In order to develop an analytical method for the discrimination of dextromethorphan (an antitussive medicine) from its enantiomer,
levomethorphan (a narcotic) in biological samples, chiral analyses of these drugs and their O-demethyl and/or N-demethyl metabolites in rat plasma, urine, and hair were carried out using LC-MS/MS. After the i.p. administration of dextromethorphan
or levomethorphan to pigmented hairy male DA rats (5 mg/kg/day, 10 days), the parent compounds and their three metabolites
in plasma, urine and hair were determined using LC-MS/MS. Complete chiral separation was achieved in 12 min on a Chiral CD-Ph
column in 0.1% formic acid–acetonitrile by a linear gradient program. Most of the metabolites were detected as being the corresponding
O-demethyl and N, O-didemethyl metabolites in the rat plasma and urine after the hydrolysis of O-glucuronides, although obvious differences in the amounts of these metabolites were found between the dextro and levo forms.
No racemation was observed through O- and/or N-demethylation. In the rat hair samples collected 4 weeks after the first administration, those differences were more clearly
detected and the concentrations of the parent compounds, their O-demethyl, N-demethyl, and N, O-didemethyl metabolites were 63.4, 2.7, 25.1, and 0.7 ng/mg for the dextro forms and 24.5, 24.6, 2.6, and 0.5 ng/mg for the
levo forms, respectively. In order to fully investigate the differences of their metabolic properties between dextromethorphan
and levomethorphan, DA rat and human liver microsomes were studied. The results suggested that there might be an enantioselective
metabolism of levomethorphan, especially with regard to the O-demethylation, not only in DA rat but human liver microsomes as well. The proposed chiral analyses might be applied to human
samples and could be useful for discriminating dextromethorphan use from levomethorphan use in the field of forensic toxicology,

Microchip analysis is a promising method for therapeutic drug monitoring. This led us to evaluate a microchip-based fluorescence
polarization immunoassay (FPIA) system for point-of-care testing on patients being treated with theophylline. The sera were
collected from 20 patients being treated with theophylline. Fluorescence polarization was measured on the microchip and theophylline
concentrations in serum were obtained. Regression analysis of the correlations was done between the results given by the microchip-based
FPIA and the conventional cloned enzyme donor immunoassay (CEDIA), and between the results given by the microchip-based FPIA
and the conventional particle-enhanced turbidimetric inhibition immunoassay (PETINIA). We successfully carried out a quantitative
analysis of theophylline in serum at values near its therapeutic range in 65 s. The results obtained by the microchip-based
FPIA correlated well with CEDIA and PETINIA results; the correlation coefficients (R
2) were 0.986 and 0.989, respectively. The FPIA system is a simple and rapid method for point-of-care testing of drugs in serum,
and its accuracy is the same as the conventional CEDIA and PETINIA. It is essential to use real samples from patients and

We generalize the division polynomials of elliptic curves to hyperelliptic Jacobians over the complex numbers. We construct them by using the hyperelliptic sigma function. Using the division polynomial, we describe a condition that a point on the Jacobian is a torsion point. We prove several properties of the division polynomials such as determinantal expressions and recurrence formulas. We have employed the proposed Silica-Silver Core-Shell (SSCS) SERS-active substrates to detect four model proteins: lysozyme (a protein without chromophore), cytochrome c (a protein with chromophore of heme), fluorescein isothiocyanate (FITC)-anti human IgG (labeled with FITC) and atto610-biotin/avidin (recognition with labeled small molecules). SERS spectra of these proteins and Raman labels on the SSCS substrates show both high sensitivity and reproducibility, which are due to electromagnetic SERS enhancement with additional localization field within closely packed Ag nanoparticles decorated on the SiO(2) nanoparticles and the aggregation of SiO(2)@Ag particles. We have found that the SERS intensities of atto610-biotin/avidin adsorbed on the SSCS substrates are about 20 times stronger than those from Ag plating on Au-decorated substrate. Moreover, the broad surface plasmon resonance (SPR) of the proposed substrates will extend SERS applications to more biological molecules with different laser excitations. 相似文献

We have investigated the pressure-induced Raman spectral changes of 1-ethyl-3-methylimidazolium tetrafluoroborate ([emim][BF4]). We found that [emim][BF4] did not crystallize up to 1.2 GPa. The Raman CH stretching spectra arising from the CH3 groups of the ethyl-chain and the CH3 group adjacent to the imidazolium-ring in [emim]+ cation largely changed against pressure. Moreover, the Raman intensity of the CH2 (N) bending band arising from the alkyl-chain drastically changes with increasing pressure, but that of the imidazolium-ring in-plane bending band arising from the imidazolium-ring is independent of pressure. Our results show that the environment around the alkyl-chain of [emim][BF4] is largely perturbed rather than that around the imidazolium-ring upon compression. 相似文献

We first show that there are in fact triangular arbitrage opportunities in the spot foreign exchange markets, analyzing the time dependence of the yen–dollar rate, the dollar–euro rate and the yen–euro rate. Next, we propose a model of foreign exchange rates with an interaction. The model includes effects of triangular arbitrage transactions as an interaction among three rates.
