The droplet-vitrification method was adapted to Pelargonium apices by optimizing the duration of the loading solution (LS) as well as the plant vitrification solution 2 (PVS2). The excised apices were dehydrated in two steps (20 min in LS and 15 min in PVS2) and then immersed directly in liquid nitrogen (LN). After thawing and unloading in the recovery solution at room temperature for 15 min, apices were plated onto semi-solid Murashige and Skoog medium. This simple protocol without any pretreatment was successfully applied to eight cultivars with a survival level ranging between 55.6 - 96.2 percent and a regrowth level between 9.1 and 70.6 percent. These results prove the feasibility of the long-term storage of Pelargonium germplasm through cryopreservation.  相似文献

To establish an improved protocol for the cryopreservation of banana (Musa spp.), a fast-freeze/fast-thaw method was applied and compared to three existing cryogenic procedures, namely, simple freezing, vitrification of proliferating meristems and vitrification of individual meristems. The average post-thaw shoot regeneration of the fast freeze/fast thaw protocol was 51.8 percent and was superior to the existing protocols which were 12.5 percent, 39.2 percent and 42.8 percent, respectively. Other factors such as time and ease of operation were considered for each method. It was found that the fast freeze and fast thaw technique was the best amongst all the four methods tested for the cryopreservation of in vitro banana meristems in the genotype Robusta (AAA).  相似文献

Motivated by recent studies on the fractal nature of folded proteins, we analyze the time-dependent autocorrelation function < x (-->)(t).x(-->)(0) >of the distance between two points on a thermally vibrating fractal. Using fractons, the vibrational excitations of a fractal, we show that for both strongly underdamped and overdamped vibrations this correlation function decays anomalously, displaying a crossover from a nearly stretched exponential decay at short times to a slow algebraic decay at long times. Relationship to single molecule experiments is discussed.  相似文献

Dy-based single-molecule magnets (SMMs) are of great interest due to their ability to exhibit very large thermal barriers to relaxation and therefore high blocking temperatures. One interesting line of investigation is Dy-encapsulating endohedral clusterfullerenes, in which a carbon cage protects magnetic Dy3+ ions against decoherence by environmental noise and allows for the stabilization of bonding and magnetic interactions that would be difficult to achieve in other molecular architectures. Recent studies of such materials have focused on clusters with two Dy atoms, since ferromagnetic exchange between Dy atoms is known to reduce the rate of magnetic relaxation via quantum tunneling. Here, two new dysprosium-containing mixed-metallic sulfide clusterfullerenes, DyScS@Cs(6)–C82 and DyScS@C3v(8)–C82, have been successfully synthesized, isolated and characterized by mass spectrometry, Vis-NIR, cyclic voltammetry, single crystal X-ray diffractometry, and magnetic measurements. Crystallographic analyses show that the conformation of the encapsulated cluster inside the fullerene cages is notably different than in the Dy2X@Cs(6)–C82 and Dy2X@C3v(8)–C82 (X = S, O) analogues. Remarkably, both isomers of DyScS@C82 show open magnetic hysteresis and slow magnetic relaxation, even at zero field. Their magnetic blocking temperatures are around 7.3 K, which are among the highest values reported for clusterfullerene SMMs. The SMM properties of DyScS@C82 far outperform those of the dilanthanide analogues Dy2S@C82, in contrast to the trend observed for carbide and nitride Dy clusterfullerenes.

Mononuclear metalloenzymes in nature can function in cooperation with precisely positioned redox-active organic cofactors in order to carry out multielectron catalysis. Inspired by the finely tuned redox management of these bioinorganic systems, we present the design, synthesis, and experimental and theoretical characterization of a homologous series of cobalt complexes bearing redox-active pyrazines. These donor moieties are locked into key positions within a pentadentate ligand scaffold in order to evaluate the effects of positioning redox non-innocent ligands on hydrogen evolution catalysis. Both metal- and ligand-centered redox features are observed in organic as well as aqueous solutions over a range of pH values, and comparison with analogs bearing redox-inactive zinc(ii) allows for assignments of ligand-based redox events. Varying the geometric placement of redox non-innocent pyrazine donors on isostructural pentadentate ligand platforms results in marked effects on observed cobalt-catalyzed proton reduction activity. Electrocatalytic hydrogen evolution from weak acids in acetonitrile solution, under diffusion-limited conditions, reveals that the pyrazine donor of axial isomer 1-Co behaves as an unproductive electron sink, resulting in high overpotentials for proton reduction, whereas the equatorial pyrazine isomer complex 2-Co is significantly more active for hydrogen generation at lower voltages. Addition of a second equatorial pyrazine in complex 3-Co further minimizes overpotentials required for catalysis. The equatorial derivative 2-Co is also superior to its axial 1-Co congener for electrocatalytic and visible-light photocatalytic hydrogen generation in biologically relevant, neutral pH aqueous media. Density functional theory calculations (B3LYP-D2) indicate that the first reduction of catalyst isomers 1-Co, 2-Co, and 3-Co is largely metal-centered while the second reduction occurs at pyrazine. Taken together, the data establish that proper positioning of non-innocent pyrazine ligands on a single cobalt center is indeed critical for promoting efficient hydrogen catalysis in aqueous media, akin to optimally positioned redox-active cofactors in metalloenzymes. In a broader sense, these findings highlight the significance of electronic structure considerations in the design of effective electron–hole reservoirs for multielectron transformations.  相似文献

We present second-order accurate central finite volume methods adapted to three-dimensional ideal magnetohydrodynamics problems. These methods alternate between two staggered grids, thus leading to Riemann solver-free algorithms with relatively favorable computing times. The div· B = 0 constraint on the magnetic field is enforced with a suitable adaptation of the constrained transport method to our central schemes. Numerical experiments show the feasibility of the proposed methods and our results are in good agreement with existing results in the recent literature. (© 2008 WILEY-VCH Verlag GmbH & Co. Palm cryobanking     
We describe the development of an efficient cryopreservation protocol for proembryogenic masses (PEMs) of date palm variety 'Barhee'. Proembryos were induced by inoculating small pieces of juvenile leaves on MS medium supplemented with 0.3 mg per liter 2,4-D. Application of these in vitro conditions led to true-to-type plants as observed after plant fructification. When compared to the standard vitrification protocol, the ultra-rapid droplet-vitrification technique proved to be superior. Sucrose preculture considerably increased post-cryopreservation recovery. The highest regeneration after cryogenic exposure reached 63.3 percent when PEMs were treated with PVS2 for 30 min at 0 degree C and 56.7 percent when PVS2 treatment lasted for 15 min at 25 degree C. The first signs of regrowth of cryopreserved PEMs were observed after 2 to 3 weeks. Cryopreservation did not affect the morphogenetic capacities of the plant material. Moreover, highly proliferating suspension cultures could be established from the cryopreserved material. The overall production of somatic embryos from 500 mg cryopreserved PEMs reached 1030 +/- 50 units after 1 month. The morphological study of date palms regenerated from cryopreserved material confirmed the stability of clonal material following cryopreservation.  相似文献

We propose two‐dimensional central finite volume methods based on our multidimensional extensions of Nessyahu and Tadmor's one‐dimensional non‐oscillatory central scheme and a constrained transport‐type method to solve ideal magnetohydrodynamic problems (MHD) and shallow water magnetohydrodynamic problems (SMHD). The main numerical scheme is second‐order accurate both in space and time and uses an original Cartesian grid coupled to a Cartesian‐ or diamond‐staggered dual grid to by‐pass the resolution of the Riemann problems at the cell interfaces. To treat the non‐vanishing magnetic field/flux divergence we have constructed an adaptation of Evans and Hawley's constrained transport method specifically designed for central schemes. Ion mobility spectrometry (IMS) has potential analytical applications in very diverse fields such as chemical, petrochemical, environmental, and, more recently, in drug, chemical warfare agent, and explosives detection. Commercially available IMS instruments are based on time-of-flight (TOF) mass spectrometry. IMS is inherently suitable for field operation as it uses relatively simple microfluidic devices and operates at atmospheric pressure. It is portable, highly sensitive with tunable selectivity, yet can be produced at relatively low cost. Key limitations of this analytical detection technique are low duty cycle, ion cluster formation, short linear dynamic range, and restriction to only positive or negative ion collection in a single analysis. Microelectromechanical system, radio frequency modulated IMS (MEMS RF-IMS), also known as differential mobility spectrometry, has recently been developed and commercialized. The technology is based on IMS, and MEMS RF-IMS offers substantially better performance. In this study, the strengths and limitations of the recently introduced differential mobility detector when used with gas chromatography in trace analyses are discussed and illustrated with applications of industrial significance.  相似文献
