Given a sequence of random variables taking values in {?v,…,0,…,+u}, let's define the local score of the sequence by Hn=max1?i?j?n(∑k=ijXk). The local score is used to analyze biological sequences pointing out regions of the sequences with interesting biological properties. In order to separate randomly events from really interesting segments, we establish here the distribution of the local score of Hn when the sequence is a Markov chain of order 1. Over the past decade, RNA conformation has been shown to respond to external stimuli. Thus, dependent on the presence of a high affinity ligand, specifically designed ribozymes can be regulated in a classical allosteric way. In this scenario, a binding event in one part of the RNA structure induces conformational changes in a separated part, which constitutes the catalytic centre. As a result activity is switched on (positive regulation) or off (negative regulation). We have developed a hairpin aptazyme responding to flavine mononucleotide (FMN). Ribozyme activity is dependent on binding of FMN and thus is switched on in the presence of FMN in its oxidized form. Under reducing conditions, however, FMN changes its molecular geometry, which is associated with loss of binding and consequently down‐regulation of ribozyme activity. While in previous experiments sodium dithionite was used for reduction of FMN, we now present an assay for electrochemically induced activity switching. We have developed an electrochemical microcell that allows for iterative cycles of reduction/oxidation of FMN in an oxygen free atmosphere and thus for reversible switching of ribozyme activity. The reaction proceeds in droplets of 3 to 10 μL at micro‐ to nanomolar concentrations of the reaction components. 相似文献

Basic algorithmic and numerical issues involved in subspace-based linear multivariable discrete-time system identification are described. A new identification toolbox—SLIDENT—has been developed and incorporated in the freely available Subroutine Library in Control Theory (SLICOT). Reliability, efficiency, and ability to solve industrial identification problems received a special consideration. Two algorithmic subspace-based approaches (MOESP and N4SID) and their combination, and both standard and fast techniques for data compression are provided. Structure exploiting algorithms and dedicated linear algebra tools enhance the computational efficiency and reliability. Extensive comparisons with the available computational tools based on subspace techniques show the better efficiency of the SLIDENT toolbox, at comparable numerical accuracy, and its capabilities to solve identification problems with many thousands of samples and hundreds of parameters. 相似文献

Homopolymers of methacrylic acid (MAA), monoethyleneglycol methyl ether methacrylate (MEOMA), diethyleneglycol methyl ether methacrylate (MEO2MA), oligo(ethyleneglycol) methyl ether methacrylate (OEGMA475 and OEGMA1100) and oligo(ethyleneglycol) ethyl ether methacrylate (OEGEMA246) were synthesized with various chain lengths via reversible addition fragmentation chain transfer (RAFT) polymerization. The homopolymers of MAA, MEOMA and OEGMA1100 did not show any cloud point (CP) in the range of 0–100 °C, whereas at a pH value of 7, the CPs were found to be 20.6, 93.7, and 20.0 °C for p(MEO2MA), p(OEGMA475) and p(OEGEMA246), respectively, with an initial monomer to initiator ratio of 50. Furthermore, statistical copolymer libraries of MAA with OEGMA475 and OEGMA1100 were prepared. The cloud points of the random copolymers of MAA and OEGMA475 were found to be in the range of 20–90 °C; surprisingly, even though the homopolymers of MAA and OEGMA1100 did not exhibit any LCST behavior, the copolymers of these monomers at certain molar ratios (up to 40% OEGMA1100) revealed a double responsive behavior for both temperature and pH. Finally, the cloud points were found to be in the range of 22–98 °C, measured at pH values of 2, 4, and 7, while no cloud point was detected at pH 10. © Wiley Periodicals, Inc. The stereoselective synthesis of tetrasubstituted tetrahydrofurans and trisubstituted tetrahydropyrans bearing a sulfoxide was achieved by reductive cyclization (Et3SiH/TMSOTf) from the corresponding enantiopure hydroxy ketones protected as a dioxolane. Biomarker discovery in cerebrospinal fluid (CSF) can provide important information helping to diagnose and monitor disease progression in brain disorders. We present a label-free LC-MS strategy to investigate the proteomic profile of CSF. We provide a framework and protocol addressing quality control, sample replication steps and the adaptation of pattern recognition methods for the detection of experimental variation and (most importantly) putative biomarkers. This strategy was tested using a total of 20 CSF samples (ten samples from healthy volunteers and ten from schizophrenia patients). A clear difference was found between healthy volunteers and schizophrenia patients. With technical and biological variation controlled, we were able to detect 77 proteins with confidence in CSF, of which seven were newly identified, without prior depletion of abundant protein species. Our strategy provides a simple and controlled method for CSF biomarker discovery. 相似文献

Absolute absorption cross sections of the absorption spectrum of the 2nu1 band of the HO2 radical in the near-IR region were measured by continuous wave cavity ring-down spectroscopy (cw-CRDS) coupled to laser photolysis in the wavelength range 6604-6696 cm(-1) with a resolution better than 0.003 cm(-1). Absolute absorption cross sections were obtained by measuring the decay of the HO2 self-reaction, and they are given for the 100 most intense lines. The most important absorption feature in this wavelength range was found at 6638.20 cm(-1), exhibiting an absorption cross section of sigma = 2.72 x 10(-19) cm2 at 50 Torr He. Using this absorption line, we obtain a detection limit for the HO2 radical at 50 Torr of 6.5 x 10(10) cm(-3). 相似文献

The objective of this study was to ascertain whether sequential sampling and isotopic analysis of bovine hooves could be used to reconstruct the dietary history of cattle. A controlled, on-farm experiment was conducted in which cattle were switched from a barley-based diet to an isotopically different diet incorporating maize, urea and seaweed (the isotopic spacing between diets was 13.6 per thousand for delta(13)C and 8.0 per thousand for delta(15)N) and maintained on that diet for 168 days. Postmortem sampling of the cleaned anterior wall of the lateral, left front claw was carried out on five individuals using a micro-drilling technique. From the first 60 mm of each claw, up to 41 samples with a spacing between them of less than 1 mm were collected. Bands were less than 1 mm deep and had a mean width of 1.2 mm. The hoof keratin showed a rapid increase followed by a slower increase in its delta(13)C and delta(15)N values following the diet switch, suggesting that C and N in hoof keratin originate from more than one pool. However, the response of the N isotope composition of the hoof was somewhat delayed compared with that of C. Estimated mean hoof growth rates for these cattle were 10.5 +/- 2.3 mm per month and 6.7 +/- 1.0 mm per month (+/-SD, n = 5) when receiving the barley-based transition diet and the maize-based experimental diet, respectively. These values are considerably higher than previous estimates obtained by visual methods and they suggest that diet may have a greater influence on hoof growth rates than seasonality. These results demonstrate that hooves are a suitable incremental tissue for high-resolution isotopic reconstruction of the dietary history of bovine animals. 相似文献

Most associate liquid crystals with their everyday use in laptop computers, mobile phones, digital cameras, and other electronic devices. However, in contrast to their rodlike (calamitic) counterparts, first described in 1907 by Vorl?nder, disklike (discotic, columnar) liquid crystals, which were discovered in 1977 by Chandrasekhar et al., offer further applications as a result of their orientation in the columnar mesophase, making them ideal candidates for molecular wires in various optical and electronic devices such as photocopiers, laser printers, photovoltaic cells, light-emitting diodes, field-effect transistors, and holographic data storage. Beginning with an overview of the various mesophases and characterization methods, this Review will focus on the major classes of columnar mesogens rather than presenting a library of columnar liquid crystals. Emphasis will be given to efficient synthetic procedures, and relevant mesomorphic and physical properties.
