Healing of tendon ruptures represents a major challenge in musculoskeletal injuries and combinations of biomaterials with biological factors are suggested as viable option for improved healing. The standard approach of repair by conventional suture leads to incomplete healing or rerupture. Here, a new elastic type of DegraPol® (DP), a polyester urethane, is explored as a delivery device for platelet‐derived growth factor—BB (PDGF‐BB) to promote tendon healing. Using emulsion electrospinning as an easy method for incorporation of biomolecules within polymers, DegraPol® supports loading and release of PDGF‐BB. Morphological, mechanical and delivery device properties of the bioactive DP scaffolds, as well as differences arising due to different electrospinning parameters are studied. Emulsion electrospun DP scaffolds result in thinner fibers than pure DP scaffolds and experience decreased strain at break [%], but high enough for successful surgeon handling. PDGF‐BB is released in a sustained manner from emulsion electrospun DP, but not completely, with still large amount of it being inside the polymeric fibers after 30 d. In vitro studies show that the bioactive scaffolds promote tenocyte proliferation in serum free and serum+ conditions, demonstrating the potential of this surgeon‐friendly bioactive delivery device to be used for tendon repair.

Alpha4/3 conotoxins: phylogenetic distribution, functional properties, and structure-function insights 总被引:1,自引:0,他引:1
This review examines the alpha4/3 conotoxins as an example of molecular diversity in a class of compounds that have evolved in a group of closely related species in a single phylogenetic lineage. The species examined belong to Stephanoconus, a clade of Conus, a genus that contains 500-700 different species of carnivorous marine snails. We examine earlier work that describes the identification and characterization of alpha-ImI, the founding alpha4/3 toxin, and two other alpha4/3 toxins, alpha-ImII and alpha-RgIA. These three toxins all inhibit nicotinic acetylcholine receptors (nAChRs) belonging to a subset of nAChRs that are composed of only alpha subunits; they are, however, diverse in terms of the all-alpha subtype they preferentially antagonize and the receptor site that they bind to. We thus speculate that the alpha4/3 toxins may be a rich source of functionally diverse all-alpha subunit nAChR inhibitors. We review extensive work that has established a detailed model for alpha-ImI binding to one of its preferred nAChR subtypes (the alpha7 nAChR) and, by comparing the alpha-ImI, alpha-ImII and alpha-RgIA sequences demonstrate how structural features of alpha4/3 peptides that account for their diverse functional properties can be identified. This approach is extended to derive models of receptor-toxin binding that may account for the different subtype specificities of alpha4/3 peptides. We also speculate on how rational modification of alpha4/3 toxins may allow engineering of ligands with desired subtype specificities. The chemical diversity produced by the closely related animals in Stephanoconus is thus functionally differentiated, although structurally homologous. 相似文献

Breathe easy : Reversible H2O and NH3 gas uptake by 2D calcium tetraphosphonates (see figure) is accompanied by framework structural changes similar to those previously reported for some carboxylate‐based hybrids. This breathing mechanism is accompanied by a volume increase of 55 %, while maintaining the topology and crystallinity of the material.

Here we report on the domain wall dynamics in amorphous glass-coated FeCuNbSiB microwires measured in the temperature range from 77 up to 400 K. At low temperatures below 200 K, the domain wall velocity is proportional to the applied magnetic field. At temperatures above 200 K, two regions have been found: one with low domain wall mobility at low fields and another one with high domain wall mobility at high fields. The different regions of the domain wall dynamics are treated in terms of the change of the domain wall configuration from transversal to vortex one. Moreover, non-linear regime is shown at low fields at the temperature 373 K as a result of the domain wall interaction with the local defects. 相似文献

The syntheses of two novel platinum(IV) complexes of formula [PtX2(S,S-eddp)]·nH2O (S,S-eddp = ethylenediamine-N,N′-di-S,S-2-propanoate ion, X = chlorido (1) or bromido (2), n = 4, 0) are reported. The complexes have been obtained by direct reaction of corresponding potassium hexahalogenidoplatinate(IV) with neutralized ethylenediamine-N,N′-di-S,S-2-propanoic acid (H2-S,S-eddp). The complexes were characterized by elemental analysis, infrared, 1H and 13C NMR spectroscopy. The spectroscopically predicted geometrical configurations of the obtained complexes were confirmed by X-ray analyses of the crystal structures of the s-cis-[Pt(S,S-eddp)Cl2]·4H2O and uns-cis-[Pt(S,S-eddp)Br2]. These complexes displayed significantly lower in vitro cytotoxicity in comparison to cisplatin. 相似文献

We study thermodynamics of black hole solutions in Lanczos–Lovelock anti-de Sitter gravity in \(d+1\) dimensions coupled to nonlinear electrodynamics and a Stückelberg scalar field. This class of theories is used in the context of gauge/gravity duality to describe a high-temperature superconductor in \(d\) dimensions. A larger number of coupling constants in the gravitational side is necessary to widen the domain of validity of physical quantities in dual quantum field theory (QFT). We regularize the gravitational action and find the finite conserved quantities for a planar black hole with scalar hair. Then we derive the quantum statistical relation in the Euclidean sector of the theory, and we obtain the exact formula for the free energy of the superconductor in the holographic QFT. Our result is analytic and it includes the effects of backreaction of the gravitational field. We further discuss on how this formula could be used to analyze second order phase transitions through the discontinuities of the free energy, in order to classify holographic superconductors in terms of the parameters in the theory. 相似文献

New salicylic (2-hydroxybenzoic) acid derivatives 1–6 were prepared by conventional heating or microwave irradiation of a mixture consisting of methyl salicylate and the corresponding amino alcohol (2,2′-dihydroxydiethylamine, 2,2′,2″-trihydroxytriethylamine or N-phenyl-2,2′-dihydroxydiethylamine) and metallic sodium as catalyst. For compounds 1, 3, and 5 X-ray structure analysis was performed, as well as molecular mechanics calculations (MMC), to define their conformation in terms of their energy minima. Comparison of crystal and MMC structures for these three compounds (1, 3, and 5) revealed that the intramolecular hydrogen bonds play an important role, stabilizing conformation of the most part of the molecule. The antioxidant activity and cytotoxicity of the synthesized derivatives were evaluated in a series of in vitro tests. The newly synthesized compounds exhibited strong activity against hydroxyl radical, as well as promising lipid peroxidation inhibition. The study showed that the electronic effects of the groups at the N atom are responsible for neutralization of the OH radical, i.e., antioxidant activity. Compounds 1–3 exhibited sub-micromolar cytotoxicity against HeLa S3, whereas compounds 1, 3 and 5 efficiently inhibited the growth of PC3 cells. 相似文献

A macroporous copolymer of glycidyl methacrylate and ethylene glycol dimethacrylate, poly(GMA-co-EGDMA), with various surface characteristics and mean pore size diameters ranging from 44 to 200 nm was synthesized, modified with 1,2-diaminoethane, and tested as a carrier for immobilization of horseradish peroxidase (HRP) by two covalent methods, glutaraldehyde and periodate. The highest specific activity of around 35 U g?1 dry weight of carrier was achieved on poly(GMA-co-EGDMA) copolymers with mean pore diameters of 200 and 120 nm by the periodate method. A study of deactivation kinetics at 65 °C and in 80 % dioxane revealed that periodate immobilization also produced an appreciable stabilization of the biocatalyst, while stabilization factor depended strongly on the surface characteristics of the copolymers. HRP immobilized on copolymer with a mean pore diameter of 120 nm by periodate method showing not only the highest specific activity but also good stability was further characterized. It appeared that the immobilization resulted in the stabilization of enzyme over a broader pH range while the Michaelis constant value (K m) of the immobilized HRP was 10.8 mM, approximately 5.6 times higher than that of the free enzyme. After 6 cycles of repeated use in a batch reactor for pyrogallol oxidation, the immobilized HRP retained 45 % of its original activity. 相似文献
