The chemo-enzymatic epoxidation of Lallemantia iberica seed oil (LISO), a novel plant oil characterized by its exceptional high content of alpha-linolenic acid (>?60%), was developed using an immobilized lipase from Pseudozyma antarctica and hydrogen peroxide as oxidant. A statistical approach was used to study the effect of enzyme amount, temperature, time, and solvent amount on the oxirane oxygen content obtained during epoxidation. An oxirane oxygen content of 8.6 ± 0.2% corresponding to a yield of 82% was obtained under optimized conditions that were identified to be at an enzyme load of 8.2 g/mol of double bonds, a solvent amount of 56.4 wt.%, a temperature of 33 °C, and an incubation time of 17 h. In addition, the experimental investigation was combined with a techno-economic and ecological assessment gaining detailed information regarding cost structure and environmental impact for the chemo-enzymatic epoxidation of the novel plant oil. 相似文献

In order to quantify autoantibodies in the sera of patients with autoimmune disease, we have created a microarray-based immunoassay that allows the simultaneous analysis of 18 known autoantigens. The microarrays contain serial dilutions of the various antigens, thereby allowing accurate determination of autoantibody titer using minimal amounts of serum. The assay is very sensitive and highly specific: as little as 40 fg of a known protein standard can be detected with little or no cross-reactivity to nonspecific proteins. The signal intensities observed from serial dilutions of immobilized antigen correlate well with serial dilutions of autoimmune sera. Miniaturized and highly parallelized immunoassays like these will reduce costs by decreasing reagent consumption and improve efficiency by greatly increasing the number of assays that can be performed with a single serum sample. This system will significantly facilitate and accelerate the diagnostics of autoimmune diseases and can be adapted easily to any other kind of immunoassay. 相似文献

The close relationship of the structure of ternary molybdenum chalcogenides to molecular cluster compounds suggests that these chalcogenides might be synthesized from such clusters or be solubilized as clusters. However, in contrast to molybdenum halide clusters which can be obtained in solution from the solid phase, many attempts to solubilize these chalcogenide clusters were unsuccessful. This is the first report that some of the pseudo-one-dimensional compounds (M2Mo6X6, X = Se, Te; M = Li, Na) can be dissolved when exposed to highly polar solvents such as dimethylsulfoxide or N-methylformamide. We show using optical microscopy, TEM, and light scattering that some of these solutions contain individual (Mo3X3)1∞ chains. This is also the first example of a purely inorganic transition-metal polymer solution. The behavior with respect to flocculation is consistent with the double layer theory. An efficient method for data processing and interpretation is needed to support and extend disulfide mass-mapping methodology based on partial reduction and cyanylation-induced cleavage to proteins containing more than four cystines. Here, the concept of "negative signature mass" is introduced as the novel feature of an algorithm designed to identify the disulfide structure of a cystinyl protein given an input of mass spectral data and an amino acid sequence. The "negative signature mass" process is different from the conventional approach in that it does not directly rule-in disulfide linkages, but rather eliminates linkages from a list of all possible theoretical linkages, with the goal of ruling out enough linkages so that only one disulfide structure can be constructed. The operating principles and the effectiveness of the algorithm are described in the context of analyzing ribonuclease A, a 124-residue protein containing eight cysteines in the form of four cystines (disulfides). 相似文献
