A 4-amido-pyrrolidone library that was intentionally synthesized as pairs of diastereomers was produced by solution-phase parallel syntheses and purified by an automated high-throughput purification system. A total of 2592 4-amido-pyrrolidinones were ultimately isolated as single diastereomers from a matrix of 1920 syntheses. After the four-step synthesis and HPLC purification, the average yield of a single diastereomer was 36.6%. The average chemical purity was >90%, and the average diastereomeric purity was >87%. The choice of chiral amines used to make amides with heterocyclic acid chlorides had a dramatic effect on success. Analysis of the relationship between amines used for synthesis and the diastereomeric separation showed that amides made from chiral 1,2-amino alcohols gave superior separation to amides from chiral morpholines. The presence of a hydrogen bond donor on the amide side chain seems to be required for a better diastereomeric separation.  相似文献

Triblock copolymer nanotubes bearing end-exposed poly(acrylic acid) or PAA core chains were prepared. The exposed PAA chains were reacted by amidization with a large excess of polystyrene spacer chains possessing amino end groups or amino-containing end blocks to graft the spacer chains. The amino groups at the other end of the spacer chains were then reacted with nanospheres bearing surface carboxyl groups to connect the nanotubes to nanospheres. The products from such a coupling reaction ranged from multiarm adduct to surfactant- and dumbbell-like objects. Product control using different strategies was explored. The products may have interesting properties and applications.  相似文献

Glucuronidation is a common mechanism in drug metabolism. In-source dissociation of glucuronides in electrospray generates fragment ions identical to those of the precursor ions of the original drugs. The effect of experimental parameters on the process was investigated in the present study using both commercially available compounds and glucuronides generated from microsomal glucuronidation incubations. It was found that cone voltage was the most critical parameter contributing to in-source fragmentation of both O- and N-glucuronides, whereas both the desolvation temperature and the source temperature had little effect. Additionally, the extent of in-source dissociation varied for different glucuronides and could be minimized by lowering cone voltage. As demonstrated in real examples, minimizing in-source dissociation can lead to higher sensitivity in detecting glucuronides in biological samples. In addition, product ions resulting from in-source dissociation of glucuronides potentially interfere with accurate determinations of corresponding drug levels if chromatographic separation is not adequate. For cases in which chromatographic separation of glucuronides from the original drugs is not readily achieved or high-throughput analyses are desired, interference caused by in-source dissociation can usually be eliminated simply by using lower cone voltage. This approach has been proven to be effective in the analysis of more than 100 glucuronides generated from in vitro microsomal incubations.  相似文献

Using the achiral diazine ligands bearing two bidentate pyridylimino groups as sources of conformational chirality, five azido-bridged coordination polymers are prepared and characterized crystallographically and magnetically. The chirality of the molecular units is induced by the coordination of the diazine ligands in a twisted chiral conformation. The use of L(1) (1,4-bis(2-pyridyl)-1-amino-2,3-diaza-1,3-butadiene) and L(2) (1,4-bis(2-pyridyl)-1,4-diamino-2,3-diaza-1,3-butadiene) induces spontaneous resolution, yielding conglomerates of chiral compounds [Mn(3)(L(1))(2)(N(3))(6)](n) (1) and [Mn(2)(L(2))(2)(N(3))(3)](n)(ClO(4))(n).nH(2)O (2), respectively, where triangular (1) or double helical (2) chiral units are connected into homochiral one-dimensional (1D) chains via single end-to-end (EE) azido bridges. The chains are stacked via hydrogen bonds in a homochiral fashion to yield chiral crystals. When L(3) (2,5-bis(2-pyridyl)-3,4-diaza-2,4-hexadiene) is employed, a partial spontaneous resolution occurs, where binuclear chiral units are interlinked into fish-scale-like homochiral two-dimensional (2D) layers via single EE azido bridges. The layers are stacked in a heterochiral or homochiral fashion to yield simultaneously a racemic compound, [Mn(2)(L(3))(N(3))(4)](n) (3a), and a conglomerate, [Mn(2)(L(3))(N(3))(4)](n).nMeOH (3b). On the other hand, the ligand without amino and methyl substituents (L(4), 1,4-bis(2-pyridyl)-2,3-diaza-1,3-butadiene) does not induce spontaneous resolution. The resulting compound, [Mn(2)(L(4))(N(3))(4)](n) (4), consists of centrosymmetric 2D layers with alternating single diazine, single EE azido, and double end-on (EO) azido bridges, where the chirality is destroyed by the centrosymmetric double EO bridges. These compounds exhibit very different magnetic behaviors. In particular, 1 behaves as a metamagnet built of homometallic ferrimagnetic chains with a unique "fused-triangles" topology, 2 behaves as a 1D antiferromagnet with alternating antiferromagnetic interactions, 3a and 3b behave as spin-canted weak ferromagnets with different critical temperatures, and 4 also behaves as a spin-canted weak ferromagnet but exhibits two-step magnetic transitions.  相似文献

The mixed micelles of cationic gemini surfactants C12C(S)C12Br2 (S=3, 6, and 12) with the nonionic surfactant Triton X-100 (TX100) have been studied by steady-state fluorescence, time-resolved fluorescence quenching, electrophoretic light scattering, and electron spin resonance. Both the surfactant composition and the spacer length are found to influence the properties of mixed micelles markedly. The total aggregation number of alkyl chains per micelle (N(T)) goes through a minimum at X(TX100)=0.8. Meanwhile, the micropolarity of the mixed micelles decreases with increasing X(TX100), while the microviscosity increases. The presence of minimum in N(T) is explained in terms of the competition of the reduction of electrostatic repulsion between headgroups of cationic gemini surfactant with the enhancement of steric repulsion between hydrophilic headgroups of TX100 caused by the addition of TX100. The variations of micropolarity and microviscosity indicate that the incorporation of TX100 to the gemini surfactants leads to a more compact and hydrophobic micellar structure. Moreover, for the C12C3C12Br2/TX100 mixed micelle containing C12C3C12Br2 with a shorter spacer, the more pronounced decrease of N(T) at X(TX100) lower than 0.8 may be attributed to the larger steric repulsion between headgroups of TX100. Meanwhile, the increase of microviscosity and the decrease of micropolarity are more marked for the C12C12C12Br2/TX100 mixed micelle, owing to the looped conformation of the longer spacer of C12C12C12Br2.  相似文献

An improved technique, pressurized capillary electrochromatography (pCEC) coupling with end-column amperometric detection (AD), was developed and used for the separation and determination of estrogens. The effects of pH value, composition of mobile phase, concentration of the surfactant sodium dodecyl sulfate (SDS) and applied voltage on separation were investigated. The electrochemical oxidation of diethylstilbestrol (DES), dienestrol (DE), and hexestrol (HEX) could be reliably monitored with a carbon electrode at 0.9 V (vs. Ag/AgCl). The pCEC analyses were performed on a capillary separation column packed with 3 microm C18 particles with an acetonitrile/water (31%: 69%) mobile phase containing Tris buffer (5 mmol/L, pH 4.5) and 4 mmol/L SDS. High voltage up to 12 kV reduced the retention time dramatically and still provided a baseline resolution. In addition, supplementary pressure prevented bubble formation and provided reliability and reproducibility of the pCEC performance. The detection limits for the three estrogens ranged from 1.2 to 2.2x10(-7) mol/L, about 10 20-fold lower than those obtained with pCEC-UV detection. To evaluate the feasibility and reliability of this system, the proposed pCEC-AD method was further demonstrated with fish muscle samples spiked with estrogens.  相似文献

Four oxamato-bridged heterotrinuclear Ni(II)Cu(II)Ni(II) complexes of formula ([Ni(bispictn)](2)Cu(pba))(ClO(4))(2).2.5H(2)O (1), ([Ni(bispictn)](2)Cu(pbaOH))(ClO(4))(2).H(2)O (2), ([Ni(cth)](2)Cu(pba))(ClO(4))(2) (3), and ([Ni(cth)](2)Cu(opba))(ClO(4))(2).H(2)O (4) and a binuclear Ni(II)Cu(II) complex of formula [Cu(opba)Ni(cth)].CH(3)OH (5) have been synthesized and characterized by means of elemental analysis, IR, ESR, and electronic spectra, where pba = 1,3-propylenebis(oxamato), pbaOH = 2-hydroxyl-1,3-propylenebis(oxamato), opba = o-phenylenebis(oxamato), bispictn = N,N'-bis(2-pyridylmethyl)-1,3-propanediamine, and cth = rac-5,7,7,12,14,14-hexamethyl-1,4,8,11-tetraazacyclotetradecane. The crystal structures of 1, 3, and 5 have been determined. The structures of complexes 1 and 3 consist of trinuclear cations and perchlorate anions, and that of 5 consists of neutral binuclear molecules which are connected by hydrogen bonds and pi-pi interactions to produce a unique supramolecular "double" sheet. In the three complexes, the copper atom in a square-planar or axially elongated octahedral environment and the nickel atom in a distorted octahedral environment are bridged by the oxamato groups, with Cu.Ni separations between 5.29 and 5.33 A. The magnetic properties of all five complexes have been investigated. The chi(M)T versus T plots for 1-4 exhibit the minimum characteristic of antiferromagnetically coupled NiCuNi species with an irregular spin state structure and a spin-quartet ground state. The chi(M)T versus T plot for 5 is typical of an antiferromagnetically coupled NiCu pair with a spin-doublet ground state. The Ni(II)-Cu(II) isotropic interaction parameters for the five complexes were evaluated and are between 102 and 108 cm(-)(1) (H = -JS(Cu).S(Ni)).  相似文献

A novel method for regioselective oxidation of phenols and anisoles has been developed in which dioxiranes, generated in situ from ketones and Oxone, oxidize phenol derivatives in an intramolecular fashion. A series of ketones with electron-withdrawing groups, such as CF(3), COOMe, and CH(2)Cl, were attached to phenols, anisoles, or aryl rings via a C(2) or C(3) methylene linker. In a homogeneous solvent system of CH(3)CN and H(2)O, oxidation of phenol derivatives 1-10 afforded spiro 2-hydroxydienones in 24-55% yields regardless of the presence of other substituents (ortho Me, meta Me or Br) on the aryl ring and the length of the linker. Experimental evidences were provided to support the mechanism that involves a regioselective pi bond epoxidation of aryl rings followed by epoxide rearrangement and hemiketal formation.  相似文献

The positively charged single walled carbon nanotubes (SWNTs~ ) were prepared by conjugating with-CONH-C_6H_(12)-NH_3~ . The double strand DNA(dsDNA) chains were loaded onto SWNTs~ via the electrostatic interactions.SWNTs~ shows improved loading efficiency (353.5μg/mg) toward dsDNA compared with that of charged free single walled carbon nanotubes (SWNTs) (82.9μg/mg).  相似文献

Reaction of nido-1,2-(Cp*RuH)2B3H7, 1, and methyl acetylene monocarboxylate under kinetic control generates nido-1,2-(Cp*Ru)2(mu-C[[CO2Me]Me])B3H7 (a pair of geometric isomers, 3 and 5) and nido-1,2-(Cp*Ru)2(1,3-mu-C[[CH2CO2Me]H])B3H7, 4, which display the first examples of exo-cluster mu-alkylidene Ru-B bridges generated by hydrometalation of an alkyne on the cluster framework. Both 3 and 5, but not 4, rearrange into arachno-2,8-mu(C)-5-eta1(O)-Me[CO2Me]C-1,2-(Cp*Ru)2B3H7, 2, in which an unprecedented intramolecular coordination of the carbonyl oxygen atom of the alkyne substituent to a boron framework site opens the ruthenaborane skeleton. Compound 2, in turn, is an intermediate in the formation of the ruthenacarborane nido-1,2-(Cp*Ru)2-3-OH-4-OMe-5-Me-4,5-C2B2H5, 12, in which the carbonyl-oxygen double bond has been cleaved as its oxygen atom inserts into a B-H bond and the carbonyl carbon inserts into the metallaborane framework. In a parallel reaction pathway, nido-1,2-(Cp*Ru)2-5-CO2Me-4,5-C2B2H7, 6, nido-1,2-(Cp*Ru)2-4-B(OH)2-5-CO2Me-4,5-C2B2H6, 16, and nido-1,2-(Cp*Ru)2(mu-H)(mu-BH2)-3-(CH2)2CO2Me-CO2Me-4,5-C2B2H4 (a pair of geometric isomers, 7 and 14, which contain an unusual Ru-B borane bridge) are formed. On heating, 7 rearranges to yield nido-1,2-(Cp*Ru)2-3-(CH2)2CO2Me-4-BH2-5-CO2Me-4,5-C2B2H5, 13, whereas 14 converts to nido-1,2-(Cp*Ru)2-3-(CH2)2CO2Me-4-CO2Me-4,5-C2B2H6, 8. Under thermodynamic control, nido-1,2-(Cp*Ru)2-4,5-B[(CH2)2CO2Me]CO(MeO)[C(CH2)CO2Me]-4,5-C2B2H6, 11, is the major product accompanied by lesser amounts of 6 and 1,2-(Cp*Ru)2-4-OMe-5-Me-4,5-C2B2H6, 10. Compound 11 features a five-membered heterocycle containing a boron atom. The structure of 7, which is an intermediate in the formation of 11, provides the basis for an explanation of this complex condensation of three alkynes. A previously unrecognized role for an exo-cluster bridging borene generated from the metallaborane skeleton by addition of the alkyne is also a feature of this chemistry. Reinsertion or loss of this boron fragment accounts for much of the chemistry observed. NMR experiments reveal labile intermediates, and one has been sufficiently characterized to provide mechanistic insight on the early stages of the alkyne-metallaborane addition reaction. All isolated compounds have been spectroscopically characterized, and most have been structurally characterized in the solid state.  相似文献
