The medical demand for useful biomarkers is large and still increasing. This is especially true for cancer, because for this disease adequate diagnostic markers with high specificity and sensitivity are still lacking. Despite advances in imaging technologies for early detection of cancer, peptidomic multiplex techniques evolved in recent years will provide new opportunities for detection of low molecular weight (LMW) proteome biomarker (peptides) by mass spectrometry. Improvements in peptidomics research were made based on separation of peptides and/or proteins by their physico-chemical properties in combination with mass spectrometric detection, respectively identification, and sophisticated bioinformatic tools for data analysis. To evaluate the potential of serological tumor marker detection by differential peptide display (DPD) we analyzed plasma samples from a tumor graft model. After subcutaneous injection of HCT-116 cells in immunodeficient mice and their growth to a palpable tumor, plasma samples were analyzed by DPD. The comparison of obtained mass spectrometric data allows discovery of tumor specific peptides which fit well into the biological context of cancer pathogenesis and show a strong correlation to tumor growth. The identified peptides indicate events associated with hyper-proliferation and dedifferentiation of cells from an epithelial origin, which are typical characteristics of human carcinomas. We conclude that these findings are a "proof of principle" to detect differentially expressed, tumor-related peptides in plasma of tumor-bearing mice.  相似文献

The primary excited state processes of phytochrome, the plant chromoprotein responsible for most photomorphogenetic responses, are investigated by picosecond intensity-dependent transmission measurements. At sufficient high excitation intensity (starting at photon flux densities of about 1′1025 cm?2 s?l) a moderate bleaching of the absorption is observed. For the quantitative analysis of the experimental data a rate equation system approach for the energy levels responsible for the ultrafast transformation Pr→ lumi-R and the photon transport through the sample was utilized. The following was found for the excited state parameters of the phytochrome Pr: (1) an excited state absorption of the same order of magnitude as the ground state absorption (cross section: S?exc= 4′10?16 cm?2 at 650 nm), (2) a fast relaxation of the higher excited singlet state on the femtosecond time scale.  相似文献

Most current device applications of ZnO are hampered by the lack of control over the electrical conductivity. As-grown ZnO usually exhibits n-type conductivity, and the cause of this residual doping is heavily debated. We have performed temperature-dependent Hall measurements and material characterization by secondary ion mass spectroscopy on nominally undoped bulk ZnO crystals as well as on material doped with potential candidates for p-conductivity in order to explore the cause of the background doping and to study the impact of possible candidates for p-doping of ZnO. Also, this paper gives an overview about surface conductivity of high-resistivity ZnO bulk material and discusses how this property might impact the difficult search for p-type ZnO. We will demonstrate the effect of a surface conducting channel on homoepitaxial MgZnO layers grown by liquid-phase epitaxy.
