Asymmetric disulfide conjugates of mercaptosuccinyl tobacco mosaic virus (TMV ~ SH) with Nα-desacetyl-Nα-5-(mercaptovaleryl)-α-melanotropin were prepared via the S-sulfoderivative of the peptide. The conjugates, TMV ~ S? S ~ α-MSH(n), contained up to n = 330 disulfide-linked peptide molecules/virion. Similarly, fluorescent conjugates, Rh(m) ~ TMV ~ S? S ~ α-MSH(n) were prepared, containing m ≈? 200 rhodamine molecules linked to the virions by thiourea bridges. Such conjugates were designed to study α-MSH receptor localization and dynamics (mainly internalization), because the carrier virions which served to enhance specific receptor binding and as fluorescent or radioactive markers may be detached from the neuropeptides at will by reduction. Reduction occurred in solution and on the cell surface, but not in the cytoplasm, thus allowing detection of internalized agonist-receptor complexes. The conjugates were superpotent agonists for tyrosinase stimulation in Cloudman S-91 melanoma cell cultures, but were inactive for cyclic AMP accumulation. Their rather rapid internalization and the influence of reducing agents and other agonists on their biologic activity suggest a close connection between receptor location and biologic response as well as the presence of essential receptor HS-groups.  相似文献

The transition from stratified to dual continuous oil–water flow (where each phase retains its continuity but there is dispersion of one phase into the other) as well as the dispersed phase fractions in the layers of the dual continuous pattern, were studied experimentally. Transition to this pattern from stratified flow occurs when drops of one phase appear into the other (onset of entrainment). The studies were carried out in a 38 mm ID horizontal stainless steel test section using two different inlet geometries, a T- and a Y-junction. The patterns were visualized through a transparent acrylic section located at 7 m from the inlet using a high speed video camera. Phase distribution measurements in a pipe cross section were obtained just before the acrylic section with a local impedance probe and the results were used to calculate the volume fraction of each phase entrained into the other. The onset of entrainment was found to occur at lower superficial water velocities as the oil superficial velocities increased. However, the inlet geometry did not affect significantly the transition line. During dual continuous flow, the dispersion of one phase into the opposite was found to extend further away from the interface with increasing water superficial velocity for a certain oil superficial velocity. An increase in the superficial water velocity increased the entrained fraction of water in oil (Ew/o) but there was no trend with the oil velocity. Similarly, an increase in the superficial oil velocity increased the fraction of oil drops in water (Eo/w) but the water velocity had no clear effect. The entrainment fractions were affected by the inlet geometry, with the T-inlet resulting in higher entrainment than the Y-inlet, perhaps because of the increased mixing induced by the T-inlet. The difference between the two inlets increased as the oil and water velocities increased.  相似文献

A novel immunosensor based on graphite screen-printed electrodes (SPEs) modified with bismuth citrate was developed for the voltammetric determination of C-reactive protein (CRP) in human serum using quantum dots (QDs) labels. The sandwich-type immunoassay involved physisorption of CRP capture antibody on the surface of the sensor, sequential immunoreactions with CRP and biotinylated CRP reporter antibody and finally reaction with streptavidin-conjugated PbS QDs. The quantification of the target protein was performed with acidic dissolution of the PbS QDs and anodic stripping voltammetric detection of the Pb(II) released. Detection was performed at bismuth nanodomains formed on the sensor surface during the electrolytic preconcentration step, as bismuth citrate was reduced to metallic bismuth simultaneously with the deposition of Pb on the surface of the immunosensor. Under optimal conditions, the response was linear over the range 0.2–100 ng mL−1 CRP and the limit of detection was 0.05 ng mL−1 CRP. Since the modified SPE serves as both the biorecognition element and the QDs reader, the analytical procedure is simplified, the drawbacks of existing electroplated immunosensors are minimized while the proposed disposable sensing platform provides convenient, low-cost and ultrasensitive detection of proteins and wider scope for mass-production.  相似文献

Three new aromatic sesquiterpenes (1, 2, and 4), one new dimeric sesquiterpene of the cyclolaurane-type (3), one sesquiterpene alcohol of bisabolene type (8) along with three previously reported metabolites (5-7), were isolated from the organic extracts of Laurencia microcladia, collected from the Chios island in the North Aegean Sea. The structures of the new natural products, as well as their relative stereochemistries, were established by means of spectral data analyses, including 2D experiments. The cytotoxicity of the isolated metabolites was evaluated against five human tumor cell lines.  相似文献

Surfaces for guided cell adhesion and growth are indispensable in several diagnostic and therapeutic applications. Towards this direction, four diblock copolymers comprising polyethylene glycol (PEG) and poly(2-tetrahydropyranyl methacrylate) (PTHPMA) are synthesized employing PEG macroinitiators of different chain lengths. The copolymer with a 5000 Da PEG block and a PEG-PTHPMA comonomers weight ratio of 43–57 provides a film with the highest stability in the culture medium and the strongest cell repellent properties. This copolymer is used to develop a positive photolithographic material and create stripe patterns onto silicon substrates. The highest selectivity regarding smooth muscle cell adhesion and growth and the highest fidelity of adhered cells for up to 3 days in culture is achieved for stripe patterns with widths between 25 and 27.5 µm. Smooth muscle cells cultured on such patterned substrates exhibit a decrease in their proliferation rate and nucleus area and an increase in their major axis length, compared to the cells cultured onto non-patterned substrates. These alterations are indicative of the adoption of a contractile rather than a synthetic phenotype of the smooth muscle cells grown onto the patterned substrates and demonstrate the potential of the novel photolithographic material and patterning method for guided cell adhesion and growth.  相似文献

A series of novel four‐arm A2B2 and A2BC and five‐arm A2B2C miktoarm star polymers, where A is poly(dimethylsiloxane) (PDMS), B is polystyrene (PS), and C is polyisoprene (PI), were successfully synthesized by the combination of chlorosilane and benzyl chloride linking chemistry. This new and general methodology is based on the linking reaction of in‐chain benzyl chloride functionalized poly(dimethylsiloxane) (icBnCl–PDMS) with the in‐chain diphenylalkyl (icD) living centers of PS‐DLi‐PS, PS‐DLi‐PI, or (PS)2‐DLi‐PI. icBnCl–PDMS was synthesized by the selective reaction of lithium PDMS enolate (PDMSOLi) with the chlorosilane groups of dichloro[2‐(chloromethylphenyl)ethyl]methylsilane, leaving the benzyl chloride group intact. The icD living polymers, characterized by the low basicity of DLi to avoid side reactions with PDMS, were prepared by the reaction of the corresponding living chains with the appropriate chloro/bromo derivatives of diphenylethylene, followed by a reaction with BuLi or the living polymer. The combined molecular characterization results of size exclusion chromatography, 1H NMR, and right‐angle laser light scattering revealed a high degree of structural and compositional homogeneity in all miktoarm stars prepared. The power of this general approach was demonstrated by the synthesis of a morphologically interesting complex miktoarm star polymer composed of two triblock terpolymer (PS‐b‐PI‐b‐PDMS) and two diblock copolymer (PS‐b‐PI) arms. © 2006 Wiley Periodicals, Inc.
