A general procedure is proposed for the rapid development of a reversed-phase liquid chromatographic (RP-LC) separation that is "orthogonal" to a pre-existing ("primary") method for the RP-LC separation of a given sample. The procedure involves a change of the mobile-phase organic solvent (B-solvent), the replacement of the primary column by one of very different selectivity, and (only if necessary) a change in mobile phase pH or the use of a third column. Following the selection of the "orthogonal" B-solvent, column and mobile phase pH, further optimization of peak spacing and resolution can be achieved by varying separation temperature and either isocratic %B or gradient time. The relative "orthogonality" of the primary and "orthogonal" RP-LC methods is then evaluated from plots of retention for one method versus the other. The present procedure was used to develop "orthogonal" methods for nine routine RP-LC methods from six pharmaceutical analysis laboratories. The relative success of this approach can be judged from the results reported here.  相似文献

The near K-edge structure of oxygen in liquid water and ices III, II, and IX at 0.25 GPa and several low temperatures down to 4 K has been studied using inelastic x-ray scattering at 9884.7 eV with a total energy resolution of 305 and 175 meV. A marked decrease of the preedge intensity from the liquid phase and ice III to ices II and IX is attributed to ordering of the hydrogen bonds in the proton-ordered lattice of the latter phases. Density functional theory calculations including the influence of the Madelung potential of the ice IX crystal correctly account for the remaining preedge feature. Furthermore, we obtain spectroscopic evidence suggesting a possible new phase of ice at temperatures between 4 and 50 K.  相似文献

A robust new solid-state nuclear magnetic resonance (NMR) method for selecting CH2 signals in magic-angle spinning (MAS) 13C NMR spectra is presented. Heteronuclear dipolar evolution for a duration of 0.043 ms, under MREV-8 homonuclear proton decoupling, converts 13C magnetization of CH2 groups into two- and three-spin coherences. The CH2 selection in the SIJ (C H H) spin system is based on the three-spin coherence S(x)I(z)J(z), which is distinguished from 13C magnetization (S(x)) by a 1H 0 degrees/90 degrees pulse consisting of two 45 degrees pulses. The two-spin coherences of the type S(y)I(z) are removed by a 13C 90 degrees x-pulse. The three-spin coherence is reconverted into magnetization during the remainder of the rotation period, still under MREV-8 decoupling. The required elimination of 13C chemical-shift precession is achieved by a prefocusing 180 degrees pulse bracketed by two rotation periods. The selection of the desired three-spin coherence has an efficiency of 13% theoretically and of 8% experimentally relative to the standard CP/MAS spectrum. However, long-range couplings also produce some three-spin coherences of methine (CH) carbons. Therefore, the length of the 13C pulse flipping the two-spin coherences is increased by 12% to slightly invert the CH signals arising from two-spin coherences and thus cancel the signal from long-range three-spin coherences. The signal intensity in this cleaner spectrum is 6% relative to the regular CP/TOSS spectrum. The only residual signal is from methyl groups, which are suppressed at least sixfold relative to the CH2 peaks. The experiment is demonstrated on cholesteryl acetate and applied to two humic acids.  相似文献

The magnetic behavior of a Bi57FeO3 powdered sample was studied at high pressures by the method of nuclear forward scattering (NFS) of synchrotron radiation. The NFS spectra from 57Fe nuclei were recorded at room temperature under high pressures up to 61.4 GPa, which were created in a diamond anvil cell. In the pressure interval 0 < P < 47 GPa, the magnetic hyperfine field HFe at the 57Fe nuclei increased reaching a value of ~52.5 T at 30 GPa, and then it slightly decreased to ~49.6 T at P = 47 GPa. As the pressure was increased further, the field HFe abruptly dropped to zero testifying a transition from the antiferromagnetic to a nonmagnetic state (magnetic collapse). In the pressure interval 47 < P < 61.4 GPa, the value of HFe remained zero. The field HFe recovered to the low-pressure values during decompression.  相似文献

We report the first detailed investigation of the (1)H, (13)C, (15)N, and (19)F nuclear magnetic resonance (NMR) spectroscopic shifts in paramagnetic metalloprotein and metalloporphyrin systems. The >3500 ppm range in experimentally observed hyperfine shifts can be well predicted by using density functional theory (DFT) methods. Using spin-unrestricted methods together with large, locally dense basis sets, we obtain very good correlations between experimental and theoretical results: R(2) = 0.941 (N = 37, p < 0.0001) when using the pure BPW91 functional and R(2) = 0.981 (N = 37, p < 0.0001) when using the hybrid functional, B3LYP. The correlations are even better for C(alpha) and C(beta) shifts alone: C(alpha), R(2) = 0.996 (N = 8, p < 0.0001, B3LYP); C(beta), R(2) = 0.995 (N = 8, p < 0.0001, B3LYP), but are worse for C(meso), in part because of the small range in C(meso) shifts. The results of these theoretical calculations also lead to a revision of previous heme and proximal histidine residue (13)C NMR assignments in deoxymyoglobin which are confirmed by new quantitative NMR measurements. Molecular orbital (MO) analyses of the resulting wave functions provide a graphical representation of the spin density distribution in the [Fe(TPP)(CN)(2)](-) (TPP = 5,10,15,20-tetraphenylporphyrinato) system (S = (1)/(2)), where the spin density is shown to be localized primarily in the d(xz) (or d(yz)) orbital, together with an analysis of the frontier MOs in Fe(TPP)Cl (S = (5)/(2)), Mn(TPP)Cl (S = 2), and a deoxymyoglobin model (S = 2). The ability to now begin to predict essentially all heavy atom NMR hyperfine shifts in paramagnetic metalloporphyrins and metalloproteins using quantum chemical methods should open up new areas of research aimed at structure prediction and refinement in paramagnetic systems in much the same way that DFT methods have been used successfully in the past to predict/refine elements of diamagnetic heme protein structures.  相似文献

Glycosaminoglycans are a family of polydisperse, highly sulfated complex mixtures of linear polysaccharides that are involved in many life processes. Defining the structure of glycosaminoglycans is an important factor in elucidating their structure-activity relationship. Capillary electrophoresis has emerged as a highly promising technique consuming an extremely small amount of sample and capable of rapid, high-resolution separation, characterization and quantitation of analytes. Numerous capillary electrophoresis methods for analysis of intact glycosaminoglycans and glycosaminoglycan-derived oligosaccharides have been developed. These methods allow for both qualitative and quantitative analysis with a high level of sensitivity. This review is concerned with separation methods of capillary electrophoresis, detection methods and applications to several aspects of research into glycosaminoglycans and glycosaminoglycan-derived oligosaccharides. The importance of capillary electrophoresis in biological and pharmaceutical samples in glycobiology and carbohydrate biochemistry and its possible applications in disease diagnosis and monitoring chemical synthesis are described.  相似文献

The diffusive behavior of ethane and ethylene in single-walled carbon nanotubes is investigated using classical molecular dynamics simulations and density functional theory calculations. At low molecular densities, these nonspherical molecules follow a spiral path inside nanotubes with diameters of 13-22 A, which maximizes the interaction of molecular C-C bonds with the C-C bonds in the nanotubes. Spherical molecules, such as methane, are not predicted to follow a spiral diffusion path. This result quantifies the manner in which molecular shape and chemical bonding affects molecule-nanotube interactions and indicates the generality of spherical transport through nanotubes.  相似文献

We report the field-orientation dependent specific heat of the spin-triplet superconductor Sr2RuO4 under the magnetic field aligned parallel to the RuO2 planes with high accuracy. Below about 0.3 K, striking fourfold oscillations of the density of states reflecting the superconducting gap structure have been resolved for the first time. We also obtained strong evidence of multiband superconductivity and concluded that the superconducting gap in the active band, responsible for the superconducting instability, is modulated with a minimum along the [100] direction.  相似文献

Mass excesses of short-lived A=2Z-1 nuclei (63)Ge, (65)As, (67)Se, and (71)Kr have been directly measured to be -46,921(37), -46,937(85), -46,580(67), and -46,320(141) keV, respectively. The deduced proton separation energy of -90(85) keV for (65)As shows that this nucleus is only slightly proton unbound. X-ray burst model calculations with the new mass excess of (65)As suggest that the majority of the reaction flow passes through (64)Ge via proton capture, indicating that (64)Ge is not a significant rp-process waiting point.  相似文献
