Uneven flow in free‐flow electrophoresis (FFE) with a gravity‐induced fraction collector caused by air bubbles in outlets and/or imbalance of the surface tension of collecting tubes would result in a poor separation. To solve these issues, this work describes a novel collector for FFE. The collector is composed of a self‐balance unit, multisoft pipe flow controller, fraction collector, and vacuum pump. A negative pressure induced continuous air flow rapidly flowed through the self‐balance unit, taking the background electrolyte and samples into the fraction collector. The developed collector has the following advantages: (i) supplying a stable and harmonious hydrodynamic environment in the separation chamber for FFE separation, (ii) effectively preventing background electrolyte and sample flow‐back at the outlet of the chamber and improving the resolution, (iii) increasing the preparative scale of the separation, and (iv) simplifying the operation. In addition, the cost of the FFE device was reduced without using a multichannel peristaltic pump for sample collection. Finally, comparative FFE experiments on dyes, proteins, and cells were carried out. It is evident that the new developed collector could overcome the problems inherent in the previous gravity‐induced self‐balance collector.  相似文献

针对新一代战斗机超声速内埋武器弹射投放分离安全性问题,采用高速风洞投放实验技术研究内埋武器从开式武器舱弹射投放分离动态运动过程,风洞投放模型试验过程中采用除垂直加速度不足外,其余全部运动严格相似的轻模型相似设计方法,并针对轻模型法垂直加速度不足所导致的投放垂直位移偏离实物位移问题,采用一种简单易行的公式修正法进行补偿,试验给出了不同初始弹射投放分离条件下,内埋武器从载机投放分离后运动轨迹与姿态角随分离时间的变化规律,试验马赫数$Ma = 1.5$.研究结果表明:初始投放分离角速度对内埋武器投放分离后的运动轨迹及姿态角有较大的影响,当初始投放分离角速度$\omega _{z0}^s = 0^\circ/{\rm s}$时,内埋导弹出舱后先向下运动远离载机的流场干扰区,之后逐渐向载机方向抬升靠近并最终碰撞载机,高速风洞投放试验结果是不安全的,但经过公式修正后投放试验结果比较乐观,垂直方向运动仍然一直下降远离载机,这说明采用高速风洞投放试验得出的导弹不安全投放分离对真实载机来说不一定会出现,高速风洞投放试验结果比较保守. 当初始投放分离角速度$\omega _{z0}^s= 15^\circ/{\rm s}$和$\omega _{z0}^s = 30^\circ/{\rm s}$时,内埋导弹投放分离后运动趋势几乎一致, 均没出现向载机靠近的现象,内埋导弹具有一定的初始投放分离角速度有利于内埋武器的安全分离.   相似文献

During the process of metabolomics profiling by using ultra high performance liquid chromatography coupled with time‐of flight mass spectrometry, blood sample pretreatment is a crucial step for biomarker discovery. Herein, in order to prevent the potential loss of metabolites and ion suppression phenomena caused by the proteins and phospholipids contained in blood fluids, a delipidation‐based solid‐phase extraction pretreatment technique for plasma broad‐coverage metabolomic profiling was performed. This technique can be summarized as a single extraction, a single elution of solid‐phase extraction plate, followed by four times measuring with electrospray ionization in positive and negative ion mode, respectively. This approach significantly increased the number of features detected in plasma, and 1572 features in positive mode and 1352 features in negative mode were detected, respectively. Besides, the stability and repeatability of the approach were greatly improved. For these advantages, the approach was employed to elucidate the potential pathogenesis of yeast‐induced fever in rats. The biomarkers associated with the pathogenesis of fever were shown to be related to amino acids metabolism and lipid metabolism. The delipidation‐based solid‐phase extraction pretreatment approach can provide a useful tool to reveal the pathological mechanisms of such systemic pathological process.  相似文献

A new complex of copper(II) picrate (pic) with 1, 3‐bis(1‐allaylbenzimidazol‐2‐yl)‐2‐oxopropane (aobb), with the composition [Cu(aobb)2](pic)2, was synthesized and characterized. The crystal structure of the copper(II) complex revealed that the coordination environment around the central copper(II) atom is a distorted octahedral arrangement. Electronic absorption spectroscopy, ethidium bromide displacement experiments and viscosity measurements indicate that the ligand and the CuII complex can strongly bind to calf thymus DNA, presumably by an intercalation mechanism. Furthermore, the antioxidant activity of the CuII complex was determined by superoxide and hydroxyl radical scavenging method in vitro, which indicate that the CuII complex has the activity to suppress OH · and O2 · –.  相似文献

An efficient synthesis of N‐substituted indole derivatives was realized by combining the Pd‐catalyzed one‐pot multicomponent coupling approach with cleavage of the C(sp3)?N bonds. Three or four components of aryl iodides, alkynes, and amines were involved in this coupling process. The cyclopentadiene–phosphine ligand showed high efficiency. A variety of aryl iodides, including cyclic and acyclic tertiary amino aryl iodides, and substituted 1‐bromo‐2‐iodobenzene derivatives could be used. Both symmetric and unsymmetric alkynes substituted with alkyl, aryl, or trimethylsilyl groups could be applied. Cyclic secondary amines such as piperidine, morpholine, 4‐methylpiperidine, 1‐methylpiperazine, 2‐methylpiperidine, and acyclic amines including secondary and primary amines all showed good reactivity. Further application of the resulting indole derivatives was demonstrated by the synthesis of benzosilolo[2,3‐b]indole.  相似文献

In this article, both thermal buckling and post-buckling of pinned–fixed beams resting on an elastic foundation are investigated. Based on the accurate geometrically non-linear theory for Euler–Bernoulli beams, considering both linear and non-linear elastic foundation effects, governing equations for large static deformations of the beam subjected to uniform temperature rise are derived. Due to the large deformation of the beam, the constraint forces of elastic foundation in both longitudinal and transverse directions are taken into account. The boundary value problem for the non-linear ordinary differential equations is solved effectively by using the shooting method. Characteristic curves of critical buckling temperature versus elastic foundation stiffness parameter corresponding to the first, the second, and the third buckling mode shapes are plotted. From the numerical results it can be found that the buckling load-elastic foundation stiffness curves have no intersection when the value of linear foundation stiffness parameter is less than 3000, which is different from the behaviors of symmetrically supported (pinned–pinned and fixed–fixed) beams. As we expect that the non-linear foundation stiffness parameter has no sharp influence on the critical buckling temperature and it has a slight effect on the post-buckling temperature compared with the linear one.  相似文献
