We have characterized the solution-phase and dry storage stability of electrochemical E-DNA sensors fabricated using mixed self-assembled monolayers (SAMs) composed of 6- or 11-carbon (C6 and C11, respectively) alpha,omega-thiol alcohols and the analogous C6- or C11-thiol-terminated stem-loop DNA probe. We find that the solution-phase and dry storage stability of C6-based E-DNA sensors are limited and poorly reproducible. The use of stabilizing agents bovine serum albumin plus either glucose or trehalose significantly improves the dry storage shelf life of such sensors: when using these preservatives, we observe only 7-9% sensor degradation after 1 month of storage in air at room temperature. In comparison, the stability of C11-based E-DNA sensors is significantly greater than that of the C6-based sensors; we observe only minor (5-8%) loss of signal upon storing these sensors for a week under ambient solution conditions or for more than a month in air in the presence of preservatives. Moreover, whereas the electron-transfer rate through C11 SAMs is slower than that observed for C6 SAMs, it is rapid enough to support good sensor performance. It thus appears that C11 SAMs provide a reasonable compromise between electron-transfer efficiency and sensor stability and are well suited for use in electronic DNA-sensing applications.  相似文献

The AOAC Use-Dilution methods do not provide procedures to enumerate the test microbe on stainless steel carriers (penicylinders) or guidance on the expected target populations of the test microbe (i.e., a performance standard). This report describes the procedures used by the U.S. Environmental Protection Agency to enumerate the test microbe (carrier counts) associated with conducting the Use-Dilution method with Staphylococcus aureus (Method 955.15) and Pseudomonas aeruginosa (Method 964.02) and the examination of historical data. The carrier count procedure involves the random selection of carriers, shearing bacterial cells from the carrier surface through sonication, and plating of serially diluted inoculum on trypticase soy agar. For each Use-Dilution test conducted, the official AOAC method was strictly followed for carrier preparation, culture initiation, test culture preparation, and carrier inoculation steps. Carrier count data from 78 Use-Dilution tests conducted over a 6-year period were compiled and analyzed. A mean carrier count of 6.6 logs (approximately 4.0 x 10(6) colony-forming units/carrier) was calculated for both S. aureus and P. aeruginosa. Of the mean values, 95% fell within +/- 2 repeatability standard deviations. The enumeration procedure and target carrier counts are desirable for standardizing the Use-Dilution methods, increasing their reproducibility, and ensuring the quality of the data.  相似文献

The two‐ and three‐dimensional mercurous cations [(Hg2)3(OH)2]4+ and [(Hg2)2O]2+ crystallize with channels and cages of roughly 1 nm diameter from aqueous solutions dependent upon the acidity of the solution. Crystal structures were determined, for example, for [Zn(H2O)6][(Hg2)3(OH)2](NO3)6 (trigonal, space group P321, a = 1183.5(2) pm, c = 534.8(1) pm, Z = 1, R1 = 0.0351 for I0 > 2σ(I0)) and for [(Hg2)2O][Pb(NO3)3]2 (cubic, space group , a = 1543.1(2) pm, Z = 8, R1 = 0.0534 I0 > 2σ(I0)).  相似文献

Nine new double-salt compounds have been prepared and six crystal structures are reported that demonstrate a general metathesis route to double-salt compounds with metallophilic interactions. The compounds contain [Pt(terpy)X]+ or [Au(bpy)X2]+ cations, Au(III) or Au(I) anions such as [AuBr4]- or [AuCl2]-, and are prepared in water and recrystallized from organic solvents. In all crystallographically characterized cases, there exist metallophilic interactions between cations and anions, demonstrating the power of this general route. In most cases, there exists an unbroken chain of metallophilic interactions through the crystal, forming single-atom-wide wires.  相似文献

A novel solution to problematic ghost images is implemented by using tilted lens elements with polynomial surfaces. Tilting the lens surfaces sends reflections out of the imaging path. The nonrotationally symmetric polynomial surfaces correct aberrations caused by tilts. Selective fluorescence labelling of oxidized cellulose functionalities followed by GPC-MALLS was used to get a deeper insight into ink-induced degradation processes. As the method is very sensitive towards oxidation and molecular weight changes, slight variations at the very beginning of aging processes, e.g. during ink corrosion of cellulose, can be studied. Five different ink modifications were applied on model papers and underwent mild accelerated aging at 55 °C and cycling humidity (7 days) followed by a short period of static humid aging at 80 °C (2 days). Pure ink constituents like tannic acid or iron sulphate do not result in the same degree of oxidation or chain scission as complete inks. Balanced ink degrades paper more than single compounds, but less than unbalanced inks. Interestingly, some degradation occurs already during or shortly after the application process of unbalanced inks on paper. It could be demonstrated that this oxidation proceeded in a rather high Mw area, while the subsequent aging steps affected predominantly regions of shorter cellulose chains.  相似文献

A combined experimental and theoretical approach has been employed to establish the basicity and proton affinity of SiF4 and the structure of SiF4H+. The kinetics and energetics for the transfer of a proton between SiF4, N2, and Xe have been explored experimentally in helium at 0.35±0.02 torr and 297±3 K with a selected-ion flow tube apparatus. The results of equilibrium constant measurements are reported that provide a basicity and proton affinity for SiF4 at 297±3 K of 111.4±1.0 and 117.7±1.2 kcal mol?1, respectively. These values are more than 2.5 kcal mol?1 lower than currently recommended values. The basicity order was determined to be GB(Xe)>GB(SiF4)>GB(N2), while the proton-affinity order was shown to be PA(Xe)>PA(N2)>PA (SiF4). Ab initio molecular orbital computations at MP4SDTQ(fc)/6-311++G(3df,3pd) using geometries from B3LYP/6-31+G(d,p) indicate a value for PA(SiF4)=118.7 kcal mol?1 that is in good agreement with experiment. Also, the most stable structure of SiF4H+ is shown to correspond to a core SiF 3 + cation solvated by HF with a binding energy of 43. 9 kcal mol?1. Support for this structure is found in separate SIFT collision induced dissociation (CID) measurements that indicate exclusive loss of HF.  相似文献

We develop a new scheme for determining molecular partial atomic charges (PACs) with external electrostatic potential (ESP) closely mimicking that of the molecule. The PACs are the ‘minimal corrections’ to a reference set of PACs necessary for reproducing exactly the tensor components of the Cartesian zero-, first- and second- molecular electrostatic multipoles. We evaluate the quality of ESP reproduction when ‘minimally correcting’ (MC) Mulliken, Hirshfeld or iterative-Hirshfeld reference PACs. In all these cases, the MC-PACs significantly improve the ESP while preserving the reference PACs’ invariance under the molecular symmetry operations. When iterative-Hirshfeld PACs are used as reference, the MC-PACs yield ESPs of comparable quality to those of the ChElPG charge fitting method.  相似文献

Supraglottic activity was rated from flexible endoscopic video recordings of subjects with normal laryngeal structure and function as they sustained vowels and repeated syllables and sentences. Judges rated these recordings for false vocal fold (FVF) adduction and anterior-to-posterior (A-P) compression at the initiation of the speech task, throughout the whole speech task (static supraglottic activity), and as brief individual adductions within a speech task (dynamic supraglottic activity). Significant differences in A-P (p < 0.0003) and FVF (p < 0.0000001) compression were found between tasks. Dynamic FVF activity was associated with glottal stops. Static A-P and FVF activities were present in males significantly more (p < 0.0001) than females. FVF activity associated with speech initiation was found in females significantly more (p = 0.0256) than males. Supraglottic activity plays a role in normal speech production, and should not necessarily be considered suggestive of a voice use pattern with excessive muscle tension.  相似文献
