Allostery, which is one of the most direct and efficient methods to fine-tune protein functions, has gained increasing recognition in drug discovery. However, there are several challenges associated with the identification of allosteric sites, which is the fundamental cornerstone of drug design. Previous studies on allosteric site predictions have focused on communication signals propagating from the allosteric sites to the orthosteric sites. However, recent biochemical studies have revealed that allosteric coupling is bidirectional and that orthosteric perturbations can modulate allosteric sites through reversed allosteric communication. Here, we proposed a new framework for the prediction of allosteric sites based on reversed allosteric communication using a combination of computational and experimental strategies (molecular dynamics simulations, Markov state models, and site-directed mutagenesis). The desirable performance of our approach was demonstrated by predicting the known allosteric site of the small molecule MDL-801 in nicotinamide dinucleotide (NAD+)-dependent protein lysine deacetylase sirtuin 6 (Sirt6). A potential novel cryptic allosteric site located around the L116, R119, and S120 residues within the dynamic ensemble of Sirt6 was identified. The allosteric effect of the predicted site was further quantified and validated using both computational and experimental approaches. This study proposed a state-of-the-art computational pipeline for detecting allosteric sites based on reversed allosteric communication. This method enabled the identification of a previously uncharacterized potential cryptic allosteric site on Sirt6, which provides a starting point for allosteric drug design that can aid the identification of candidate pockets in other therapeutic targets.Using reversed allosteric communication, we performed MD simulations, MSMs, and mutagenesis experiments, to discover allosteric sites. The oxidation-reduction reaction between U(VI) and Ti(III) in HCl solution was studied spectrophotometrically. The reaction is second-order at all concentrations of reactants, HCl, ferrous chloride and mannitol used in this work. In 5M HCl the rate constantk increases with increasing Ti(III) concentration, whereas it decreases with increasing U(VI) concentration, with increasing HCl concentration from 1.00M to 7.17M and increases thereafter from 7.17M to 11.79M. The addition of mannitol causes a consistent decrease in the rate of reaction, whereas ferrous chloride has no effect. The activation energy for this oxidation-reduction reaction was 47.90±0.11 kJ·mol–1. The values of H

The present paper demonstrates the review of some acid processes as well as development of some new solvent processes for reclamation of used lubricating oils. The conventional processes are found to be of low yield (? 50%), laborious, time consuming and environmentally hazardous, because of residual acidic sludge. Based on the findings, a new modified Aluminium sulphate‐sodium silicate‐acid‐base method employing a small quantity of acid and giving a high yield (? 60%) is proposed. Further, to avoid use of acid, new regeneration processes based on solvent extraction were investigated. They are termed CCl4‐alcohol method, Dodecane‐alcohol method and Toluene‐alcohol method. These processes are not only cost effective in terms of complete solvent recovery, but are rapid, less time consuming, more environmentally friendly and gave a high yield (70–75%). The virgin lubricants (Castrol GTX and Rimula‐C) as well as oils recovered by different methods were also characterized physicochemically to determine kinematic viscosity, density, refractive index, carbon distribution, wear scar diameter, % Conradson carbon residue, % ash, % chloride, pour point, etc. Results obtained show that many of the physico‐chemical properties of the recovered oils are in good agreement with those of virgin oils. The n.d.M analysis was also performed which shows that virgin oils have 73 ± 3% paraffinic carbon, 26 ± 3% naphthenic carbon and about 1% aromatic carbon. The recovered oils also showed nearly the same chemical composition. The UV‐Visible spectra of the recovered oils are all similar to those of virgin lubricants. The results suggest that the oils recovered by solvent treatments, particularly Dodecan‐alcohol and Toluene‐alcohol methods, may serve for lubrication purposes and can be rendered as excellent as virgin lubricants with the addition of certain additives. The proposed methods may be considered as alternative cost effective green techniques for acid reclamation processes and being the motivation of the present investigation. 相似文献

Gas‐solid reactions in chemical and metallurgical industries often involve solid pellets and a gaseous reactant. The progress of chemical reaction is measured by the movement of zones within the pellet and has been explained in terms of diffusion and chemical reaction processes. Earlier models identified a single reaction zone, in addition to product layer and unreacted core. In the present article, two reaction zones are envisaged as a more plausible explanation of the movement of the zones as the reaction proceeds. Earlier models for reversible reactions have assumed that conditions at the interface between the reaction zone and the unreacted core correspond to equilibrium at the prevailing temperature. The gaseous concentrations were assumed to permeate the core at the interfacial values so that no reaction occured in the core. More realistically, the present article envisages an additional zone within which the gaseous concentrations fall from the equilibrium values to zero. It is assumed that in the reaction zone proper, referred to as zone I, having thickness zI, the concentration profile is sigmoidal. This agrees with the earlier work of Khan and Bowen [1] and Prasannan and Doraswamy. [2] In zone I and the concentration of the reactant gas varies between [Ai] and [A*]. In the zone II, having thickness z2, concentration varies linearly between [A*] and zero. This model has been applied successfully to the data of the reduction of hematite [3] at different temperatures. The contribution of different forms of resistance, diffusion in product layer, chemical reaction and diffusion in the reactant core, is assessed as function of time (start to the end of reaction). The thickness of the zones remain almost constant as the reaction progresses. In particular, the influences of the product and core diffusion coefficients and chemical equilibrium constant on the extant reaction are evaluated. The dependence of concentration profile and zone thickness on equilibrium constant, K, velocity constant, k, diffusional coefficients DC and DP has been investigated thoroughly. Conventional cancer treatments have shown several unfavourable adverse effects, as well as an increase in anticancer drug resistance, which worsens the impending cancer therapy. Thus, the emphasis is currently en route for natural products. There is currently great interest in the natural bioactive components from medicinal plants possessing anticancer characteristics. For example, clove (Syzygium aromaticum L.) (Family Myrtaceae) is a highly prized spice that has been historically utilized as a food preservative and for diverse medical uses. It is reckoned amongst the valued sources of phenolics. It is indigenous to Indonesia but currently is cultivated in various places of the world. Among diverse active components, eugenol, the principal active component of S. aromaticum, has optimistic properties comprising antioxidant, anti-inflammatory, and anticancer actions. Eugenol (4-allyl-2-methoxyphenol) is a musky oil that is mainly obtained from clove. It has long been utilized all over the world as a result of its broad properties like antioxidant, anticancer, anti-inflammatory, and antimicrobial activities. Eugenol continues to pique investigators’ interest because of its multidirectional activities, which suggests it could be used in medications to treat different ailments. Anticancer effects of eugenol are accomplished by various mechanisms like inducing cell death, cell cycle arrest, inhibition of migration, metastasis, and angiogenesis on several cancer cell lines. Besides, eugenol might be utilized as an adjunct remedy for patients who are treated with conventional chemotherapy. This combination leads to a boosted effectiveness with decreased toxicity. This paper studies the new families of exact traveling wave solutions with the modified nonlinear Schrödinger equation, which models the propagation of rogue waves in ocean engineering. The extended Fan sub-equation method with five parameters is used to find exact traveling wave solutions. It has been observed that the equation exhibits a collection of traveling wave solutions for limiting values of parameters. This method is beneficial for solving nonlinear partial differential equations, because it is not only useful for finding the new exact traveling wave solutions, but also gives us the solutions obtained previously by the usage of other techniques (Riccati equation, or first-kind elliptic equation, or the generalized Riccati equation as mapping equation, or auxiliary ordinary differential equation method) in a combined approach. Nanohybrid superconducting junctions using antimony telluride (Sb2Te3) topological insulator nanoribbons and Nb superconducting electrodes are fabricated using electron beam lithography and magnetron sputtering. The effects of bias current, temperature, and magnetic field on the transport properties of the junctions in a four-terminal measurement configuration are investigated. Two features are observed. First, the formation of a Josephson weak-link junction. The junction is formed by proximity-induced areas in the nanoribbon right underneath the inner Nb electrodes which are connected by the few tens of nanometers short Sb2Te3 bridge. At 0.5 K a critical current of 0.15 µA is observed. The decrease of the supercurrent with temperature is explained in the framework of a diffusive junction. Furthermore, the Josephson supercurrent is found to decrease monotonously with the magnetic field indicating that the structure is in the small-junction limit. As a second feature, a transition is also observed in the differential resistance at larger bias currents and larger magnetic fields, which is attributed to the suppression of the proximity-induced superconductive state in the nanoribbon area underneath the Nb electrodes. 相似文献

Preclassification of raw infrared spectra has often been neglected in scientific literature. Separating spectra of low spectral quality, due to low signal-to-noise ratio, presence of artifacts, and low analyte presence, is crucial for accurate model development. Furthermore, it is very important for sparse data, where it becomes challenging to visually inspect spectra of different natures. Hence, a preclassification approach to separate infrared spectra for sparse data is needed. In this study, we propose a preclassification approach based on Multiplicative Signal Correction (MSC). The MSC approach was applied on human and the bovine knee cartilage broadband Fourier Transform Infrared (FTIR) spectra and on a sparse data subset comprising of only seven wavelengths. The goal of the preclassification was to separate spectra with analyte-rich signals (i.e., cartilage) from spectra with analyte-poor (and high-matrix) signals (i.e., water). The human datasets 1 and 2 contained 814 and 815 spectra, while the bovine dataset contained 396 spectra. A pure water spectrum was used as a reference spectrum in the MSC approach. A threshold for the root mean square error () was used to separate cartilage from water spectra for broadband and the sparse spectral data. Additionally, standard noise-to-ratio and principle component analysis were applied on broadband spectra. The fully automated MSC preclassification approach, using water as reference spectrum, performed as well as the manual visual inspection. Moreover, it enabled not only separation of cartilage from water spectra in broadband spectral datasets, but also in sparse datasets where manual visual inspection cannot be applied. 相似文献
