Specific carbon-13 labelling of leucine residues in human growth hormone.

Biosynthetic human growth hormone specifically 13C-labelled in the carbonyl positions of all 26 leucine residues has been obtained by recombinant DNA techniques using 13C-labelled leucine and an E. coli strain that requires leucine. It is shown that, on the whole, the labelling is specific with no significant mislabelling as would have been the case had the 13C-labelled leucine been metabolized.     

The fluorite-related anion-excess structure of CeN0.222O0.667F1.333: Ordering of defect clusters

This investigation presents the preparation of CeN_0.222O_0.667F_1.333 by a solid-state reaction from a mixture of CeN:CeF₃:CeO₂ = 1:2:1.5 and its structural investigation. The samples were annealed at 900°C in platinum tubes for different times. The basic structure found by powder neutron diffraction is anion-excess fluorite-related. The unit cell is an orthorhombic distortion of the cubic fluorite cell and has the space group Abm2. The lattice constants are a = 577.71(2) pm, B = 572.76(5) pm, and c = 573.32(6) pm. The structure refined by Rietveld analysis shows that [1:0:2]- defect clusters are present. In samples prepared by longer annealing times an ordering of these clusters to larger aggregates, i.e., toward the vernier phases, was observed. This was deduced from full profile analysis without refining a structural model by comparing the instrumental resolution curves of several models.

Deposition of amorphous hydrogenated semiconductors by magnetron assisted silane decomposition

Magnetron assisted silane decomposition (MASD) is proposed as a method for deposition of a-Si:H and its alloys. In this method a silane containing gas mixture is passed through the magnetron plasma near a target and decomposed there. The deposition rate in the case of the c-Si target is increased 3 times compared to magnetron sputtering and film properties are changed. a-SiSn:H is obtained with a Sn target.     

Average and local magnetic moments on Fe atoms in microcrystalline and amorphous Fe100−xPx alloys

We have measured the concentration dependence of the average magnetic moment per Fe atom _Fe(x) in microcrystalline and amorphous Fe-P alloys obtained over a wide concentration range using electrochemical deposition. The model of local magnetic moments has been used to described _Fe(x). On the basis of this model the effects of phosphorus on the value _Fe are explained in terms of the parameters of the local environment of the Fe atom.

The influence of synthetic conditions on the stability of methotrexate-monoclonal antibody conjugates determined by reversed phase high performance liquid chromatography.

Methotrexate (MTX) has been convalently attached to an IgG-type monoclonal antibody (791T/36) directed to tumour-associated antigen gp72. Conjugates were synthesized by the active ester method using MTX N-succinimidyl ester at various pH values (7.5-10.5). Following purification by gel filtration, high performance liquid chromatography was used to assess the free drug or its derivatives in samples of MTX-791T/36 conjugates previously treated (or not) with hydroxylamine. Quantitative analysis, performed on a reversed phase column (pore size 300 A) with isocratic acetonitrile-sodium acetate buffer (pH 4.8) as mobile phase, indicated no detectable amount of free methotrexate in hydroxylamine-treated conjugates even six months after their preparation. Similar observations were made with conjugates, whose synthesis were performed at pH greater than or equal to 10. In contrast, the presence of increasing amounts of drug/metabolite could be demonstrated in samples produced at lower pH values. Based on these findings, the pH-dependent kinetics of MTX release has been determined and used to design conditions under which stable MTX-791T/36 conjugates could be prepared without hydroxylamine reaction.
