This paper deals with strategies for computing efficiently the propagation of sound waves in ducts containing passive components. In many cases of practical interest, these components are acoustic cavities which are connected to the duct. Though standard Finite Element software could be used for the numerical prediction of sound transmission through such a system, the method is known to be extremely demanding, both in terms of data preparation and computation, especially in the mid-frequency range. To alleviate this, a numerical technique that exploits the benefit of the FEM and the BEM approach has been devised. First, a set of eigenmodes is computed in the cavity to produce a numerical impedance matrix connecting the pressure and the acoustic velocity on the duct wall interface. Then an integral representation for the acoustic pressure in the main duct is used. By choosing an appropriate Green?s function for the duct, the integration procedure is limited to the duct–cavity interface only. This allows an accurate computation of the scattering matrix of such an acoustic system with a numerical complexity that grows very mildly with the frequency. Direct measurements of individual head-related transfer functions (HRTFs) with a probe microphone at the eardrum are unpleasant, risky, and unreliable and therefore have not been widely used. Instead, the HRTFs are commonly measured from the blocked ear canal entrance, which excludes the effects of the individual ear canals and eardrums. This paper presents a method that allows obtaining individually correct magnitude frequency responses of HRTFs at the eardrum from pressure-velocity (PU) measurements at the ear canal entrance with a miniature PU sensor. The HRTFs of 25 test subjects with nine directions of sound incidence were estimated using real anechoic measurements and an energy-based estimation method. To validate the approach, measurements were also conducted with probe microphones near the eardrums as well as at blocked ear canal entrances. Comparisons between the different methods show that the method presented is a valid and reliable technique for obtaining magnitude frequency responses of HRTFs. The HRTF filters designed using the PU measurements are also shown to yield more correct frequency responses at the eardrum than the filters designed using measurements from the blocked ear canal entrance. 相似文献

The recently discovered SAFit class of inhibitors against the Hsp90 co‐chaperone FKBP51 show greater than 10 000‐fold selectivity over its closely related paralogue FKBP52. However, the mechanism underlying this selectivity remained unknown. By combining NMR spectroscopy, biophysical and computational methods with mutational analysis, we show that the SAFit molecules bind to a transient pocket in FKBP51. This represents a weakly populated conformation resembling the inhibitor‐bound state of FKBP51, suggesting conformational selection rather than induced fit as the major binding mechanism. The inhibitor‐bound conformation of FKBP51 is stabilized by an allosteric network of residues located away from the inhibitor‐binding site. These residues stabilize the Phe67 side chain in a dynamic outward conformation and are distinct in FKBP52, thus rationalizing the basis for the selectivity of SAFit inhibitors. Our results represent a paradigm for the selective inhibition of transient binding pockets. 相似文献

A microfluidic nebulizer chip for mass spectrometry is presented. It is an all-glass device which consists of fusion bonded Pyrex wafers with embedded flow channels and a nozzle at the chip edge. A platinum heater is located on the wafer backside. Fabrication of the chip is detailed, especially glass deep etching, wafer bonding, and metal patterning. Various process combinations of bonding and metallization have been considered (anodic bonding vs. fusion bonding; heater inside/outside channel; metallization before/after bonding; platinum lift-off vs. etching). The chip vaporizes the liquid sample (0.1-10 microL min(-1)) and mixes it with a nebulizer gas (ca. 100 sccm N2). Operating temperatures can go up to 500 degrees C ensuring efficient vaporization. Thermal insulation of the glass ensures low temperatures at the far end of the chip, enabling easy interconnections. 相似文献
