Despite first evidence for the cytochrome P450-mediated enantioselective biosynthesis and activity of cis-epoxyeicosatrienoic acids (EETs), as yet little is known about the stereospecifity of EET generation and physiology, because the existing chiral methods are time consuming, labor intensive, and not sensitive enough. We present a method for highly sensitive, direct, and simultaneous chiral analysis of all eight EET enantiomers consisting of (i) solid-phase extraction, (ii) reversed-phase high-performance liquid chromatographic purification followed by (iii) consecutive regio- and enantiomeric separation of the four underivatized EET regioisomers within one chromatographic run employing capillary tandem column chiral-phase liquid chromatography with (iv) reliable dual online photodiode array and gentle electrospray ionization tandem mass spectrometric identification and quantitation of the eluting optical antipodes. This one-step, simple, expeditious, and highly sensitive measurement allows profiling of all eight EET enantiomers at once, thus avoiding substance loss and enabling high sample throughput. Limits of quantification in the low picogram range were achieved by the use of capillary columns with typical high quantitative sensitivity instead of conventional columns with low chromatographic signal intensity employed by previous methods. Application to tissue homogenates demonstrated the suitability of this approach for routine and reliable “enantioprofiling” of free endogenous EETs, i.e., EETs not esterified into cellular membrane phospholipids, typically occurring at very low concentrations. The technique can readily be employed for preparative purification of enantiomers in the microgram range using large-inner-diameter columns. This review provides a broad overview of the literature related to the importance of ligands in homogeneous catalysis. In particular, it describes the types of nitrogen donor ligands that have typically been used for homogeneous catalysis. We surveyed the important transition metal homogenous catalysts explicitly from 2011 up to early 2014 and summarize their comparative catalytic activities. Generally, the main factors observed are the ligand structure, electron donor property and steric bulk which can affect the catalytic activity. Electron count and inductive effect can also influence the efficiency of homogeneous catalyst.  相似文献

We have considered the generalized version of chiral schwinger model with the Lorentz covariant masslike term for gauge field with the choice a ? r2 =?0. We carry out the quantization by the canonical Dirac method of both the gauge-invariant and non-invariant version of this model to determine the phase space structure. Therefore we have shown that the gauge invariant theory has the same physical spectrum as that of the original gauge noninvariant formulation.

In the present investigation we have analyzed the peristaltic flow when the viscous fluid with variable viscosity is bounded with convective walls. The combined effects of thermaldiffusion (Soret effects) and diffusion-thermo (Dufour effect) are considered. Convective form of the heat and mass transport phenomenon has been given special attention in addition to the effects of magnetic field. Mathematical formulation for the physical systemis derived and simplified using long wavelength and low Reynolds number approximation. Series solutions for the velocity, temperature andmass concentration fields are computed and elaborated for different physical quantities including magnetic parameter, Soret and Dufour effects, Biot numbers, etc. Streamlines analysis is presented showing the effects of bolus movement.  相似文献

The current research work describes the development of a rapid HPLC method for the concurrent detection of pregabalin and piperine in dual drug-loaded nanoformulations. The primary goal was to recognize the chromatographic conditions wherein propitious segregation of the integrants with quality peaks can be attained. An attempt to expound the target analytical profile was made to accomplish this goal, and critical method attributes (CMAs), viz. percentage acetonitrile content, injection volume and pH, which affect critical quality attributes (CQAs), were identified using systemic risk analysis. Box–Behnken design was employed to develop a relationship between CMAs and CQAs, which engenders an analytical design space. Efficient chromatographic separation for pregabalin and piperine was attained using an analytical C18 column and mobile phase comprising acetonitrile–water (pH 6.9; 70:30%, v/v) in an isocratic elution mode with a 1 ml/min flow rate. The elution was descried at an isosbestic wavelength of 221 nm using a photodiode array detector. The International Conference on Harmonization guidelines were adopted for the developed HPLC method. The validated HPLC method can be further utilized for the simultaneous quantification and detection of pregabalin and piperine in other lipid-based nanopharmaceuticals such as polymeric nanoparticles, nanocrystals, solid-lipid nanoparticles, metallic nanoparticles, etc., in in vitro and in vivo studies.  相似文献

Achieving in a straightforward way the synthesis of enantioenriched elaborated three‐dimensional molecules related to bioactive natural products remains a long‐standing quest in organic synthesis. Enantioselective organocatalysis potentially offers a unique opportunity to solve this problem, especially when combined with complementary modes of activation. Here, we report the sequential association of organocatalytic and superacid activations of simple linear achiral readily available precursors to promote the formation of unique highly elaborated chiral methylene‐bridged benzazocanes exhibiting three to five fully‐controlled stereocenters. This peculiar backbone, difficult to assemble by standard synthetic approaches, is closely related to bioactive natural and synthetic morphinans and benzomorphans. The formation of a highly reactive chiral 7‐membered ring N‐acyl iminium superelectrophilic ion, evidenced by low‐temperature in situ NMR experiments, triggers a challenging stereoselective Friedel–Crafts‐type cyclization.  相似文献

In this work the surface of LiNibegin{document}$_{0.5}$end{document}Mnbegin{document}$_{1.5}$end{document}Obegin{document}$_{4}$end{document} (LMN) particles is modified by Mnbegin{document}$_{3}$end{document}Obegin{document}$_{4}$end{document} coating through a simple wet grinding method, the electronic conductivity is significantly improved from 1.53begin{document}$times$end{document}10begin{document}$^{-7}$end{document} S/cm to 3.15begin{document}$times$end{document}10begin{document}$^{-5}$end{document} S/cm after 2.6 wt% Mnbegin{document}$_{3}$end{document}Obegin{document}$_{4}$end{document} coating. The electrochemical test results indicate that Mnbegin{document}$_{3}$end{document}Obegin{document}$_{4}$end{document} coating dramatically enhances both rate performance and cycling capability (at 55 ℃) of LNM. Among the samples, 2.6 wt% Mnbegin{document}$_{3}$end{document}Obegin{document}$_{4}$end{document}-coated LNM not only exhibits excellent rate capability (a large capacity of 108 mAh/g at 10 C rate) but also shows 78% capacity retention at 55 ℃ and 1 C rate after 100 cycles.  相似文献

Artificial counterparts of conical‐shaped transmembrane protein channels are of interest in biomedical sciences for biomolecule detection and selective ion permeation based on ionic size and/or charge differences. However, industrial‐scale applications such as seawater desalination, separation of mono‐ from divalent cations, and treatment of highly‐saline industrial waste effluents are still big challenges for such biomimetic channels. A simple monomer seeding experimental approach is used to grow ionically conductive biomimetic charged nanocone pores at the surface of an acid‐functionalized membrane. These readily scalable nanocone membranes enable ultra‐fast cation permeation (Na+=8.4× vs. Mg2+=1.4×) and high ion charge selectivity (Na+/Mg2+=6×) compared to the commercial state‐of‐the‐art permselective membrane (CSO, Selemion, Japan) owing to negligible surface resistance and positively charged conical pore walls.  相似文献
