Radioligand therapy (RLT) is an emergent drug class for cancer treatment. The dose administered to cancer patients is constrained by the radiation exposure to normal tissues to maintain an appropriate therapeutic index. When a radiopharmaceutical or its radiometabolite is retained in the kidneys, radiation dose deposition in the kidneys can become a dose-limiting factor. A good exemplar is [177Lu]Lu-DOTATATE, where patients receive a co-infusion of basic amino acids for nephroprotection. Besides peptides, there are other classes of targeting vectors like antibody fragments, antibody mimetics, peptidomimetics, and small molecules that clear through the renal pathway. In this review, we will review established and emerging strategies that can be used to mitigate radiation-induced nephrotoxicity, with a focus on the development and incorporation of cleavable linkers for radiopharmaceutical designs. Finally, we offer our perspectives on cleavable linkers for RLT, highlighting future areas of research that will help advance the technology.  相似文献

Combination of wet-grinding and laser fragmentation is a promising approach to advance both methods: Laser fragmentation will be more efficient when combined with mechanical treatment and wet-grinding may take advance of the abrasion-free laser process to achieve fabrication of smaller particles. By mechanical pre-treatment of zinc oxide microparticles in a stirred-media mill, the starting material is activated by generation of crystallographic defects, which strongly enhance the efficiency of subsequent laser fragmentation. Picosecond-laser irradiation of mechanically treated and untreated microparticles suspended in water yielded in colloidal zinc oxide nanoparticles. Furthermore, nanoparticle productivity and properties can be controlled by variation of anionic surfactant concentration.  相似文献

This pilot study elaborates the development of novel epoxy/electrospun polylactic acid (PLA) nanofiber composites at the fiber contents of 3, 5, and 10 wt % to evaluate their mechanical and thermal properties using flexural tests and differential scanning calorimetry (DSC). The flexural moduli of composites increase remarkably by 50.8 and 24.0% for 5 and 10 wt % fiber contents, respectively, relative to that of neat epoxy. Furthermore, a similar trend is also shown for corresponding flexural strengths being enhanced by 31.6 and 4.8%. Fractured surface morphology with scanning electron microscopy (SEM) confirms a full permeation of cured epoxy matrix into nanofiber structures and existence of nondestructive fibrous networks inside large void cavities. The glass transition temperature (Tg) of composites increases up to 54–60 °C due to embedded electrospun nanofibers compared to 50 °C for that of epoxy, indicating that fibrous networks may further restrict the intermolecular mobility of matrix in thermal effects. © 2014 Wiley Periodicals, Inc. The synthesis and X-ray structure of a new manganese(V) mesitylimido complex with a tetraamido macrocyclic ligand (TAML), [MnV(TAML)(N-Mes)] ( 1 ), are reported. ‘Slotting fee’ (hereafter ‘SF’) is an upfront fee a ‘supplier’ is required to pay a retailer in order to have his product sold on the retailer's shelves. It is becoming increasingly common, but also widely reviled. This paper considers a newsvendor product whose expected demand is dependent on retail price and sales effort. The question we pose is: given that the Stackelberg-dominant retailer has to choose a pricing contract with which she transacts with the supplier, how would the supply-chain stakeholders fare when the retailer implements SF instead of another practical pricing contract? We show that, contradicting its negative public image, SF empowers the dominant retailer to specify contract terms that will benefit all the stakeholder-groups. That is, the supplier's and the retailer's profits are higher, the production workers are asked to produce more, and the consumers pay a lower retail price. We also propose a new ‘composite’ contract format that incorporates both the SF and ‘buyback’ features. This composite format empowers the retailer to provide even greater benefits to the supply-chain's stakeholders.

The kinetics and mechanism of the reaction of SIV (SO32?+HSO3?) with a ruthenium(VI) nitrido complex, [(L)RuVI(N)(OH2)]+ (RuVIN, L=N,N′‐bis(salicylidene)‐o‐cyclohexyldiamine dianion), in aqueous acidic solutions are reported. The kinetic results are consistent with parallel pathways involving oxidation of HSO3? and SO32? by RuVIN. A deuterium isotope effect of 4.7 is observed in the HSO3? pathway. Based on experimental results and DFT calculations the proposed mechanism involves concerted N?S bond formation (partial N‐atom transfer) between RuVIN and HSO3? and H+ transfer from HSO3? to a H2O molecule.  相似文献
