Piperazine-based buffers for liposome coating of capillaries for electrophoresis

Anionic liposomes can be coated on fused-silica capillaries for electrophoresis in the presence of N-(hydroxyethyl)piperazine-N'-(2-ethanesulfonic acid) (HEPES) as background electrolyte (BGE) solution. In this work, the interaction of various compounds with zwitterionic and anionic phospholipid coatings was studied with HEPES at pH 7.4 as BGE solution. The chromatographic and electrophoretic behavior of three test sample solutions (anionic, cationic, and neutral) was investigated for evaluation of the phospholipid coatings. Our results show that hydrophobic interactions between analytes and the phospholipid coating are important for the migration of charged analytes. In addition, the performances of other piperazine-based buffers, i.e., N-(2-hydroxyethyl)piperazine-N'-(2-hydroxypropanesulfonic acid), piperazine-N,N'-bis(2-ethanesulfonic acid), and piperazine-N,N'-bis(hydroxypropane sulfonic acid), at pH 7.4, as liposome solvent and BGE solution were evaluated and compared with the performance of HEPES at pH 7.4. The anionic liposome solution comprised 80/20 mol% phosphatidylcholine/phosphatidylserine. A simple test solution was selected and the chromatographic and electrophoretic migration behavior of the analytes was evaluated. The results show that, in addition to HEPES, other piperazine-based buffers at pH 7.4 are suitable for coating of fused-silica capillaries with anionic liposomes.     

CHARACTERIZATION OF THE RED LIGHT INDUCED REDUCTION OF A PARTICLE ASSOCIATED b-TYPE CYTOCHROME FROM CORN IN THE PRESENCE OF METHYLENE BLUE*

Methylene blue transfers electrons to a membrane-associated b-type cytochrome in particulate fractions from corn coleoptiles. The K_m for methylene blue is less than 1 µM under optimal conditions. This reaction is destroyed by boiling, but not by 7 M urea. Kinetic analyses of the influence of light intensity on cytochrome reduction suggest that a first order photochemical reaction is limiting. Free EDTA may serve as an electron donor in this system at least at high methylene blue and protein concentrations. The photoactivity does not coincide either with mitochondrial or endoplasmic reticulum markers, and may be localized in plasma membrane. There is an estimated 5 times 10^-10 mol photoreducible cytochrome per g coleoptile tissue. Studies on the effect of pH on the reaction in the presence of methylene blue or thionine indicate that dye photoreduction itself is not rate-limiting. Wavelength dependence studies suggest that it is methylene blue monomer and not dimer which mediates the reaction. Although oxygen is apparently required for the reaction, neither superoxide nor excited singlet oxygen appear to be involved. The reaction mechanism is still unknown.     

Scanning electron microscopic studies on different habits of gel grown doped manganese carbonate crystals

The effect of impurities on the growth of manganese carbonate crystals has been studied. Stereophotogrammetric analysis showed that all crystal faces have the same Miller indices and the peanut-shaped forms are twins with twin axis 0001.     

Über assoziierte lineare und lokalkonvexe Topologien

Certain properties E of linear topological or locally convex spaces induce a functor in the corresponding category, which assigns to every space (X,F) an associated topologyF ^E. The well-known notions of the coarsest barrelled topology stronger than a given locally convex topology or of the strongest locally convex topology weaker than a given linear topology are examples of this concept. In the first two parts of this paper we consider the problem, whether the above functors commute with other processes, such as forming products, linear and locally convex direct sums, inductive limits and completions. With help of two technical lemmas we prove in the third part, that every separated locally convex space is a quotient of a complete locally convex space, in which every bounded set has a finite dimensional linear span. This sharpens results of Y. Kōmura [12], M. Valdivia [18] and W.J. Wilbur [20].     

Electronic transport in graphene nanostructures on SiO2

We report two experiments on graphene nanostructures. The first was performed on a graphene nanoribbon, where the nature of electronic transport was investigated in detail. Electrons or holes are found to localize in pockets of the potential along the ribbon. Transport is governed by the joint action of localization and Coulomb interaction. The temperature-dependence of the conductance shows activated behavior at temperatures above a few Kelvin. The activation energy retraces the edges of Coulomb blockade diamonds found in nonlinear transport. In the second experiment the metallic tip of a low-temperature scanning force microscope was scanned above a graphene quantum dot. In addition to the familiar Coulomb blockade fringes, localized states are detected forming in the constrictions connecting the dot to source and drain.     

Total and Semi‐Syntheses of Antimicrobial Thuggacin Derivatives

The total and semi‐synthesis of 13 new macrolactones derived from thuggacin, which is a secondary metabolite from the myxobacterium Sorangium cellulosum, are reported. The thuggacins have attracted much attention due to their strong antibacterial activity, particularly towards Mycobacterium tuberculosis. This study focuses on 1) thuggacin derivatives that cannot equilibrate by transacylation between the three natural thuggacins A–C, 2) the roles of the thiazole ring, and 3) the hexyl side chain at C2. Semi‐synthetic O‐methylation at C17 suppressed the transacylations without a substantial loss of antibacterial activity. Exchanging the C17–C25 side chain for simplified hydrophobic chains led to complete loss of antibacterial activity. Exchange of the thiazole by an oxazole ring or removal of the hexyl side chain at C2 had no substantial effect on the biological properties.     

The impact of blood on liver metabolite profiling – a combined metabolomic and proteomic approach

Metabolomics has entered the well‐established omic sciences as it is an indispensable information resource to achieve a global picture of biological systems. The aim of the present study was to estimate the influence of blood removal from mice liver as part of sample preparation for metabolomic and proteomic studies. For this purpose, perfused mice liver tissue (i.e. with blood removed) and unperfused mice liver tissue (i.e. containing blood) were compared by two‐dimensional gas chromatography time of flight mass spectrometry (GC × GC‐TOFMS) for the metabolomic part, and by liquid chromatography tandem mass spectrometry (LC‐MS/MS) for the proteomic part. Our data showed significant differences between the unperfused and perfused liver tissue samples. Furthermore, we also observed an overlap of blood and tissue metabolite profiles in our data, suggesting that the perfusion of liver tissue prior to analysis is beneficial for an accurate metabolic profile of this organ.
