We present a disposable microarray hybridization chamber with an integrated micropump to speed up diffusion based reaction kinetics by generating convective flow. The time-to-result for the hybridization reaction was reduced from 60 min (standard protocol) down to 15 min for a commercially available microarray. The integrated displacement micropump is pneumatically actuated. It includes two active microvalves and is designed for low-cost, high volume manufacturing. The setup is made out of two microstructured polymer parts realized in polycarbonate (PC) separated by a 25 μm thermoplastic elastomer (TPE) membrane. Pump rate can be controlled between 0.3 μl s(-1) and 5.7 μl s(-1) at actuation frequencies between 0.2 Hz and 8.0 Hz, respectively. 相似文献

Functionalization of surfaces with highly branched dendrimer molecules has gained attractiveness for various applications because the number of functional groups exceeds those of surfaces functionalized with self-assembled monolayers. So far, little is known about the physicochemical properties of dendrimer functionalized surfaces, especially if the flexibility of dendrimer structure remains after covalent immobilization. Therefore, the purpose of this study was to covalently immobilize polyamidoamine (PAMAM) dendrimer molecules exhibiting terminal amine and carboxyl groups to silicon model surfaces and to explore their properties and structure at the solid-air and solid-liquid interface. Our results show that the surface free energy is higher for PAMAM coatings than for analogously terminated SAMs and also higher for carboxyl than amine functionalized coatings. Furthermore, several findings suggest that conformational freedom of the dendrimers was preserved after surface immobilization. Wet compared to dry PAMAMNH(2) surfaces show reduced hydrophilicity and increased contact angle hysteresis, whereas PAMAMCOOH surfaces become more hydrophilic and showed decreased hysteresis. Streaming current measurements showed an unexpected behavior for PAMAMCOOH surfaces in that they reveal a net positive surface charge over a wide pH range in spite of the carboxylated periphery. All of these results indicate a certain degree of masking, burrowing, back-folding and unfolding of functional groups upon environmental changes. 相似文献

An autofocusing method for two-wavelength digital holographic microscopy (TWDHM) based on the wavelength dependence of the diffraction process is proposed. Red and green lights are employed for the illumination of the TWDHM, and the generated holograms are recorded simultaneously by a color CCD camera. Due to the wavelength dependency of the diffraction process, the farther the reconstruction plane is from the image plane, the larger the difference is between the red and green light distributions. Thus, the image plane can be determined by finding the minimum of the variation between the red and green lights on their amplitude distributions. The feasibility of the proposed method is demonstrated by simulation and experiment. 相似文献

Femtosecond x-ray laser flashes with power densities of up to 10(14) W/cm(2) at 13.7 nm wavelength were scattered by single xenon clusters in the gas phase. Similar to light scattering from atmospheric microparticles, the x-ray diffraction patterns carry information about the optical constants of the objects. However, the high flux of the x-ray laser induces severe transient changes of the electronic configuration, resulting in a tenfold increase of absorption in the developing nanoplasma. The modification in opaqueness can be correlated to strong atomic charging of the particle leading to excitation of Xe(4+). It is shown that single-shot single-particle scattering on femtosecond time scales yields insight into ultrafast processes in highly excited systems where conventional spectroscopy techniques are inherently blind. 相似文献

An enantioselective assay for the determination of methadone and its main metabolite 2‐ethylidene‐1,5‐dimethyl‐3,3‐diphenylpyrrolidine in equine plasma based on capillary electrophoresis with highly sulfated γ‐cyclodextrin as chiral selector and electrokinetic analyte injection is described. The assay is based on liquid/liquid extraction of the analytes at alkaline pH from 0.1 mL plasma followed by electrokinetic sample injection of the analytes from the extract across a buffer plug without chiral selector. Separation occurs cationically at normal polarity in a pH 3 phosphate buffer containing 0.16% (w/v) of highly sulfated γ‐cyclodextrin. The developed assay is precise (intra‐ and interday RSD < 4% and < 7%, respectively), is capable to determine enantiomer levels of methadone and 2‐ethylidene‐1,5‐dimethyl‐3,3‐diphenylpyrrolidine in plasma down to 2.5 ng/mL, and was successfully applied to monitor enantiomer drug and metabolite levels in plasma of a pony that was anesthetized with racemic ketamine and isoflurane and received a bolus of racemic methadone and a bolus followed by constant rate infusion of racemic methadone. Recent experimental signals have led to a revival of tetraquarks,the hypothetical q~2q~2 hadronic states proposed by Jaffe in 1976 to explain the light scalar mesons.Mesonic structures with exotic quantum numbers have indeed been observed recently,though a controversy persists as to whether these are true resonances and not merely kinematical threshold enhancements,or otherwise states not of a true q~2q~2 nature.Moreover,puzzling non-exotic mesons are also often claimed to have a tetraquark configuration.However,the corresponding model calculations are practically always carried out in pure and static bound-state approaches,ignoring completely the coupling to asymptotic two-meson states and unitarity,especially the dynamical effects thereof.In this short paper we argue that these static predictions of real tetraquark masses are highly unreliable and provide little evidence of the very existence of such states. 相似文献
