In this work, we describe an optimized procedure based on gradual hemolysis for the isolation of hemoglobin derived from bovine
slaughterhouse erythrocytes in a membrane bioreactor. The membrane bioreactor system that provided high yields of hemoglobin
(mainly oxyhemoglobin derivate) and its separation from the empty erythrocyte membranes (ghosts) was designed at a pilot scale.
Ten different concentrations of hypotonic media were assessed from the aspect of the extent of hemolysis, hematocrit values
of the erythrocyte suspensions, cell swelling, and membrane deformations induced by decreased salt concentration. Effective
gradual osmotic hemolysis with an extent of hemolysis of 88% was performed using 35 mM Na-phosphate/NaCl buffer of pH 7.2–7.4.
Under these conditions most of the cell membranes presented the appearance of the normal ghosts under phase contrast microscope.
The hemoglobin purity of >80% was confirmed by SDS-PAGE. Kinetic studies showed that maximal concentration of hemoglobin was
reached after 40 min, but the process cycle at which recovery of 83% was achieved lasted for 90 min. The dynamics of both
steps, (1) transport through the membrane of erythrocytes during process of hemolysis and (2) transport through the reactor

Summary. In the present work, rutin (3,3′ ,4′ ,5,7-pentahydrohyflavone-3-rhamnoglucoside) was determinated via a complexing reaction with a titanyloxalate anion. K2[TiO(C2O4)2] and rutin react in 50% ethanol forming a 1:2 complex in a pH range from 4.00 to 11.50, in which the TiO(C2O4)2
2− ion is linked to rutin through the 4-carbonyl and 5-hydroxyl group. The thermodynamic stability constant log β2
0 of the complex is determined to 10.80 at pH = 6.50. The change of the standard Gibbs free energy Δ G0 amounts to −61 kJċ mol−1, indicating that the process of complex formation is spontaneous. The optimal conditions for the spectrophotometric determination
of microconcentrations of rutin are at pH=6.40 and λ= 430 nm, where the complex shows an absorption maximum with a molar absorption coefficient a

In this work, room temperature interaction of n-hexane with HZSM-5 (Si/Al=20) and ion-exchanged
samples containing one (CuZSM-5, FeZSM-5 and MnZSM-5) or two transition-metal
cations (Fe,CuZSM-5; Cu,MnZSM-5 and Fe,MnZSM-5) was studied by microcalorimetry
and TPD methods. Both differential heats and the amounts of n-hexane
adsorbed per one unit cell were quantitatively determined. Higher heats of
adsorption and higher amounts of adsorbed gas were found for ion-exchanged
samples than for HZSM-5. The experiments of n-hexane
adsorption on hydrated samples were also performed. The amounts of n-hexane
adsorbed on hydrated ZSM-5 were lower in comparison with dehydrated samples,

Currant fruit extracts were characterized by (2,2-diphenyl-1-picrylhydrazyl) radical scavenging activity, 2,2-azinobis-3 ethyl benzothiazoline-6-sulfonic acid cation decolorization activity, total reducing power, cupric ion reducing antioxidant power, and ferric ion reducing antioxidant power (FRAP) assays to evaluate their antioxidant activity. All five antioxidant assays revealed the highest antioxidant activity to be present for the black currants. The highest concentrations of phenolics were present in the black currants (1690?±?10?mg gallic acid equivalents (GAE)/100?g fresh weight), while the lowest value was obtained in the white currants (579?±?5?mg GAE/100?g fresh weight). The correlation between the total phenolic content and antioxidant activity assays was evaluated using regression analysis. A significant positive correlation was obtained between the total phenols and the cupric ion reducing antioxidant power (r?=?0.97, p?0.05), followed by that between the total phenols and FRAP (r?=?0.93, p?0.05). Hierarchical cluster analysis separated these selected fruit species into two statistically significant clusters with black currants as a distinctive cluster. 相似文献

An iron(III) complex with the hexadentate ligand 1,3-propanediamine-N,N′-diacetate-N,N′-di-3-propionate (1,3-pddadp) was prepared, chromatographically isolated as its isomer trans(O5O6)-Cs[Fe(1,3-pddadp)] · 2H2O, and characterized. The trans(O5O6) configuration of the iron(III) compound was found to dominate and this geometry was established by means of IR spectroscopy
and Density Functional Theory (DFT). Structural data correlating the octahedral geometry of the [Fe(1,3-pddadp)]− unit and an extensive strain analysis are discussed in relation to the information obtained for similar complexes. Antibacterial

Abstract Although strawberry tree (Arbutus unedo L.) leaves have long been used as a herbal remedy, insufficient information is available on their nephrotoxicity. We assessed the safety of strawberry tree water leaf extract and its key component arbutin, administered per os to Lewis rats of both genders at 200?mg/kg b.w./day for 14 and 28 days. The effects of the tested compounds on DNA integrity in renal cells was evaluated using alkaline comet assay, while kidney function was studied using serum creatinine and urea levels. Strawberry tree water leaf extract showed high biocompatibility with kidney tissue. It did not impair DNA integrity of renal cells and kidney function, either in male or female rats. However, exposure to single arbutin affected the levels of primary DNA damage in renal cells which could be related to metabolic conversion of arbutin into hydroquinone, whose nephrotoxicity has previously been proven. 相似文献
