Several disubstituted pyrazolo[3,4-d]pyrimidine, pyrazolo[1,5-a]pyrimidine and thiazolo[4,5-d]pyrimidine ribonucleosides have been prepared as congeners of uridine and cytidine. Glycosylation of the trimethylsilyl (TMS) derivative of pyrazolo[3,4-d]pyrimidine-4,6(1H,5H,7H)-dione ( 4 ) with 1-O-acetyl-2,3,5-tri-O-benzoyl-D-ribofuranose ( 5 ) in the presence of TMS triflate afforded 7-(2,3,5-tri-O-benzoyl-β-D-ribofuranosyl)pyrazolo-[3,4-d]pyrimidine-4,6(1H,5H)-dione ( 6 ). Debenzoylation of 6 gave the uridine analog 7-β-D-ribofuranosylpyrazolo[3,4-d]pyrimidine-4,6(1H,5H)-dione ( 3 ), identical with 7-ribofuranosyloxoallopurinol reported earlier. Thiation of 6 gave 7 , which on debenzoylation afforded 7-β-D-ribofuranosyl-6-oxopyrazolo[3,4-d]pyrimidine-4(1H,5H)-thione ( 8 ). Ammonolysis of 7 at elevated temperature gave a low yield of the cytidine analog 4-amino-7-β-D-ribofuranosylpyrazolo[3,4-d]pyrimidin-6(1H)-one ( 11 ). Chlorination of 6 , followed by ammonolysis, furnished an alternate route to 11 . A similar glycosylation of TMS-4 with 2,3,5-tri-O-benzyl-α-D-arabinofuranosyl chloride ( 12 ) gave mainly the N7-glycosylated product 13 , which on debenzylation provided 7-β-D-arabinofuranosylpyrazolo[3,4-d]pyrimidine-4,6(1H,5H)-dione ( 14 ). 4-Amino-7-β-D-arabinofuranosyl-pyrazolo[3,4-d]pyrimidin-6(1H)-one ( 19 ) was prepared from 13 via the C4-pyridinium chloride intermediate 17 . Condensation of the TMS derivatives of 7-hydroxy- ( 20 ) or 7-aminopyrazolo[1,5-a]pyrimidin-5(4H)-one ( 23 ) with 5 in the presence of TMS triflate gave the corresponding blocked nucleosides 21 and 24 , respectively, which on deprotection afforded 7-hydroxy- 22 and 7-amino-4-β-D-ribofuranosylpyrazolo[1,5-a]pyrimidin-5-one ( 25 ), respectively. Similarly, starting either from 2-chloro ( 26 ) or 2-aminothiazolo[4,5-d]pyrimidine-5,7-(4H,6H)-dione ( 29 ), 2-amino-4-β-D-ribofuranosylthiazolo[4,5-d]pyrimidine-5,7(6H)-dione ( 28 ) has been prepared. The structure of 25 was confirmed by single crystal X-ray diffraction studies.  相似文献

Ligands derived from the tripodal N4 ligand tris(pyridylmethyl)amine ((pyCH2)3N, tpa) of general formula (6-RNHpyCH2)nN(CH2py)(3-n)(R = H, n= 1-3 L(1-3); R = neopentyl, n= 1-3 L'(1-3)) were used to elucidate and quantify the magnitude of the effects exerted by hydrogen bonding and hydrophobic environments in the zinc-water acidity of their complexes. The pKa of the zinc-bound water molecule of [(L(1-3))Zn(OH2)]2+ and [(L'(1-3))Zn(OH2)]2+ 1'-3' was determined by potentiometric pH titrations in water (1-3) or water-ethanol (1:1) (1'-3'). The zinc(II) water acidity gradually increases as the number of -NH2 hydrogen bonding groups adjacent to the water molecule increases. Thus, the zinc-bound water of [(L3)Zn(OH2)]2+ and [(tpa)Zn(OH2)]2+ deprotonate with pKa values of 6.0 and 8.0, respectively. The pKa of the water molecule, however, is only raised from 8.0 in [(tpa)Zn(OH2)]2+ to 9.1 in [(bpg)Zn(OH2)]+ (bpa =(pyCH2)2N(CH2COO-)). Moreover, the acidity of the zinc-bound water of several of the five-coordinate zinc(II) complexes with the hydrogen bonding groups is greater than that of four-coordinate [((12)aneN3)Zn(OH2)]2+ (pKa = 7.0). This result shows that the magnitude of the effect exerted by the hydrogen bonding groups can be larger than that induced by changing one neutral by one anionic ligand, and/or even by changing the coordination number of the zinc(II) centre. The X-ray structure of [(L'2)Zn(OH)]ClO4 2' and [(L'3)Zn(OH)]ClO4.CH3CN 3'.CH3CN is reported, and show the neopentylamino groups forming N-H...O hydrogen bonds with the zinc-bound hydroxide. Although, which have hydrogen bonding and hydrophobic groups, have a zinc-bound water more acidic than [(tpa)Zn(OH2)]2+, their pKa is not always lower than that of 1-3. This result suggests that a hydrogen bonding microenvironment may be more effective than a hydrophobic one to increase the zinc-water acidity.  相似文献

Abstract— In this study we compared the plasma distribution and arterial accumulation of a photosensitizer, benzoporphyrin derivative (BPD), in two models of atherosclerosis: the spontaneous lesions of the Watanabe heritable hyperlipidemic (WHHL) rabbit and induced lesions of the balloon-injured, cholesterol-fed New Zealand white (NZW) rabbit. Selective uptake and retention of a photosensitizer by the abnormal portion of a vessel is a necessity in order for photodynamic therapy to become a successful modality for inhibition of intimal hyperplasia, selective removal of atherosclerotic tissue or imaging of diseased arteries. Liposome-based formulations were compared to freshly isolated native low density lipoprotein (LDL) and acetylated-LDL (Ac-LDL) as delivery vehicles for BPD. Plasma distribution of the photosensitizer was analyzed by KBr density gradient ultracentrifuga-tion. Although the delivery vehicle influenced plasma distribution immediately postinjection, BPD subsequently partitioned according to the plasma concentration of the lipoproteins. Photosensitizer level in plaque and normal artery specimens was determined by ethyl acetate extraction and spectrofluorometric measurement. The measurement of BPD in normal and atherosclerotic arterial tissue demonstrated a selective accumulation in atherosclerotic tissue. Preassociation with LDL and Ac-LDL enhanced accumulation of BPD in atherosclerotic tissue when compared with normal artery (mean ratios of 2.8 and 4.1 were achieved, respectively). These results indicate that the preferential uptake of BPD by atherosclerotic plaque can be enhanced by preassociation with plasma lipoproteins, suggesting that light activation could lead to a highly selective destruction of diseased vascular tissue.  相似文献

A method based on the complexometric titration of thorium using ethylene diaminetetra-acetic acid (EDTA) as complexant has been developed for the determination of thorium in thorium-plutonium solution without resorting to prior separation of plutonium. Plutonium in the form of Pu(VI) does not affect the thorium determination when present up to 10% in thorium—plutonium solution. For oxidation of plutonium to Pu(VI), HClO4 or AgO was used. HClO4 is preferred. The thorium values obtained without prior separation of plutonium are compared with those obtained after separating plutonium by anion exchange technique. A precision of ±0.5% has been obtained for 5–10 mg of thorium in the aliquot.  相似文献

Oximidobenzotetronic acid is recommended for the separation and gravimetric determination of palladium and cobalt An ethanolic solution of the reagent quantitatively precipitates palladium(II) from solutions which are 0.75 N in acid up to pH 5.1, the complex is weighed as Pd(C9H5NO4)2. Cobalt(II) can be determined in the filtrate after the precipitation of palladium. With 0.5 N acid solutions, no interference was found from Pt(IV), Ir(IV), Rh(III), Ru(III), Os(IV), Au(III), Ag(I), Cu(II), Fe(III), Ni(II), Hg(II). Pb(II), Bi(III), Cd(II), As(V), Se(VI), Te(IV), Mo(VI), Sb(III), Al(III), Cr(III), Zn(II), Ti(IV), Zr(IV). acetate, oxalate, citrate, tartrate, phosphate and fluoride.  相似文献

The effect of 4th generation poly(amidoamine) dendrimer (4G PAMAM) present in an anionic phospholipid composition, consisting of hydrogenated soyphosphatidylcholine (HSPC), cholesterol (CH), dicetyl phosphate (DCP), and poly(ethylene glycol) (Mw approximately 2000) derivatized phosphatidylethanolamine (PEG2000-PE), on the hydration and liquid crystalline structure formation was investigated. The optical and polarized light microscopies of the liposomal dispersion obtained from the hydrated lipid composition show two types of birefringent structures (mesophases): plastic, wormlike microstructures and conventional, over-elongated lamellae. Differential scanning calorimetry (DSC) shows an increase in the liquid crystalline phase transition (Tg) of the lipid composition from 60 to 94 degrees C with increasing 4G PAMAM concentrations from 0 to 0.011 mM, respectively. The Tg values of the two microstructures were 68 and 84 degrees C, respectively, indicating that the plastic microstructures were 4G PAMAM/DCP-complexes-rich (alpha mesophases) and the conventional and elongated lamellae were dendrimer-doped HSPC/CH-rich microstructures (beta mesophases). Optical microscopy shows that the alpha mesophases convert into various other types of vesicular structures such as giant unilamellar vesicles and biliquid foams, upon heating above the phase transition temperature of the lipid composition (approximately 60-65 degrees C). The microstructure transformation is a result of an osmotic influx of water and the detergent action of PEG2000-PE present in the lipid composition. The transmission electron microscopy (TEM) images of the liposomal dispersion show particles embedding circular transparent domains that exactly correlate to the theoretical 4G PAMAM/DCP complex sizes, thus, providing evidence of 4G PAMAM interspersed within the two mesophases. Small-angle X-ray scattering (SAXS) measurements indicate that the alpha mesophases are a dendrimer-interlinked, symmetrically undulated lamellar phase and the beta mesophases are dendrimer-doped, occasionally kinked lamellae. An increase in dendrimer concentration in the lipid composition was found to decrease interlamellar spacing. On the basis of optical microscopy, DSC, TEM, and SAXS data, a model of dendrimer-doped mesophase structure and lamellae fusion is proposed. This investigation provides new self-assembled materials for drug/gene delivery and supplements the understanding of mechanisms involved in various biological processes such as membrane fusion, transmembrane permeation, and endocytosis.  相似文献

Three-component Grieco condensation reaction of 3-aminopyrazolo[3,4-b]pyridine, formaldehyde/benzaldehyde and electron rich alkenes in presence of iron(III) chloride gave tetrahydropyrido[2′,3′:3,4]pyrazolo[1,5-a]pyrimidines/quinazolines in single pot. The sequence of reactions is formation of di-azadiene in situ and the subsequent regioselective addition of alkenes in aza-Diels Alder type reaction. The structure and stereochemistry of the products were confirmed by spectral data and single crystal X-ray crystallography.  相似文献

The kinetics of decomposition of hydrogen peroxide have been studied on mixed Fe(III)–Al(III) hydroxide and oxide catalysts. While iron hydroxide possesses considerable catalytic activity, aluminium hydroxide has very little activity. The rate of decomposition on mixed hydroxides increases with increasing concentration of aluminium hydroxide up to about 1.5 mol% and decreases thereafter. The mixed oxides possess negligible activity compared to the corresponding hydroxides. The energy of activation, as calculated from the Arrhenius equation, is 10.1 kcal/mol for sample S4, containing 1.52 mol% of alumina. The rate of decomposition of S4 increases with increasing pH up to 6.8 and decreases thereafter. The rate is first order in all these cases. A suitable mechanism is suggested.

Nuclear prenylation of naringenin (7) with 2-methylbut-3-en-2-ol in the presence of boron trifluoride etherate gives a mixture of 6-C-prenyl-(11), 8-C-prenyl-(15) and 6,8-di-C-prenyl-(8) derivatives. On formic acid cyclisation, 11 yielded two monodihydropyrans (12 and 13), but 15 afforded only one viz 16; similarly 8 formed the bisdihydropyran 10. Methylation of 8-C-prenyl naringenin (15) with Me2SO4 resulted in the formation of di-O-methyl derivatives of xanthohumol (22) and isoxanthohumol (23).

Cyclodehydrogenation of 6,8-di-C-prenyl-naringenin (8) with DDQ gave a mono-C-prenyl-2,2-dimethylpyran (1) corresponding to (±) lupinifolin. The angular isomer (2) was also formed. The structure of natural flemichin-B therefore needs further consideration. Similarly, cyclodehydrogenation of 6-C-(11)- and 8-C-prenyl-(15) naringenins afforded the corresponding linear (24) and angular (25) derivatives which have been characterized by conversion into known chalcones 26 and 27 by O-methylation.  相似文献
