2-Substituted homophthalimides 2a-c were reduced regioselectively with sodium borobydride to carbinol-lactam intermediates 3a-c , which were dehydrated, followed by hydrogenation, to give 1-oxo-tetrabydroisoquinolines or 3,4-dihydroisoquinolin-1(2H)ones 5a-c . Nemonoxacin (TG‐873870) is a novel C‐8‐methoxy nonfluorinated quinolone with higher activity than ciprofloxacin, levofloxacin and moxifloxacin against Gram‐positive pathogens including methicillin‐susceptible or methicillin‐resistant Staphylococcus aureus and Streptococcus pneumoniae with various resistant phenotypes. A rapid, sensitive and selective liquid chromatography–tandem mass spectrometric (LC‐MS/MS) method was developed and validated to determine the concentration of nemonoxacin in human plasma and urine. Protein precipitation and liquid–liquid extraction were employed for plasma and urine sample preparations, respectively, and extract was then injected into the system. Separation was performed on a C18 reversed‐phase column using acetonitrile–0.1% formic acid as a mobile phase. Both analyte and internal standard (gatifloxacin) were determined using electrospray ionization and the MS data acquisition via the selected reaction monitoring in positive‐ion mode. The lower limit of quantification was 5 ng/mL and the calibration curves were linear in the concentration range of 5–1000 ng/mL. The accuracy, precision, selectivity, linearity, recovery, matrix effect and stability were validated for TG‐873870 in human plasma and urine. Take five: A unique quintuply bonded dimolybdenum complex [Mo(2)(μ-Li){μ-HC(N-2,6-Et(2)C(6)H(3))(2)}(3)] (see picture) was synthesized and characterized. The Mo-Mo interaction includes an unexpected bridging Li(+) ion. In this work, the effects of electrolytes used in roughening gold substrates by electrochemical methods on surface-enhanced Raman scattering (SERS) were first investigated. First, gold substrates were roughened by triangular-wave oxidation–reduction cycles (ORC) in aqueous solutions containing different kinds of 0.1 M electrolytes. Then Rhodamine 6G (R6G) was used as Raman probe to examine this effect of electrolytes used on the SERS observed. The result indicates that the highest intensity of SERS of R6G was obtained on the roughened Au substrate prepared in 0.1 M NaCl, which was less used in the literature. Meanwhile, it was also found that the rougher surface morphology observed, which is contributive to the higher SERS obtained, is corresponding to the smaller cathodic peak area shown in the cyclic voltammograms for roughening the Au substrate. 相似文献

This paper utilizes molecular-dynamics simulations to investigate the mechanical characteristics of a suspended (10, 10) single-walled carbon nanotube (SWCNT) during atomic force microscopy (AFM) nanoindentation at different temperatures. Spontaneous topological transition of the Stone-Wales (SW) defects is clearly observed in the indentation process. The present results indicate that under AFM-bending deformation, the mechanical properties of the SWCNT, e.g., the bending strength, are dependent on the wrapping angle. In addition, it is also found that the radial dependence of the reduced formation energy of the SW defects is reasonably insensitive only for the small tubes. However, for tube diameters greater than 2.4 nm [corresponding to the (18, 18) CNT], the SW defects tend to be more radius sensitive. The results indicate that the bending strength decreases significantly with increasing temperature. This study also investigates the variation in the mechanical properties of the nanotube with the density of C60 encapsulated within the nanotube at various temperatures. It is found that, at lower temperatures, the bending strength of the C60-filled nanotube increases with C60 density. However, the reverse tendency is observed at higher temperatures. Finally, the "sharpest tip" phenomena between the probe and the tube wall and the elastic recovery of the nanotube during the retraction process are also investigated. 相似文献

Pancreatic ductal adenocarcinoma is one of the most lethal malignancies: more than half of patients are diagnosed with a metastatic disease, which is associated with a five-year survival rate of only 3%. 5-epi-Sinuleptolide, a norditerpene isolated from Sinularia sp., has been demonstrated to possess cytotoxic activity against cancer cells. However, the cytotoxicity against pancreatic cancer cells and the related mechanisms are unknown. The aim of this study was to evaluate the anti-pancreatic cancer potential of 5-epi-sinuleptolide and to elucidate the underlying mechanisms. The inhibitory effects of 5-epi-sinuleptolide treatment on the proliferation of pancreatic cancer cells were determined and the results showed that 5-epi-sinuleptolide treatment inhibited cell proliferation, induced apoptosis and G2/M cell cycle arrest, and suppressed the invasion of pancreatic cancer cells. The results of western blotting further revealed that 5-epi-sinuleptolide could inhibit JAK2/STAT3, AKT, and ERK phosphorylation, which may account for the diverse cytotoxic effects of 5-epi-sinuleptolide. Taken together, our present investigation unveils a new therapeutic and anti-metastatic potential of 5-epi-sinuleptolide for pancreatic cancer treatment. 相似文献

Intramolecular interactions are shown to be key for favoring a given structure in systems with a variety of conformers. In ortho-substituted benzene derivatives including a beryllium moiety, beryllium bonds provide very large stabilizations with respect to non-bound conformers and enthalpy differences above one hundred kJ·mol−1 are found in the most favorable cases, especially if the newly formed rings are five or six-membered heterocycles. These values are in general significantly larger than hydrogen bonds in 1,2-dihidroxybenzene. Conformers stabilized by a beryllium bond exhibit the typical features of this non-covalent interaction, such as the presence of a bond critical point according to the topology of the electron density, positive Laplacian values, significant geometrical distortions and strong interaction energies between the donor and acceptor quantified by using the Natural Bond Orbital approach. An isodesmic reaction scheme is used as a tool to measure the strength of the beryllium bond in these systems in terms of isodesmic energies (analogous to binding energies), interaction energies and deformation energies. This approach shows that a huge amount of energy is spent on deforming the donor–acceptor pairs to form the new rings. 相似文献

Abstract Treatment of ethyl oxalyl chloride or methyl oxalyl chloride with lithium diisopropyl(carboethoxyfluoromethyl)phosphonate[(i-PrO)2P(O)CFCO2Et]?Li+ 2 followed by in siru nucle-ophilic addition with methylmagnesium iodide or vinyl magnesium bromide affords with exclusive E-stereoselectivity formation of diethyl-2-fluoro-3-methyl fumarate (CH3)(C02Et)C[dbnd]CFCO2Et 4 or 75% of the E-isomer of a-fluoro-P-vinyl-a,P-unsaturated diester (E,Z)-(CH2[dbnd]CH)(CO2C2H5)C[dbnd]CFCO2Et 5, respectively. However, direct reaction of ethyl pyruvate with 2 gives the fluoro-olefin (CH3)(CO2Et)C[dbnd]CFCO2Et 4 with 79% E-stere-oselectivity. The E/Zratio of (CH2[dbnd]CH)(CO2Et)C[dbnd]CFCO2Et 5 depends on the HMFT or DMPU cosolvents present in the reaction mixture. 相似文献
