A liquid chromatography-electrospray ionization mass spectrometry (LC-ESI-MS) method was developed and validated for the assay of glucosamine sulfate in human plasma. Plasma proteins were precipitated by acetonitrile, followed by vortex mixing and centrifugation. The supernatant was transferred and derivatized with phenyl iso-thiocyanate in acetonitrile at 60 degrees C for 40 min. Chromatographic separation was performed on a C(18) column (Inertsil ODS-3 150 x 2.1 mm i.d., 5 microm, JP) with a mobile phase gradient consisting of 0.2% acetic acid (aqueous) and methanol at a flow-rate of 0.3 mL/min. MS detection using electrospray ionization (ESI) as an interface was used in single ion monitoring mode to determine positive ions at m/z 297. This method was shown to be selective and sensitive for glucosamine sulfate. The limit of detection was 35 ng/mL for glucosamine sulfate in plasma and the linear range was 0.1-20 microg/mL in plasma with a correlation coefficient (r) of 0.9991. The relative standard deviations (RSDs) of intra-day and inter-day assays were 8.7-11.4 and 9.8-12.6%, respectively. Extraction recoveries of glucosamine sulfate in plasma were greater than 73%. This method proved to be simple, reproducible and feasible for pharmacokinetic studies of glucosamine sulfate in healthy volunteers after a single oral administration (1500 mg). The pharmacokinetic parameters and relative bioavailabilities were investigated for both domestic glucosamine sulfate tablet and capsule preparations compared with an imported capsule product.  相似文献

The great performance of a fibrous bed bioreactor (FBB) is mainly dependent on the cell adhesion and immobilization into the fibrous matrix. Therefore, understanding the mechanism and factors controling cell adhesion in the fibrous matrix is necessary to optimize the FBB setup and further improve the fermentability. The adhesion behavior of a strain of Clostridium tyrobutyricum isolated from an FBB was studied, which was proven to be affected by the different environmental conditions, such as growth phase of cells, pH, ionic strength, ionic species, and composition of media. Our results also suggested that electrostatic interactions played an important role on bacteria adhesion into the fibrous matrix. This study demonstrated that the compositions of fermentation broth would have a significant effect on cell adhesion. Consequently, a two-stage glucose supply control strategy was developed to improve the performance of FBB with higher viable cell density in the operation of the FBB setup.  相似文献

The title reactions occur stepwise, the first and fastest being MeReO3 + Eu2+ --> Re(VI) + Eu3+ (k298 = 2.7 x 10(4) L mol(-1) s(-1)), followed by rapid reduction of Re(VI) by Eu2+ to MeReO2. The latter species is reduced by a third Eu2+ to Re(IV), a metastable species characterized by an intense charge transfer band, epsilon410 = 910 L mol(-1) cm(-1) at pH 1; the rate constant for its formation is 61.3 L mol(-1) s(-1), independent of [H+]. Yet another reduction step occurs, during which hydrogen is evolved at a rate v = k[Re(IV)][Eu2+][H+](-1), with k = 2.56 s(-1) at mu = 0.33 mol L(-1). The 410 nm Re(IV) species bears no ionic charge on the basis of the kinetic salt effect. We attribute hydrogen evolution to a reaction between H-ReVO and H3O+, where the hydrido complex arises from the unimolecular rearrangement of Re(III)-OH in a reaction that cannot be detected directly. Chromium(II) ions do not evolve H2, despite E(Cr) degrees approximately E(EU) degrees. We attribute this lack of reactivity to the Re(IV) intermediate being captured as [Re(IV)-O-Cr(III)]2+, with both metals having substitutionally inert d3 electronic configurations. Hydrogen evolution occurs in chloride or triflate media; with perchlorate present, MeReO2 reduces perchlorate to chloride, as reported previously [Abu-Omar, M. M.; Espenson, J. H. Inorg. Our study has demonstrated for the first time that zirconia nanoparticles modified by a simple carboxylic surfactant of a very long alkyl chain can significantly enhance the activity of the immobilized lipases for asymmetric synthesis in organic media. Zirconia nanoparticles of ca. 20 nm diameter were grafted with carboxylic surfactant modifiers from Tween 85 and erucic acid. The surface of nanoparticles was successfully changed from hydrophilic to hydrophobic. Lipases from Candida rugosa and Pseudomonas cepacia were immobilized on the modified zirconia nanoparticles by adsorption in aqueous solution. The immobilized lipases were used for the resolution of ( R, S)-ibuprofen and ( R, S)-1-phenylethanol through esterification and acylation, respectively, in isooctane organic solvent. When immobilized on erucic acid-modified zirconia, both lipases gave significantly higher activity and enantioselectivity compared with those from their corresponding crude lipase powders. The nanohybrid biocatalysts are stable and can be reused for eight cycles without loss in activity and selectivity. The interaction between the hydrophobic surface of zirconia support and lipases probably induces the conformational rearrangement of lipases into an active, stable form.  相似文献

One-electron oxidation of alcohols such as methanol, ethanol, and 2-propanol by 1,3,5-trimethoxybenzene radical cation (TMB*+) in the excited state (TMB*+*) was observed during the two-color two-laser flash photolysis. TMB*+ was formed by the photoinduced bimolecular electron-transfer reaction from TMB to 2,3,5,6-tetrachlorobenzoquinone (TCQ) in the triplet excited-state during the first 355-nm laser flash photolysis. Then, TMB*+* was generated from the selective excitation of TMB*+ during the second 532 nm laser flash photolysis. Hole transfer rate constants from TMB*+* to methanol, ethanol, and 2-propanol were calculated to be (5.2 +/- 0.5) x 10(10), (1.4 +/- 0.3) x 10(11), and (3.2 +/- 0.6) x 10(11) M-1 s-1, respectively. The order of the hole transfer rate constants is consistent with oxidation potentials of alcohol. Formation of TCQH radical (TCQH*) with a characteristic absorption peak at 435 nm was observed in the microsecond time scale, suggesting that deprotonation of the alcohol radical cation occurs after the hole transfer and that TCQ radical anion (TCQ*-), generated together with TMB*+ by the photoinduced electron-transfer reaction, reacts with H+ to give TCQH*.  相似文献

This paper studies the propagation of disturbances superimposedon a pre-stressed incompressible hyperelastic thin rod. Startingfrom the incremental equations given by Haughton and Ogden (1979,J. Mech. Phys. Solids 27, 179-212 and 489-512), we derive athree parameter-dependent one-dimensional rod equation as thegoverning equation. In particular, it is found that one parameterplays a crucial role. Depending on whether it is larger or smallerthan or equal to a critical value, the shear-wave velocity islarger or smaller than or equal to the bar-wave velocity. Inthe case that these two velocities are equal, there exist travelling-wavesolutions of arbitrary form. This implies that for this particularcase the initial disturbance would propagate along the rod withoutdistortion. To see the influence of the pre-stress in detail,we further consider an initial-value problem with an initialsingularity in the shear strain. The solutions are expressedin terms of integrals through the method of Fourier transform.We then conduct an asymptotic analysis for the solutions. Fora material point in a neighbourhood behind the shear-wave front,the phase function of these integrals has a stationary pointat infinity. Here, we use a technique of uniform asymtotic expansionto handle this case. An asymptotic expansion, correct up toorder O(t-1), for the shear strain, which is uniformly validin a neighbourhood behind the shear-wave front, is derived.For material points in other spatial domains, the method ofstationary point is applicable, and asymptotic expansions (correctup to order O(t-1)) are obtained. A novelty is that we are ableto deduce precise qualitative information about the waves inthe far field from our analytic results. Wave profiles for twoconcrete examples are also provided.  相似文献

A general synthesis of 2-fluoro-1-naphthols in two steps from 1-indanones is reported. The 1-indanones are first converted to difluoromethyl 2-fluoro-1-napthyl ethers by reaction with difluorocarbene source, trimethylsilyl 2-fluorosulfonyl-2,2-difluoroacetate (TFDA). These ethers are then converted in high yield to the respective naphthols by heating with a mixture of acetic acid and 48% HBr.  相似文献
