以大孔微球硅胶(30nm,8μm)为基体,经γ-氨丙基三乙氧基硅烷活化与间隔臂氯代环氧丙烷反应后,再与螯合剂亚氨基二乙酸键合,后者与铜离子(Ⅱ)螯合后,构成定位金属离子亲和色谱固定相。用磷酸盐缓冲溶液(pH=7.0)作流动相,经紫外吸收检测(260nm)对核苷酸混合物进行了高效液相色谱分析,取得较满意的结果。 相似文献

The absorption spectra of Co(II) chloride complexes, containing variable concentrations of chloride ligand, in a molten mixture of 80 mol% acetamide–20 mol% calcium nitrate tetrahydrate, were studied at 313, 333, 353, and 363 K, in the wavelength range 400-800 nm. The melt contains three possible ligands (CH3CONH2, H2O, and NO3
-) for competition with added chloride ligand. Addition of chloride caused a shift of the absorption maximum of octahedral cobalt(II) nitrate towards lower energies and pronounced changes in the shape of the initial spectrum of cobalt(II) nitrate. The effect of temperature changes on the molar absorption coefficient of the Co(II) species was dependent on the chloride concentration and was attributed to the structural changes occurring in the cobalt(II) species. The STAR and STAR FA programs were applied to identify the complex ionic species and to calculate the stability constants of Co(II) complexes formed in this solvent. The results indicate the highest probability of formation of the following complex species: Co(NO3)4
2-, Co(NO3)2Cl2
2-, and CoCl4
2-. Stability constants of each complex were presented for the equilibria occurring at 313, 333, 353, and 363 K. Distribution of the Co(II) species was also calculated over the ranges of chloride concentration and temperature investigated. 相似文献

Zusammenfassung Die Reaktion von 2-Nitroso-1-naphthol mit Resorcin bzw. Orcin in Äther bei Anwesenheit von HNO3 liefert Benzo[c]phenoxazon-(9) (I), Benzo[c]phenoxazon-(9)-12-oxid (II), Methyl-benzo[c]phenoxazon-(9) (III) sowie 11-Methyl-benzo[c]phenoxazon-(9)-12-oxid (IV). Die Struktur der isolierten Substanzen wurde durch Reduktion mittels TiCl3 sowie durch die UV- und IR-Spektren gestützt.

The reaction of 2-nitroso-1-naphthol with resorcinol and orcinol in ether solution in the presence of nitric acid has been studied. From the reaction mixture the benzophenoxazones I–IV have been isolated. Structures were assigned on the grounds of UV and IR spectra and partly by using TiCl3 as a reducing agent.

A pressurized gradient capillary electrochromatography (pCEC) instrument was developed to separate 18 amino acid derivatives. A reversed-phase C18 column (3 microm, 130 mm x 75 microm I.D.) and an acetate buffer (50 mmol/l NaAc, pH 6.4) with an ion-pair reagent (1% N,N-dimethylformamide) were used to separate derivatized amino acids from a standard solution (2 microg/ml), and the wavelength of the UV-Vis detector was 360 nm. The pressure on the capillary column was kept at approx. 70 Pa and 3 kV positive voltage was added on the outlet end of column. The effect of voltage on the eluting order of amino acids and the resolution of separation were studied, and it was found that when the voltage was higher than 3 kV, the adsorption of amino acids in the porous C18 column occurred. The effect of salt concentration, injection volume, and column length on the separation of amino acids was determined. The amino acid sample was separated by pCEC, and RSDs of the migration times of each amino acid were all less than 2.5%. 相似文献

The mechanisms for the reaction of CH3S with NO2 are investigated at the QCISD(T)/6‐311++G(d,p)//B3LYP/6‐311++G(d,p) on both single and triple potential energy surfaces (PESs). The geometries, vibrational frequencies, and zero‐point energy (ZPE) correction of all stationary points involved in the title reaction are calculated at the B3LYP/6‐311++G(d,p) level. More accurate energies are obtained at the QCISD(T)/6‐311++G(d,p). The results show that 5 intermediates and 14 transition states are found. The reaction is more predominant on the single PES, while it is negligible on the triple PES. Syntheses of the cis-configurated isopenam 9 (Scheme 1), isocephem 14 (Scheme 2), and isocephem 19 (Scheme 3) are described. The key step in the preparation of 14 and 19 involved a Pummerer-type rearrangement of the corresponding sulfoxides 12 and 18 . These β-lactams were found to possess biological activity against several pathogenic microorganisms in vitro. The electronic activation of the lactam moiety of 19 remarkably enhanced its biological activity. A retinoic moiety was attached to 19 via an amino linker. The resultant retinamido-β-lactam 21 showed significant cytostatic activity in tracheal organ cultures obtained from vitamin-A-deficient hamsters. 相似文献

Zusammenfassung 1,3-Dimethyl-4,5-diaminouracil (DAL) hat sich als sehr empfindliches Reagens zum selektiven Nachweis von Co2+-Ionen im stark alkalischen Medium einer Phosphatpufferlösung (pH=11,20) erwiesen. Es wurden vier Additionsverbindungen von DAL mit den Zentralatomen Cd(II), Pd(II), Ni(II) und Co(II) im Verhältnis Me DAL=1 2 hergestellt. Spektrophotometrische Untersuchungen weisen darauf hin, daß bei pH 6 bis 7 eine Komplexverbindung im Verhältnis CoDAL=12 gebildet wird.
Summary 1,3-dimethyl-4,5-diaminouracil (DAL) has been found to be a very sensitive reagent for the selective detection of Co2+ ions in strong alkaline medium of a phosphate buffer solution (pH=11.20). Four addition compounds of DAL with the central atoms Cd(II), Pd(II), Ni(II) and Co(II) with the ratio MeDal=12 have been prepared. An on-line high-performance liquid chromatography/mass spectrometry (HPLC/MS) method is described for the rapid characterization of any type of oligosaccharide released from glycoproteins. The procedure can be applied without further manipulation to fractions collected from a high-performance anion-exchange chromatography-pulse amperometric detection (HPAEC-PAD) system commonly used for glycosylation mapping of glycoproteins, or to a pool of oligosaccharides directly released from glycoproteins. The system consists of a porous graphitized high-performance chromatography column (Hypercarb) coupled to a quadrupole time-of-flight (TOF) mass spectrometer. Oligosaccharides are eluted from the column with a gradient of ammonium acetate/acetonitrile and directly identified following in-source fragmentation. Some applications of the method are presented, as well as information about the spectra and fragmentation behavior observed for N- and O-linked oligosaccharides released from some recombinant glycoproteins. Low femtomole limits of detection are achieved using proper miniaturization. 相似文献
