Melanin-concentrating hormone (MCH) is implicated in the feeding behavior in mammals affording a potential target to control overeating in people. Compound 1 (AMG 076) has been identified as a potent MCHr1 antagonist for the treatment of obesity. A synthesis suitable for the large-scale preparation of this lead candidate was developed to support preclinical studies. A Robinson annulation of benzylpiperidone and resolution of the desired enone from a mixture of the diastereomers afforded key intermediate 6 after a stereoselective hydrogenation. Subsequent Fischer indole synthesis with hydrazine 5 then provided the advanced intermediate, indole 2. Two complementary reductive amination strategies employing either aldehyde 3 or lactol 4 led to the synthesis of title compound 1.  相似文献

A new Re-doped nano-TiO2 photocatalyst was synthesized by immersion method. The novel doped nano-TiO2 photocatalyst utilizing visible light was firstly prepared. The doped nano-TiO2 powder was charactered by XRD, FTIR, UV-Vis, and its photocatalytic activity was tested through the photocatalytic degradation of methamidophos as a model compound under ultraviolet irradiating and in sunlight, respectively. In order to compare the photocatalytic activities, the same experiment was carried out for undoped nano-TiO2. The degradation ratio of methamidophos in the presence of doped nano-TiO2 reached 64.40% under sunlight for 12 h, which was 2.64% in the presence of undoped nano-TiO2. The degradation ratio of methamidophos in the presence of doped nanoTiO2 reached 90.39% under UV irradiationat 2.5 h, which was 51.29% in the presence of undoped nano-TiO2. All the results show that the doped TiO2 is a promising photocatalyst using sunlight for treating the organophosphorous pesticide wastewater. The article is published in the original.  相似文献

The Er3+:Y3Al5O12 as an upconversion luminescence agent, which can transform visible light into ultraviolet light, was synthesized by nitrate?Ccitrate acid and calcined method. Then, a novel photocatalyst, Er3+:Y3Al5O12/TiO2?CZrO2, was prepared using ultrasonic dispersion and liquid boiling method. The samples were characterized using X-ray diffraction (XRD) and scanning electron microscopy (SEM). In succession, the degradation process of organic dye was monitored by UV?CVis spectrum and ion chromatography for verifying the photocatalytic activity of Er3+:Y3Al5O12/TiO2?CZrO2. The influences on its photocatalytic activity such as Ti/Zr molar ratio, heat-treated temperature, and time were studied. In addition, the influences of initial concentration, Er3+:Y3Al5O12/TiO2?CZrO2 amount, solar light irradiation time, and organic dye category on the photocatalytic degradation efficiency were also investigated. It was found the photocatalytic activity of Er3+:Y3Al5O12/TiO2?CZrO2 was superior to Er3+:Y3Al5O12/TiO2 and Er3+:Y3Al5O12/ZrO2. Therefore, the Er3+:Y3Al5O12/TiO2?CZrO2 is a useful photocatalytic material for the wastewater treatment duo to efficient utilization of solar light.  相似文献

In this work, three anthraquinone derivants (Alizarin: 1,2-dihydroxy-9, 10-anthraquinone, Alizarin–DA: 1,2-dihydroxy-9, 10-anthraquinone-3-aminomethyl-N, N-diacetic acid and Alizarin–DA–Fe: 1,2-dihydroxy-9, 10-anthraquinone-3-aminomethyl-N, N-diacetate-Ferrous(III)) were used to study the sonodynamic and sonocatalytic damage of bovine serum albumin (BSA) molecules according to the hyperchromic effect of UV–vis spectra and quenching effect of intrinsic fluorescence. Meanwhile, some influencing factors such as ultrasonic irradiation time, anthraquinone derivants concentration and ionic strength on the damage of BSA molecules were also considered. The results show that the synergetic effect of anthraquinone derivants and ultrasonic irradiation can efficiently damage the BSA molecules. Finally, some special radical scavengers were used to determine the kind of generated reactive oxygen species (ROS) in the presence of three anthraquinone derivants under ultrasonic irradiation. The results show that the ROS, at least, including singlet oxygen (1O2) and hydroxyl radicals (OH) are generated during the sonodynamic and sonocatalytic processes. It is wished that this paper could offer some valuable references for the application of anthraquinone derivants in sonodynamic therapy (SDT) and sonocatalytic therapy (SCT) for tumor treatment.  相似文献

Synthesis of five and six membered heterocycles, indolines, 2,3-dihydrobenzofurans, chromans, isochromans, 1,2,3,4-tetrahydroquinolines, and 1,2,3,4-tetrahydroisoquinolines, in 70-99% yield by a ligand-free palladium catalyzed reductive Heck cyclization of phenyl bromides and chlorides, under mild conditions, is reported. Soybean protein has long been recognized as a source of dietary allergens for humans and animals with β-conglycinin being the major allergen. This paper presents a sandwich enzyme-linked immunosorbent assay (ELISA) that allows for the detection of trace amount of β-conglycinin in soybean and soybean products. In the sandwich ELISA, mouse anti-β-conglycinin monoclonal antibody (Mab 5C5) was used as coating antibody, and rabbit anti-β-conglycinin polyclonal antibody (Pab) was used as secondary antibody. The assay showed high specificity for β-conglycinin with minimum cross-reactions with other soy proteins. The practical working range for the determination of β-conglycinin using the developed assay was 3–100 ng mL−1 and the limit of determination (LOD) was 1.63 ng mL−1. The recoveries of β-conglycinin in spiked soybean samples were between 88.1% and 106.6% with relative standard deviation less than 8.9% (intra-day) and 13.1% (inter-day). The developed method was used to analyze 469 soybean seed samples from different sources as well as five soybean products treated with different processing techniques. The data showed that the concentration of β-conglycinin decreased significantly after processing, especially for soybean protein isolation, where the concentration of β-conglycinin dropped to nearly zero. The assay provides a specific and sensitive method for the screening of β-conglycinin and allows for further investigation into hypersensitive mechanisms of soybean proteins and development of soybean processing techniques to reduce their negative effects.  相似文献

This Letter focuses on the synchronization in a class of dynamical complex networks with each node being a deterministic ratchet. In virtue of the technique derived from pendulum-like nonlinear analytic theory and Kalman–Yakubovich–Popov (KYP) lemma, simple linear matrix inequality (LMI) formulations are established to guarantee the stable synchronization of such networks. An interesting conclusion is reached that the stability of synchronization in the coupled whole N-dimensional networks can be converted into that of the simplest 2-dimensional space.  相似文献

Phase separation of the poly(3-hexylthiophene) (P3HT) and [6,6]-phenyl-C61-butyric acid methyl ester (PCBM) active layer (ATL) was investigated by varying their relative ratio in the organic solar cells (OSCs). With the help of the UV/visible spectrophotometer, optical microscopy and scanning electron microscope, we found that the cluster of PCBM at the interface or surface was affected by Al cathode, the composition of the blends and thermal annealing. The disc-like shape crystals of PCBM substituted for the needle-like ones at higher PCBM compositions at the ATL/Al interface, which led to stronger contacts and bigger contact area. It could make short circuit current density increase, but may affect the blend morphology and result in parallel resistance and open circuit voltage decreased with the PCBM ratio increasing from 40 to 60%. The microstructure of the P3HT:PCBM ATL, determined by the composition dependent phase separation, supported the optimized performance of the OSCs with the composition of 40-50% PCBM.  相似文献

The objective of this research was to develop a multiplex dipstick immunoassay method for the simultaneous determination of multi-veterinary drug residues, such as β-agonists, sulfonamides, and tetracyclines in milk, urine, and serum. The multiplex dipstick assay format was based on an indirect competitive approach: Three test lines (different antigens) and one control line (goat anti-mouse IgG) were located on the strip membrane. Labeled antibodies were freeze-dried in microwells. Samples did not require pretreatment and could be directly analyzed within 10 min. Threshold levels in different sample matrices were visually estimated at 0.3–0.45 ng mL−1 for clenbuterol; 3–4 ng mL−1 for sulfadiazine; and 4.5–6 ng mL−1 for tetracycline, respectively. The linear relationship between the concentrations of veterinary drug residues and the Au nanoparticles plasmon absorbance allowed quantitative determination of these veterinary drug residues. The recoveries of clenbuterol, sulfadiazine and tetracycline in spiked samples ranged from 78.4% to 112.6%, and the relative standard deviations were below 11.2%. Analysis of animal samples suggested that the proposed multiplex dipstick assay method was consistent with the LC-MS/MS method. The percentage of false results was less than or equal to 5%. Thus, the proposed multiplex dipstick assay is inexpensive, easy-to-use, and suitable for the purposes of rapid and comprehensive screening of 3 families of β-agonists, sulfonamides and tetracyclines including 26 drugs in animal body fluids.  相似文献
