Whole treechips obtained from softwood forest thinnings were pretreated via single-and two-stage dilute-sulfuric acid pretreatment.
Whole-tree chips were impregnated with dilute sulfuric acid and steam treated in a 4-L steam explosion reactor. In single-stage
pretreatment, wood chips were treated using a wide range of severity. In two-stage pretreatment, the first stage was carried
out at low severity tomaximize hemicellulose recovery. Solubilized sugars were recovered from the first-stage prehydrolysate
by washing with water. In the second stage, water-insoluble solids from first-stage prehydrolysate were impregnated with dilute
sulfuric acid, then steam treated at more severe conditions to hydrolyze a portion of the remaining cellulose to glucose and
to improve the enzyme digestibility. The total sugar yields obtained after enzymatic hydrolysis of two-stage dilute acid-pretreated
samples were compared with sugar yields from single-stage pretreatment. The overall sugar yield from two-stage dilute-acid
pretreatment was approx 10% higher, and the net enzyme requirement was reduced by about 50%. Simultaneous saccharification
and fermentation using an adapted Saccharomyces cerevisiae yeast strain further improved cellulose conversion yield and lowered the enzyme requirement. 相似文献

The process of converting renewable lignocellulosic biomass to ethanol requires a number of steps, and pretreatment is one
of the most important. Pretreatment usually in volves a hydrolysis of the easily hydrolyzed hemicellulosic component of biomass
using some form of thermal/chemical/mechanical action that results in a product that can be further hydrolyzed by cellulase
enzymes (the cellulosic portion). The sugars produced can then befermented to ethanol by fermentative microorganisms. If the
pretreatment step is not severe enough, the resultant residue is not as easily hydrolyzed by the cellulase enzyme. More severe
pretreatment conditions result in the production of degradation products that are toxic to the fermentative microorgan ism.
In this article, wereport the quantitative analysis of glucose, mannose, xylose, and acetic acid using Fourier transform infrared
(FTIR) spectroscopy on liquors from dilute-acid-pretreated softwood and hard wood slurries. Comparison of FTIR and high-performance
liquid chromatography quantitative analyses of these liquorsare reported. Recent developments in infrared probe technology
has enabled the rapid quantification of these sugars by FTIR spectroscopy in the batch reactor during optimization of the
pretreatment conditions, or interfaced to the computer controlling a continuous reactor for on-line monitoring and control. 相似文献

A series of sulfoalkyl ether -cyclodextrin derivatives, including sulfoethyl, sulfopropyl and sulfobutyl ethyl -cyclodextrins, have been synthesized and characterized. Each sulfoalkyl ether -cyclodextrin is a mixture of various degrees of substitution and different positional isotherms. Elemental analysis, 1H NMR, MS, and Differential Scanning Calorimetry analysis were used to determine the average degree of substitution for each -cyclodextrin derivative. The average degrees of substitution are 3.4, 1.6 and 2.5 for sulfoethyl, sulfopropyl and sulfobutyl ether -cyclodextrin, respectively. The water solubility of these derivatives is substantially higher than that of -cyclodextrin. 1H NMR indicates thatsulfoethyl ether -cyclodextrin may have major substitution on the secondaryhydroxyl group while the major substitution in sulfopropyl and sulfobutyl ether-cyclodextrin could be on the primary hydroxyl group. Approximately 9% of the 9.7 billion bushels of corn harvested in the United States was used for fuel ethanol production in 2002, half of which was prepared for fermentation by dry grinding. The University of Illinois has developed a modified dry grind process that allows recovery of the fiber fractions prior to fermentation. We report here on conversion of this fiber (Quick Fiber [QF]) to ethanol. QF was analyzed and found to contain 32%wt glucans and 65%wt total carbohydrates. QF was pretreated with dilute acid and converted into ethanol using either ethanologenic Escherichia coli strain FBR5 or Saccharomyces cerevisiae. For the bacterial fermentation the liquid fraction was fermented, and for the yeast fermentation both liquid and solids were fermented. For the bacterial fermentation, the final ethanol concentration was 30 g/L, a yield of 0.44 g ethanol/g of sugar(s) initially present in the hydrolysate, which is 85% of the theoretical yield. The ethanol yield with yeast was 0.096 gal/bu of processed corn assuming a QF yield of 3.04 lb/bu. The residuals from the fermentations were also evaluated as a source of corn fiber oil, which has value as a nutraceutical. Single-stage cocurrent dilute acid pretreatments were carried out on yellow poplar (Liriodendron tulipifera) sawdust using an as-installed and short residence time modified pilot-scale Sunds hydrolyzer and a 4-L bench-scale NREL
digester (steam explosion reactor). Pretreatment conditions for the Sunds hydrolyzer, installed in the NREL process development
unit (PDU), which operates at 1 t/d (bone-dry t) feed rate, spanned the temperature range of 160 – 210°C, 0.1 – 1.0% (w/w)
sulfuric acid, and 4-10-min residence times. The batch pretreatments of yellow poplar sawdust in the bench-scale digester
were carried out at 210 and 230°C, 0.26% (w/w) sulfuric acid, and 1-, 3-, and 4-min residence times. The dilute acid prehydrolysis
solubilized more than 90% of the hemicellulose, and increased the enzymatic digestibility of the cellulose that remained in
the solids. Compositional analysis of the pretreated solids and liquors and mass balance data show that the two pretreatment

Selective thinning of forests in the western United States will generate a large, sustainable quantity of softwood residues
that can be an attractive feedstock for fuel ethanol production. The major species available from thinning of forests in northern
California and the eastern Rocky Mountains include white fir (Abies concolor), Douglas fir (Pseudotsuga menziesii), and Ponderosa pine (Pinus ponderosa). Douglas fir chips were soaked in 0.4% sulfuric acid solution, then pretreated with steam at 200 – 230°C for 1 – 5 min.
After pretreatment, 90 – 95% of the hemicellulose and as much as 20% of the cellulose was solubilized in water, and 90% of
the remaining cellulose can be hydrolyzed to glucose by cellulase enzyme. The prehydrolysates, at as high as 10% total solid

The impact of the nonionic surfactant, dodecyl triethyleneglycol ether (C(12)E(3)) on the solution microstructure of the dialkyl chain cationic surfactant, dihexadecyl dimethyl ammonium bromide, (DHDAB) has been investigated. The variation in solution microstructure has been studied using a combination of small angle neutron scattering, ultra small angle neutron scattering, optical texture and photon correlation spectroscopy. At low surfactant concentrations (1.5 mM) the microstructure takes the form of bilamellar vesicles (BLV) for compositions containing less than 20 mol % of added C(12)E(3). Multilamellar vesicles (MLV) are the predominant microstructure for solutions richer in composition than 20 mol % C(12)E(3). At more than 80 mol % C(12)E(3), the solution microstructure reverts to that of a lamellar phase dispersion consistent with studies on the pure nonionic surfactant. At higher concentrations (60 mM) a wide continuous L beta phase region is observed for compositions in the range 20 to 80 mol % C(12)E(3). The fine details of the phase diagram were obtained from quantitative analysis of the SANS data using a well-established lamellar membrane model. Irrespective of the nonionic content, the bilayers are in general highly rigid, consistent with those stabilized by charge interactions. Furthermore estimates of the product of membrane moduli (compressibility and bending modulus) indicate that the different phase regions have very different membrane properties, however the magnitude of the variations observed are not predicted using existing theoretical treatments. 相似文献
