A low-cost, disposable card for rapid polymerase chain reaction

A low-cost, disposable card for rapid polymerase chain reaction (PCR) was developed in this work. Commercially available, adhesive-coated aluminum foils and polypropylene films were laminated with structured polycarbonate films to form microreactors in a card format. Ice valves [1] were employed to seal the reaction chambers during thermal cycling and a Peltier-based thermal cycler was configured for rapid thermal cycling and ice valve actuation. Numerical modeling was conducted to optimize the design of the PCR reactor and investigate the thermal gradient in the reaction chamber in the direction of sample thickness. The PCR reactor was experimentally characterized by using thin foil thermocouples and validated by a successful amplification of 10 copy of E. coli tuf gene in 27 min.     

Electronic spectroscopy of the 3d Rydberg states of NO-Rg (Rg=Ne,Ar,Kr,Xe) van der Waals complexes

We have employed (2+1) resonance-enhanced multiphoton ionization spectroscopy to record electronic absorption spectra of NO-Rg (Rg=Ne,Ar,Kr) van der Waals complexes. The nitric oxide molecule is the chromophore, and the excitation corresponds to an electron being promoted from the 2ppi* orbital to 3dsigma, 3dpi, and 3ddelta Rydberg states. We review the ordering of the 3dlambda states of NO and use this as a basis for discussing the 3d components in the NO-Rg complexes, in terms of the interactions between the Rydberg electron, the core, and the Rg atom. Predissociation of the H' 2Pi state occurs through the F2Delta state for NO-Ar and NO-Kr, and this will be considered. We shall also outline problems encountered when trying to record similar spectra for NO-Xe, related to the presence of atomic Xe resonances.     

Experimental studies of the Progressing Batch Reactor

The data obtained in the single reactor experiments has shown the use of 1/2-in. chips to produce results that are reliable and repeatable. These data are not sensitive to variations in wood chip packing or prehydrolysis procedures. The latter is important since multireactor studies will try various prehydrolysis strategies while investigating the operational characteristics of a multireactor system. The possible importance of hydrodynamics within the wood chip bed was demonstrated by the data from the 1/4- and 1/8-in. chip experiments. The importance of shrinking wood chip bed was demonstrated by the reactor’s inferior performance in the upflow versus the downflow mode. A hydrodynamic study should be undertaken to investigate these phenomena. This study can proceed concurrently with work on a tworeactor system that can give a preliminary glimpse at the operation and benefits of a multireactor system.     

A Note on Local Rigidity

The aim of this note is to give a geometric proof for classical local rigidity of lattices in semisimple Lie groups. We are reproving well known results in a more geometric (and hopefully clearer) way.     

Observation of laser satellites in a plasma produced by a femtosecond laser pulse

Laser satellites are detected in the emission spectra of magnesium and aluminum plasmas produced by femtosecond laser pulses. This is made possible by the realization of picosecond time resolution in a high-luminosity x-ray spectrograph with a spherically curved mica crystal. The temporal characteristics of these newly recorded spectral lines show unequivocally that they are formed as a result of nonlinear processes. Pis’ma Zh. éksp. Teor. High-dynamic-range laser-pulse-contrast measurement with a plasma-shuttered streak camera

The dynamic range of a picosecond visible streak camera has been improved by the combination of a plasma shutter and multishot averaging performed with a photoconductive switch sweep circuit. We use this technique to measure the contrast of a 100-fs laser pulse over 2 ns with a dynamic range of 7 orders of magnitude.     

Phenol derivatized hexahydropyrimidines prepared from Mannich condensations

Six new phenol derivatized hexahydropyrimidines (1,3-bis-(2-hydroxy-3,5-di(X)benzyl) hexahydropyrimidine and 5,5′-dimethyl-1,3-bis-(2-hydroxy-3,5-di(X)benzyl) hexahydropyrimidine where X = Me, t-Bu, and Cl) were synthesized in one-pot in 33-79% yield using Mannich condensations.     

Dilute sulfuric acid hydrolysis of biomass for ethanol production

A series of pilot plant experiments have been conducted to compare the performance of a system utilizing two percolation reactors in series to a single reactor system. Although theoretically capable of producing higher glucose yields or concentrations, the two-reactor system concentrations were approximately the same and the yields were considerably lower than those from the single reactor study. An associated kinetics study found the glucose degradation kinetics to be accelerated by chromium ions, but this effect was greatly reduced in the presence of wood. The presence of metal surfaces also increased the rate of degradation even without large ion concentrations. The poor performance of the reactor system is proposed to be caused by intraparticle glucose diffusion effects and the catalytic effect on glucose degradation reactions of chromium ions from the corrosion of stainless steel by the acid. Strategies for reducing the effects of these phenomena on PBR performance are presented.

     

Electronic spectroscopy of para-fluorotoluene clusters

We report (1 + 1) resonance-enhanced multiphoton ionization spectra for clusters of para-fluorotoluene (pFT)_n (n = 1–11). After n = 2, the spectra appear to have converged in appearance, suggestive of a dimer chromophore, with weak bonding for subsequent additions of pFT molecules. The spectra also indicate dramatic, vibrational-mode-sensitive changes of oscillator strength between the monomer and the clusters. We also briefly describe the results of probing different parts of the expansion, and varying the laser power.     

Design, Synthesis, and Testing of Polyamine Vectored Iron Chelators

Iron chelators have been shown to control the growth of cancer cells in culture by sequestering exogenous iron in the media. Thus, the ligands prevent cellular access to the metal. However, because transferrin provides iron to tumor cells in animals, chelators have not been effective antitumor agents. Polyamine chelator conjugates in which the polyamine vectored ligands into cells were far more active than the free chelators themselves. However, the free ligands were not released from the vector once in the cell. The current study focuses on the synthesis and preliminary evaluation of a polyamine chelator conjugate capable of releasing the free ligand intracellularly via a nonspecific esterase.
