New chemically modified oligonucleotides at the site of the backbone are needed to improve the properties of oligonucleotides. A practical synthesis for a triazole‐linked nucleoside dimer based on a PNA‐like structure has been developed. This involves synthesizing two uracil‐based monomers that contain either an azide or an alkyne functionality, followed by copper‐catalyzed 1,3‐dipolar cycloaddition. This dimer was incorporated within an oligonucleotide via phosphoramidite chemistry and UV‐monitored thermal denaturation data illustrates slight destabilization relative to its target complementary sequence. A fast and direct competitive biomimetic enzyme‐linked immunosorbent assay (BELISA) method was developed for the determination of methimazole (MMZ) in urine sample based on a molecularly imprinted film as an artificial antibody. This is the first example to monitor methimazole with a direct competitive biomimetic enzyme‐linked immunosorbent assay (BELISA) method. The imprinted film was directly synthesized on the well surface of MaxiSorp polystyrene 96‐well plate and characterized. The results showed that it exhibited an antibody‐like binding ability, rapid adsorption speed, high stability, which was particularly advantageous and suitable for BELISA development. The BELISA method established in this paper had a higher selectivity for MMZ than for the structurally related compounds and the IC50 (calculated as the concentration giving 50% inhibition of color development) and the detection limit values under optimized experimental conditions were 70 ± 4 μg L‐1 and 0.9 ± 0.04 μg L‐1, respectively. The method was applied to the determination of MMZ in spiked urine sample with excellent recoveries ranging from 90% to 95%, and the imprinted film was able to be reused for 20 times without loss of sensitivity. Au/TiO2 nanorod composites with different ratios of [TiO2]:[Au] have been prepared by chemically reducing AuCl4‐ on the positively charged TiO2 nanorods surface and used to modify boron‐doped diamond (BDD) electrodes. The electrochemical behaviors of catechol on the bare and different Au/TiO2 nanorod composites‐modified BDD electrodes are studied. The cyclic voltammetric results indicate that these different Au/TiO2 nanorod composites‐modified BDD electrodes can enhance the electrocatalytic activity toward catechol detection, as compared with the bare BDD electrode. Among these different conditions, the Au/TiO2‐BDD3 electrode (the ratio of [TiO2]:[Au] is 27:1) is the most choice for catechol detection. The electrochemical response dependences of the Au/TiO2‐BDD3 electrode on pH of solution and the applied potential are studied. The detection limit of catechol is found to be about 1.4 × 10‐6 M in a linear range from 5 × 10‐6 M to 200 × 10‐6 M on the Au/TiO2‐BDD3 electrode. 相似文献

The bifunctional activities of α-amylase and pullulanase are found in the cloned recombinant amylopullulanase. It was encoded
in a 2.9-kb DNA fragment that was amplified using polymerase chain reaction from the chromosomal DNA of Thermoanaerobacter ethanolicus 39E. An estimated 109-kDa recombinant protein was obtained from the cloned gene under the prokaryotic expression system.
The optimum pH of the recombinant amylopullulanase was 6.0. The most stable pH for the α-amylase and pullulanase activity
was 5.5 and 5.0, respectively. The optimum temperature for the α-amylase activity was 90°C, while its most stable temperature
was 80°C. Regarding pullulanase activity, the optimum temperature and its most stable temperature were found to be 80 and
75°C, respectively. Pullulan was found to be the best substrate for the enzyme. The enzyme was activated and stabilized by
the presence of Ca2+, whereas EDTA, N-bromosuccinimide, and α-cyclodextrin inhibited its bifunctional activities. A malto-2–4-oligosac-charide was the major product
obtained from the enzymatic reaction on soluble starch, amylose, amylopectin, and glycogen. A single maltotriose product was
found in the pullulan hydrolysis reaction using this recombinant amylopullulanase. Kinetic analysis of the enzyme indicated
that the K
m
values of α-amylase and pullulanase were 1.38 and 3.79 mg/mL, respectively, while the V

A pure algebraic treatment of the eigenvalue equation corresponding to the asymmetric top is presented. The algebraic method employs the Holstein–Primakoff bosonic realization of the angular momentum algebra. Explicit determination of the linear boson transformation coefficients of the eigenstates is carried out by means of the coherent states formalism. Polyaniline–emeraldine base (EB) fiber with excellent mechanical and electrical properties have been spun from highly concentrated (20% w/w), EB/N‐methyl‐2‐pyrrolidinone (NMP)/2‐methylaziridine (2 MA) solution. These solutions had gelation times, which varied from hours to days depending on the molar ratio of 2 MA to EB tetramer repeating unit in the N‐methyl‐2‐pyrrolidinone (NMP) solvent. To better compare the mechanical and electrical properties, dense films were also prepared by thermal evaporation of less concentrated solution (1% w/w). Both fibers and films were amenable to thermal stretching with maximum draw ratios of 4 : 1 and these stretched samples exhibited the greatest tensile strength overall. Wide‐angle X‐ray diffraction (WAXD) of as‐spun and 4‐times stretched fiber showed a completely amorphous structure. Fiber subjected to heat treatment at 250 °C under N2 flux for 2 h displayed further improvements in mechanical properties because of crosslinking between the polymer chains. Fibers and films were later doped by immersion in a variety of aqueous acid solutions. Room temperature DC conductivities for the doped samples ranged from 6 × 10−4 to 45 S/cm depending on the specific choice of acid. Scanning electron microscopy of fiber samples shows the presence of macrovoid formation during fiber spinning. Continued refinement of the processing parameters and fiber post‐treatment, to enhance chain alignment and increase fiber density, will likely lead to additional improvements in the fiber mechanical and electrical properties. Characterization of emeraldine base (EB) powder, solution, films, and fibers by UV‐Vis, DSC, TGA, and WAXD were also performed. © 2000 John Wiley & Sons, Inc. The effect of liquid–liquid phase separation (LLPS) on the crystallization behavior of poly(ethylene‐ran‐vinyl acetate) with a vinyl acetate content of 9.5 wt % (EVA‐H) in the critical composition of a 35/65 (wt/wt) EVA‐H/paraffin wax blend was investigated by small‐angle light and X‐ray scattering methods and rheometry. This blend exhibited an upper critical solution temperature (UCST) of 98°C, and an LLPS was observed between the UCST and the melting point of 88°C for the EVA‐H in the blend. As the duration time in the LLPS region increased before crystallization at 65°C, both the spherulite size and the crystallization rate of the EVA‐H increased, but the degree of the lamellar ordering in the spherulite and the degree of crystallinity of the EVA‐H in the blend decreased. © 2000 John Wiley & Sons, Inc. A quantitative study was undertaken of the anisotropy of low‐strain mechanical behavior for specially oriented polyethylene with controlled crystalline and lamellar orientation. The samples were prepared by the die drawing of injection‐molded rods of polyethylene and annealing. This produced a parallel lamellar structure for which a simple, three‐dimensional composite laminate model could be used to calculate the expected anisotropy. Experimental data, including X‐ray strain measurements of the lateral crystalline elastic constants, showed good quantitative agreement with the model prediction. The X‐ray strain measurements confirmed that the amorphous regions exert large constraints on the crystalline phase in the lateral directions, where an order of magnitude difference was found between the measured apparent lateral crystalline compliances in the lamellar‐stack sample and the expected values for a perfect crystal. © 2000 John Wiley & Sons, Inc. The isothermal crystallization kinetics of poly(trimethylene terephthalate) (PTT) have been investigated using differential scanning calorimetry (DSC) and polarized light microscopy (PLM). Enthalpy data of exotherm from isothermal crystallization were analyzed using the Avrami theory. The average value of the Avrami exponent, n, is about 2.8. From the melt, PTT crystallizes according to a spherulite morphology. The spherulite growth rate and the overall crystallization rate depend on crystallization temperature. The increase in the spherulitic radius was examined by polarized light microscopy. Using values of transport parameters common to many polymers (U* = 1500 cal/mol, T∞= Tg − 30 °C) together with experimentally determined values of T (248 °C) and Tg (44 °C), the nucleation parameter, kg, for PTT was determined. On the basis of secondary nucleation analyses, a transition between regimes III and II was found in the vicinity of 194 °C (ΔT ≅ 54 K). The ratio of kg of these two regimes is 2.1, which is very close to 2.0 as predicted by the Lauritzen–Hoffman theory. The lateral surface‐free energy, σ = 10.89 erg/cm2 and the fold surface‐free energy, σe = 56.64 erg/cm2 were determined. The latter leads to a work of chain‐folding, q = 4.80 kcal/mol folds, which is comparable to PET and PBT previously reported. © 2000 John Wiley & Sons, Inc. The positron annihilation lifetime (PAL) of a series of copolyimides and copolyamides with microphase‐separated structures was measured to investigate the effects of different hard‐segment polymers on the PAL properties of soft‐segment domains of poly(dimethyl‐siloxane) (PDMS) and poly(ethylene oxide) (PEO). The lifetime (τ3) and intensity (I3) of the long‐lived component are given as a function of the PDMS or PEO content for a series of copolymers, of which the density roughly obeys the additive rule except for the PDMS‐segmented copolyamides. The PDMS‐segmented copolyimides and copolyamides show much smaller I3 values than those estimated from the additive rule. The lifetime distribution of the long‐lived component for the PDMS‐segmented copolyamides is composed of two components. The longer‐lifetime component is attributed to pure PDMS domains, and the shorter‐lifetime component is attributed to the polyamide domains, intermediate phases, and PDMS domains containing small amounts of short amide blocks. Despite the high PDMS content, the latter component is rather large. Thus, the positronium formation in the PDMS domains of the copolyimides and copolyamides is effectively reduced. This can be explained by the combination of the difference in the electron affinity of the PDMS and polyimide or polyamide segments and the incomplete phase separation. The PEO‐segmented copolyimides show much smaller I3 values than those predicted from the additive rule.
