Heterochromatin bodies in single- and multichromocentered interphase cell nuclei of Triatoma infestans, a vector of Chagas disease, have been suggested to contain AT-rich DNA, based on their positive response to Q-banding and Hoechst 33248 treatment. No information exists on whether GC-rich DNA is also present in these nuclei and whether it plays a role on chromatin condensation. Considering that methodologies more precise than those previously used to determine DNA base composition in situ are currently available, and that the spatial distribution of chromatin areas differing in composition in interphase cell nuclei of different species is a matter of interest, the localization of AT- and GC-rich DNA in T. infestans nuclei is revisited here. The methodologies used included DAPI/AMD and CMA(3)/Distamycin differential staining, Feulgen-DNA image analysis following Msp I and Hpa II enzymatic digestion, 5-methylcytidine immunodetection, AgNOR response, confocal microscopy, and the 5-aza-2'-deoxycytidine (5-AZA) demethylation assay. The results identified the presence of AT-rich/GC-poor DNA in chromocenters and evenly distributed AT and GC sequences in euchromatin. A GC-rich DNA zone encircling the chromocenters was also found but it could not be associated with NOR regions. To corroborate the DNA AT-richness in T. infestans nuclei, bioinformatic analyses were also performed. Methylated cytosine was evident at some points of the chromocenters' edge in single- and multichromocentered nuclei and at the euchromatin of multichromocentered nuclei and could be transiently affected by the 5-AZA treatment. The present results suggest that in the particular case of chromocenters of the hemipteran T. infestans, cytosine methylation is not a relevant factor involved in chromatin condensation. 相似文献

Thermal ionisation mass spectrometry (TIMS) method has been developed for the simultaneous detection of different cerium isotopes in biological samples (i.e., blood and urine) at very low concentrations. The work has been done in the frame of a biokinetic study, where different stable cerium isotopes have been administered orally and intravenously as tracers to the human body. In order to develop an appropriate detection method for the tracers in the biological samples, an optimum sample preparation technique has been set and adapted to the specific requirements of the analysis technique used, i.e., TIMS. For sample evaporation and ionisation, the double tantalum filament technique showed the best results. The ions produced were simultaneously collected on a secondary electron multiplier so that the isotopic ratios of the cerium isotopes in the biological samples could be measured. The technique has been optimised for the determination of cerium down to 1?ng loaded on the evaporation filament corresponding to cerium concentrations of down to 1?ng?ml(-1) in the blood or urine samples. It has been shown that the technique is reliable in application and enables studies on cerium metabolism and biokinetics in humans without employing radioactive tracers. 相似文献

Cardiac elastography is a useful diagnostic technique for detection of heart function abnormalities, based on analysis of echocardiograms. The analysis of the regional heart motion allows assessing the extent of myocardial ischemia and infarction. In this paper, a new two-stage algorithm for cardiac motion estimation is proposed, where the data is taken from a sequence of 2D echocardiograms. The method combines the advantages of block-matching and optical flow techniques. The first stage employs a standard block-matching algorithm (sum of absolute differences) to provide a displacement estimate with accuracy of up to one pixel. At the second stage, this estimate is corrected by estimating the parameters of a local image transform within a test window. The parameters of the image transform are estimated in the least-square sense. In order to account for typical heart motions, like contraction/expansion, translation and rotation, a local affine model is assumed within the test window. The accuracy of the new algorithm is evaluated using a sequence of 500 grayscale B-mode images, which are generated as distorted, but known copies of an original ROI, taken from a real echocardiogram. The accuracy of the motion estimation is expressed in terms of errors: maximum absolute error, root-mean-square error, average error and standard deviation. The errors of the proposed algorithm are compared with these of the known block-matching technique with cross-correlation and interpolation in the sub-pixel space. Statistical analysis of the errors shows that the proposed algorithm provides more accurate estimates of the heart motion than the cross-correlation technique with interpolation in the sub-pixel space. 相似文献

In the present work we report the synthesis of MoS2 nanotubes with diameters greater than 10 nm using a template method. The length and properties of these nanotubes are a direct result of the preparation method. High-resolution transmission electron microscopy is used to study the structure of these highly curved entities. Molecular dynamics simulations of MoS2 nanotubes reveal that one of the stable forms of the nanotubes is a twisted one. The twisting of the nanotubes produces a characteristic contrast in the images, which is also studied using simulation methods. The analysis of the local contrast close to the perpendicular orientation shows geometrical arrays of dots in domain-like structures, which are demonstrated to be a product of the atomic overlapping of irregular curvatures in the nanotubes. We consider the stability of periodic gravity free-surface water waves traveling downstream at a constant speed over a shear
flow of finite depth. In case the free surface is flat, a sharp criterion of linear instability is established for a general
class of shear flows with inflection points and the maximal unstable wave number is found. Comparison to the rigid-wall setting
testifies that the free surface has a destabilizing effect. For a class of unstable shear flows, the bifurcation of nontrivial
periodic traveling waves is demonstrated at all wave numbers. We show the linear instability of small nontrivial waves that
appear after bifurcation at an unstable wave number of the background shear flow. The proof uses a new formulation of the
linearized water-wave problem and a perturbation argument. An example of the background shear flow of unstable small-amplitude
periodic traveling waves is constructed for an arbitrary vorticity strength and for an arbitrary depth, illustrating that

Background
White matter hyperintensities (WMHs) on MRI have been associated with age, cardiovascular risk factors and falls in the elderly. This study evaluated the relationship between WMHs and dynamics of postural control in older adults without history of falls.Methods
We studied 76 community-living subjects without history of falls (age 64.5±7.3 years). Brain and WMH volume calculations and clinical rating were done on fluid-attenuation inversion recovery (FLAIR) and MP-RAGE MR images on 3 T. Balance was assessed from the center of pressure displacement using the force platform during 3 min of quiet standing using traditional and dynamic measures (using stabilogram-diffusion analysis). Gait speed was measured from 12-min walk.Results
Age-adjusted periventricular and focal WMHs were associated with changes in certain dynamic balance measures, including reduced range of postural sway in anteroposterior direction (fronto-temporal WMHs, P=.045; parieto-occipital WMHs, P=.009) and more irregular long-term mediolateral fluctuations (P=.046). Normal walking speed was not affected by WMHs.Conclusions
Periventricular and focal WMHs affect long-term dynamics of postural control, which requires engagement of feedback mechanisms, and may contribute to mobility decline in the elderly. 相似文献38.
Line-integral approximations to the acoustic path integral have been used to estimate the magnitude of the fluctuations in an acoustic signal traveling through an ocean filled with internal waves. These approximations for the root-mean-square (rms) fluctuation and the bias of travel time, rms fluctuation in a vertical arrival angle, and the spreading of the acoustic pulse are compared here to estimates from simulations that use the parabolic equation (PE). PE propagations at 250 Hz with a maximum range of 1000 km were performed. The model environment consisted of one of two sound-speed profiles perturbed by internal waves conforming to the Garrett-Munk (GM) spectral model with strengths of 0.5, 1, and 2 times the GM reference energy level. Integral-approximation (IA) estimates of rms travel-time fluctuations were within statistical uncertainty at 1000 km for the SLICE89 profile, and in disagreement by between 20% and 60% for the Canonical profile. Bias estimates were accurate for the first few hundred kilometers of propagation, but became a strong function of time front ID beyond, with some agreeing with the PE results and others very much larger. The IA structure functions of travel time with depth are predicted to be quadratic with the form theta(2)vc0(-2)deltaz(2), where deltaz is vertical separation, c0 is a reference sound speed, and thetav is the rms fluctuation in an arrival angle. At 1000 km, the PE results were close to quadratic at small deltaz, with values of thetav in disagreement with those of the integral approximation by factors of order 2. Pulse spreads in the PE results were much smaller than predicted by the IA estimates. Results imply that acoustic tomography of internal waves at ranges up to 1000 km can use the IA estimate of travel-time variance with reasonable reliability. 相似文献

Synthesis, electrical and magnetic characterization of a superconducting FeSe0.85 compound is reported. An anomaly in the magnetization against temperature around 90 K is observed. The magnetic characterization of a commercial compound with nominal FeSe stoichiometry is also presented.
