To determine simultaneously the relative molecular mass (Mr) of a native oligomeric protein, and the number and Mr of its subunits, a method using two-dimensional electrophoresis was developed. To determine the Mr of a native oligomeric protein, pore gradient gel electrophoresis was performed for the first dimension. Native proteins were dissociated into their subunits by sodium dodecyl sulphate (SDS) in a gel slice, then applied to SDS polyacrylamide gel electrophoresis for the second dimension to determine the Mr of subunits. The advantage, accuracy, limitations and application of the method are discussed.  相似文献

The title carbenes where aryl groups are phenyl, 2,4, 6-trimethylphenyl, and octahydro-1,4:5,8-di(ethano)anthryl were generated for the first time by irradiation of the corresponding diazo precursors and fully characterized by ESR in a rigid matrix at low temperature. It has been demonstrated that anthryl groups can act as excellent reservoirs for the unpaired electrons as well as relatively effective kinetic protectors for carbene.  相似文献

We designed a new enteric coated preparation which is pH independent and functions by pancreatic lipase activity in the duodenum. Triolein (TO) and trilaurin (TL) were selected as lipase sensitive components and ethylcellulose (EC) was used as the support film for TO and TL. Tablets (330 mg, d = 10 mm) containing a model drug, sulfamethizole (SMZ), were coated with 1% each of TO, TL and EC solution by the fluidized bed coating technique. Disintegration tests were carried out in the media including JPXI 1st fluid (pH 1.2, JP-1), 2nd fluid (pH 6.8, JP-2) and JP-2 with gall powder and pancreatic lipase (JP-2-GL). The lag time of disintegration of the tablet (TOTL-Tab) coated 5-7 mg/tab with TO, TL and EC was about 10 min and all of the tablets disintegrated completely within 30 min in JP-2-GL. However, in the other media, which did not contain lipase, TOTL-Tab did not disintegrate for at least 2 h. It was confirmed that TO and TL in the coating film were digested by lipase. In addition, the tensil strength of the film decreased quickly after incubation in JP-2-GL. These results suggest that the application of TO, TL and EC to tablet coating is useful for an enteric release preparation sensitive to pancreatic lipase, even if patients have low gastric acidity or are taking antacids.  相似文献

A study of the polymerization of vinyl monomers with binary systems of tertiary amines and various organic halides containing chemical bonds such as C? Cl, N? Cl, O? Cl, S? Cl, and Si? Cl has been made at 60°C. Some of the binary systems were found to be effective as radical initiator in the polymerization of methyl methacrylate. The relative initiating activities of the halides in the presence of dimethylaniline were found to be in the following order: tert-C4H9OCl > n-C4H9NCl2 > (n-C4H9)2NCl ? CH3SiCl3 ? C6H5SiCl3 > C6H5SO2Cl > C6H5Cl > C6H5PCl2. Styrene and vinyl acetate polymerized only with the initiator system of dimethylaniline and benzyl chloride. Tri-n-butylamine was less active than dimethylaniline. Pyridine and 4-vinylpyridine, in combination with some organic halides, also initiated the polymerization of methyl methacrylate. The N-vinylcarbazole–benzenesulfonyl chloride system, in the presence of methyl methacrylate, gave only the homopolymer of N-vinylcarbazole.  相似文献

In this study, molecular dynamics simulations were performed to elucidate the effects of stacking fault energy (SFE) on the physical interactions between an edge dislocation and a spherical void in the crystal structure of face-centred cubic metals at various temperatures and for different void sizes. Four different types of interaction morphologies were observed, in which (1) two partial dislocations detached from the void separately, and the maximum stress corresponded to the detachment of the trailing partial; (2) two partial dislocations detached from the void separately, and the maximum stress corresponded to the detachment of the leading partial; (3) the partial dislocations detached from the void almost simultaneously without jog formation; and (4) the partial dislocations detached from the void almost simultaneously with jog formation. With an increase in void size or SFE, the interaction morphology changed in the above-mentioned order. It was observed that the magnitude of the critical resolved shear stress (CRSS) and its dependence on the SFE were determined by these interaction morphologies. The value of the CRSS in the case of interaction morphology (1) is almost equal to an analytical one based on the linear elasticity by employing the Burgers vector of a single partial dislocation. The maximum value of the CRSS is also obtained by the analytical model with the Burgers vector of the two partial dislocations.  相似文献

Potential confinement was demonstrated experimentally under various electron cyclotron resonance heating (ECRH) scenarios. The particle confinement time and the plasma confining potential increased with ECRH power. The plug potential formation by means of only ce ECRH was studied by Monte-Carlo simulation. Potential confinement experiments have advanced in higher density region up to 4×1012 cm-3. The higher density plasma was obtained by high frequency ICRF heating and recently installed neutral beam injector in the central cell. Studies of scaling relation of the potential confinement with respect to the plasma density have started with the high density plasma.  相似文献
