Alteration of insect growth regulators by the action of inhibitors is becoming an attractive strategy to combat disease-transmitting insects. In the present study, we investigated the larvicidal effect of 1,2,3-triazolyl-pyrimidinone derivatives against the larvae of the mosquito Anopheles arabiensis, a vector of malaria. All compounds demonstrated insecticidal activity against mosquito larvae in a dose-dependent fashion. A preliminary study of the structure–activity relationship indicated that the electron-withdrawing substituent in the para position of the 4-phenyl-pyrimidinone moiety enhanced the molecules’ potency. A docking study of these derivatives revealed favorable binding affinity for the sterol carrier protein-2 receptor, a protein present in the intestine of the mosquito larvae. Being effective insecticides against the malaria-transmitting Anopheles arabiensis, 1,2,3-triazole-based pyrimidinones represent a starting point to develop novel inhibitors of insect growth regulators. 相似文献

Constitutive heterochromatin comprises a substantial fraction of the eukaryotic genomes and is mainly composed of tandemly arrayed satellite DNAs (satDNA). These repetitive sequences represent a very dynamic and fast evolving component of genomes. In the present work we report the isolation of Cricetus cricetus (CCR, Cricetidae, Rodentia) centromeric repetitive sequences from chromosome 4 (CCR4/10sat), using the laser microdissection and laser pressure catapulting procedure, followed by DOP-PCR amplification and labelling. Physical mapping by fluorescent in situ hybridisation of these sequences onto C. cricetus and another member of Cricetidae, Peromyscus eremicus, displayed quite interesting patterns. Namely, the centromeric sequences showed to be present in another C. cricetus chromosome (CCR10) besides CCR4. Moreover, these almost chromosome-specific sequences revealed to be present in the P. eremicus genome, and most interestingly, displaying a ubiquitous scattered distribution throughout this karyotype. Finally and in both species, a co-localisation of CCR4/10sat with constitutive heterochromatin was found, either by classical C-banding or C-banding sequential to in situ endonuclease restriction.The presence of these orthologous sequences in both genomes is suggestive of a phylogenetic proximity. Furthermore, the existence of common repetitive DNA sequences with a different chromosomal location foresees the occurrence of an extensive process of karyotype restructuring somehow related with intragenomic movements of these repetitive sequences during the evolutionary process of C. cricetus and P. eremicus species. 相似文献

Magnetometry results have shown that gold NPs (∼2 nm in size) protected with phosphine and chlorine ligands exhibit permanent
magnetism. When the NPs size decreases down to the subnanometric size range, e.g. undecagold atom clusters, the permanent
magnetism disappears. The near edge structure of the X-ray absorption spectroscopy data points out that charge transfer between
gold and the capping system occurs in both cases. These results strongly suggest that nearly metallic Au bonds are also required
for the induction of a magnetic response. Electron paramagnetic resonance observations indicate that the contribution to magnetism

Human immunodeficiency virus type-1 integrase (HIV-1 IN) is an essential enzyme for effective viral replication. Diketo acids such as L-731,988 and S-1360 are potent and selective inhibitors of HIV-1 IN. In this study, we used molecular dynamics simulations, within the hybrid quantum mechanics/molecular mechanics (QM/MM) approach, to determine the protein-ligand interaction energy between HIV-1 IN and L-731,988 and 10 of its derivatives and analogues. This hybrid methodology has the advantage that it includes quantum effects such as ligand polarisation upon binding, which can be very important when highly polarisable groups are embedded in anisotropic environments, as for example in metal-containing active sites. Furthermore, an energy decomposition analysis was performed to determine the contributions of individual residues to the enzyme-inhibitor interactions on averaged structures obtained from rather extensive conformational sampling. Analysis of the results reveals first that there is a correlation between protein-ligand interaction energy and experimental strand transfer into human chromosomes and secondly that the Asn-155, Lys-156 and Lys-159 residues and the Mg(2+) ion are crucial to anti-HIV IN activity. AbstractTwo new oxamate-containing manganese(II) complexes, [{Mn(H2edpba)(H2O)2}2]n (1) and [Mn(H2edpba)(dmso)2]?dmso?CH3COCH3?H2O (2) (H4edpba = N,N′-ethylenediphenylenebis(oxamic acid) and dmso = dimethylsulfoxide), have been synthesized and the structures of 1 and 2 were characterized by single crystal X-ray diffraction. The structure of 1 consists of neutral honeycomb networks in which each manganese(II) is six-coordinate by one H2edpba2? ligand and two carboxylate–oxygens from two other H2edpba2? ligands building the equatorial plane. Each manganese is connected to its nearest neighbor through two carboxylate(monoprotonated oxamate) bridges in an anti-syn conformation. A dmso solution of single crystals of 1 was placed under acetone atmosphere affording 2, whereas putting 2 in equimolar water:ethanol mixture results in 1. The molecular structure of 2 is made up of mononuclear manganese(II) units which are interlinked by weak C–H?π and edge-to-face π-stacking interactions leading to supramolecular chains along the crystallographic b axis. Magnetic measurements reveal the occurrence of an antiferromagnetic coupling between two manganese(II) ions through anti-syn carboxylate bridges for 1 [J = ?1.18 cm?1, the Hamiltonian being defined as H = ?J S1.S2] and very weak intrachain ferromagnetic interactions in 2 [J = + 0.046 cm?1, H = ?J ∑iSi.Si + 1]. 相似文献

Despite the central importance of aqueous amphiphile assemblies in science and industry, the size and shape of these nano‐objects is often difficult to control with accuracy owing to the non‐directional nature of the hydrophobic interactions that sustain them. Here, using a bioinspired strategy that consists of programming an amphiphile with shielded directional Watson–Crick hydrogen‐bonding functions, its self‐assembly in water was guided toward a novel family of chiral micelle nanotubes with partially filled lipophilic pores of about 2 nm in diameter. Moderate levels of a proinflammatory macrophages phenotype are indispensable and play an important role in the skeletal muscle repair process since this response depends on their secreted products concentration to influence and modulate muscle inflammation as well as the differentiation of myoblasts. This study investigated the effects of photobiomodulation (PBM) on undifferentiated and differentiation-induced C2C12 myoblasts cultivated in different concentrations of M1 phenotype macrophage-conditioned media of J774 cells (MCM1) also submitted to PBM using the same irradiation parameters. Irradiation was performed once with low-level laser (780 nm, 70 mW, 1 J) and was evaluated cell viability, proliferation and differentiation, nitric oxide (NO) synthesis and IL-6 and TNF-α protein levels 24 and 48 h after C2C12 irradiation. PBM treatment in undifferentiated myoblasts exhibited lower IL-6 levels in the presence of nonirradiated MCM1 at both concentrations. Myoblasts in proliferation condition cultivated with irradiated MCM1 showed lower IL-6 and TNF-α levels after 48 h in the presence of both concentrations evaluated. PBM induced a decrease in the synthesis of NO on undifferentiated and differentiation-induced myoblasts. PBM was able to reduce the level of proinflammatory protein and markers, which are important to allow the differentiation of myoblasts during the muscle repair process. 相似文献

The orientation of the orthopalladation of iminophosphoranes R3P=NCH2Aryl (R=Ph, Aryl=Ph (1a), C6H(4)-2-Br (1b), C6H4-Me-2 (1e), C6H3-(Me)(2)-2,5 (1f); R=p-tolyl, Aryl=Ph (1c); R=m-tolyl, Aryl=Ph (1d); R3P=MePh2P, and Aryl=Ph (1g)) has been studied. 1a reacts with Pd(OAc)2 (OAc=acetate) giving endo-[Pd(micro-Cl){C,N-C6H4(PPh2=NCH2Ph)-2}]2 (3a), while exo-[Pd(micro-Br){C,N-C6H4(CH2N=PPh3)-2}]2 (3b) could only be obtained by the oxidative addition of 1b to Pd2(dba)3. The endo form of the metalated ligand is favored kinetically and thermodynamically, as shown by the conversion of exo-[Pd(micro-OAc){C,N-C6H4(CH2N=PPh3)-2}]2 (2b) into endo-[Pd(micro-OAc){C,N-C6H4(PPh2=NCH2Ph)-2}]2 (2a) in refluxing toluene. The orientation of the reaction is not affected by the introduction of electron-releasing substituents at the Ph rings of the PR3 (1c and 1d) or the benzyl units (1e and 1f), and endo complexes (3c-3f) were obtained in all cases. The palladation of MePh2P=NCH2Ph (1g) can be regioselectively oriented as a function of the solvent. The exo isomer [Pd(micro-Cl){C6H4(CH2N=PPh2Me)-2}]2 (exo-3g) is obtained in refluxing CH2Cl2, while endo-[Pd(micro-Cl){C,N-C6H4(PPh(Me)=NCH2Ph)-2}]2 (endo-3g) can be isolated as a single isomer in refluxing toluene. In this case, the exo metalation is kinetically favored while an endo process occurs under thermodynamic control, as shown through the rearrangement of [Pd(micro-OAc){C6H4(CH2N=PPh2Me)-2}]2 (exo-2g) into [Pd(micro-OAc){C,N-C6H4(P(Ph)Me=NCH2Ph)-2}]2 (endo-2g) in refluxing toluene. The preference for the endo palladation of 1a and the kinetic versus thermodynamic control in 1g has been explained through DFT studies of the reaction mechanism. 相似文献
