Galectin-1 (GAL1) is a beta-galactoside-binding protein that has been implicated in the regulation of viability of lymphoid cells. However, the signaling pathway governed by the binding of GAL1 to the cell membrane is not understood yet. As a first step toward the elucidation of GAL1-initiated signaling events, electrophoresis techniques such as sodium dodecyl sulfate-polyacrylamide gel electrophoresis (SDS-PAGE), and two-dimensional electrophoresis (2-DE) were used together with precipitation techniques. This allowed us to identify the membrane receptor of GAL1, and to characterize the signal resulting from the binding of GAL1 to this receptor. Our results demonstrate that the tyrosine phosphatase CD45 is the receptor for GAL1, and that the src-type tyrosine kinase Lyn is a target for the effects of GAL1/CD45 interactions in B-cells. Furthermore, these results show the usefulness of combined precipitation and electrophoresis techniques to analyze phosphotyrosine-dependent mechanisms during the study of cell functions.  相似文献

We prove the Bianchi permutability (existence of superposition principle) of Bäcklund transformations for asymmetric quad-equations. Such equations and their Bäcklund transformations form 3D consistent systems of a priori different equations. We perform this proof by using 4D consistent systems of quad-equations, the structural insights through biquadratics patterns and the consideration of super-consistent eight-tuples of quad-equations on decorated cubes.  相似文献

The problem of sequence comparison via optimal alignments occurs naturally in many areas of applications. The simplest such technique is based on evaluating a score given by the length of a longest common subsequence divided by the average length of the original sequences. In this paper we investigate the expected value of this score when the input sequences are random and their length tends to infinity. The corresponding limit exists but is not known precisely. We derive a theoretical large deviation, convex analysis and Monte Carlo based method to compute a consistent sequence of upper bounds on the unknown limit. We applied the solvation models SM8, SM8AD, and SMD in combination with the Minnesota M06-2X density functional to predict vacuum-water transfer free energies (Task 1) and tautomeric ratios in aqueous solution (Task 2) for the SAMPL2 test set. The bulk-electrostatic contribution to the free energy of solvation is treated as follows: SM8 employs the generalized Born model with the Coulomb field approximation, SM8AD employs the generalized Born approximation with asymmetric descreening, and SMD solves the nonhomogeneous Poisson equation. The non-bulk-electrostatic contribution arising from short-range interactions between the solute and solvent molecules in the first solvation shell is treated as a sum of terms that are products of geometry-dependent atomic surface tensions and solvent-accessible surface areas of the individual atoms of the solute. On average, three models tested in the present work perform similarly. In particular, we achieved mean unsigned errors of 1.3 (SM8), 2.0 (SM8AD), and 2.6 kcal/mol (SMD) for the aqueous free energies of 30 out of 31 compounds with known reference data involved in Task 1 and mean unsigned errors of 2.7 (SM8), 1.8 (SM8AD), and 2.4 kcal/mol (SMD) in the free energy differences (tautomeric ratios) for 21 tautomeric pairs in aqueous solution involved in Task 2.  相似文献

Weinberg's seminal prediction of the cosmological constant relied on a provisional method for regulating eternal inflation which has since been put aside. We show that a modern regulator, the causal patch, improves agreement with observation, removes many limiting assumptions, and yields additional powerful results. Without assuming necessary conditions for observers such as galaxies or entropy production, the causal patch measure predicts the coincidence of vacuum energy and present matter density. Their common scale, and thus the enormous size of the visible Universe, originates in the number of metastable vacua in the landscape.  相似文献
