The electron impact mass spectra of the acetates of zinc, magnesium, cobalt and manganese have been investigated using a direct inlet probe. Volatile tetrahedral complexes are produced on heating, and ions of the form [M4(OCOCH3)6O] (M = Co and Mn) and [N4(OCOCH3)5O]+ (N = Zn or Mg) are observed. A mixture of magnesium and cobalt acetates produces ions of the form [MgnCo4–n(OCOCH3)6O].  相似文献

The amino acid analog azaproline (azPro) contains a nitrogen atom in place of the C(alpha) of proline. Peptides containing azPro were shown to stabilize the cis-amide conformer for the acyl-azPro bond and prefer type VI beta-turns both in crystals and in organic solvents by NMR. The increased stability for cis-amide conformers was relatively minor with respect to the trans-conformers. Further, their conformational preferences were depended on solvent. To elucidate the impact of azPro substitution on amide cis-trans isomerism and peptide conformation, this paper reports ab initio studies on azPro derivatives and a comparison with their cognate Pro derivatives: 1-acetyl-2-methyl pyrrolidine (1), 1-acetyl-2-methyl pyrazolidine (2), Ac-Pro-NHMe (3), Ac-azPro-NHMe (4), Ac-azPro-NMe(2) (5), Ac-azAzc-NHMe (6), and Ac-azPip-NHMe (7). Conformational preferences were explored at the MP2/6-31+G** level of theory in vacuo. Solvation effects for 1 and 2 were studied implicitly using the polarizable continuum model and explicitly represented by interactions with a single water molecule. An increase in the conformational preference for the cis-amide conformer of azPro was clearly seen. An intramolecular hydrogen bond occurred solely in the trans-amide conformer that reduced the preference for the cis-conformer by 2.2 kcal/mol. The larger ring homolog aza-pipecolic acid (azPip), in which this internal hydrogen bond was diminished, significantly augmented stabilization of the cis-amide conformer. In aqueous solution, the preference for the cis-amide conformers was greatly reduced, mainly as a result of interaction between water and the lone pair of the alpha-nitrogen in the trans-amide conformer that was 3.8 kcal/mol greater than that in the cis-conformer. In the azPro analog, the energy barrier for cis-trans amide isomerization was 6 kcal/mol less than that in the cognate Pro derivative. Because the azPro derivatives can stabilize the cis-amide bond and mimic a type VI beta-turn without incorporation of additional steric bulk, such a simple chemical modification of the peptide backbone provides a useful conformational constraint when incorporated into the structure of selected bioactive peptides. Such modifications can scan receptors for biological recognition of reverse turns containing cis-amide bonds by the incorporation of type VI beta-turn scaffolds with oriented appended side chains.  相似文献

Fibronectin and proteoglycans as determinants of cell-substratum adhesion.   总被引:17,自引:0,他引:17  
When normal or SV40-transformed Balb/c 3T3 cells are treated with the Ca++-specific chelator EGTA, they round up and pull away from their footpad adhesion sites to the serum-coated tissue culture substrate, as shown by scanning electron microscope studies. Elastic membranous retraction fibers break upon culture agitation, leaving adhesion sites as substrate-attached material (SAM) (Cells leave "footprints" of substrate adhesion sites during movement by a very similar process.) SAM contains 1-2% of the cell's total protein and phospholipid content and 5-10% of its glucosamine-radiolabeled polysaccharide, most of which is glycosaminoglycan (GAG). By one- and two-dimensional sodium dodecyl sulfate-polyacrylamide gel electrophoresis, there is considerable enrichment in SAM for specific GAGs; for the glycoprotein fibronectin; and for the cytoskeletal proteins actin, myosin, and the subunit protein of the 10 nm-diameter filaments. Fibrillar fibronectin of cellular origin and substratum-bound fibronectin of serum origin (cold-insoluble globulin, CIg) have been visualized by immunofluorescence microscopy. The GAG composition in SAM has been examined under different cellular growth and attachment conditions. Heparan sulfate content correlates with glycopeptide content (derived from glycoprotein). Newly attaching cells deposit SAM with principally heparan sulfate and fibronectin and little of the other GAGs. Hyaluronate and chrondroitin proteoglycans are coordinately deposited in SAM as cells begin spreading and movement over the substrate. Cells attaching to serum-coated or CIg-coated substrates deposited SAM with identical compositions. The proteoglycan nature of the GAGs in SAM has been examined, as well as the ability of proteoglycans to form two classes of reversibly dissociable "supramolecular complexes" - one class with heparan sulfate and glycopeptide-containing material and the second with hyaluronate-chondroitin complexes. Enzymatic digestion of "intact" SAM with trypsin or testicular hyaluronidase indicates that (1) only a small portion of long-term radiolabeled fibronectin and cyto-skeletal protein is bound to the substrate via hyaluronate or chondroitin classes of GAG; (2) most of the fibronectin, cytoskeletal protein and heparan sulfate coordinately resist solubilization; and (3) newly synthesized fibronectin, which is metabolically labile in SAM, is linked to SAM by hyaluronate- and/or chondroitin-dependent binding. All of our studies indicate that heparan sulfate is a direct mediator of adhesion of cells to the substrate, possibly by binding to both cell-surface fibronectin and substrate-bound CIg in the serum coating; hyaluronate-chondroitin complexes in SAM appear to be most important in motility of cells by binding and labilizing fibronectin at the periphery of footpad adhesions, with subsequent cytoskeletal disorganization.  相似文献

An inexpensive multisample fast-atom bombardment (FAB) probe assembly was designed for high-throughput analysis of samples on a VG ZAB-SE mass spectrometer. The system consists of a vacuum lock system and a FAB probe whose tip contains five or more sample wells. The probe enters the mass spectrometer source region perpendicular to the secondary ion beam axis, The probe is maintained at high voltage on contact with a spring clip attached to the screen plate of the source block. Sample throughput with the multisample probe is twice that of a coaxial probe, with about twice the sensitivity and no sample cross contamination.  相似文献

The cyclic voltammetric peaks for charging trapping and untrapping reactions between the inner and outer redox polymer films of five bilayer electrodes are compared to a theory for control of the rate of charge trapping by electron diffusion rates in the inner polymer film. The five bilayer electrodes use various different redox polymer films (electropolymerized poly-pyridine complexes of Fe, Ru, and Os, and polyvinylferrocene) arranged in different orders. The currents on the rising edge of the bilayer trapping and untrapping peaks follow the electron diffusion theory up to ca. 80% of the peak current; currents thereafter are controlled by another process(es). The analysis yields values for the electron diffusion constants in the inner bilayer polymer films, which agree with one another for different bilayers having the same inner film polymer films and which also agree with independent determinations by other methods. Two of the bilayers are made from the same two polymers, arranged in different inner-outer order. These bilayers also illustrate the occurrence of a “leak reaction”, in which charge trapped in the outer film is discharged via a thermodynamically unfavorable electron transfer reaction with the inner polymer film.  相似文献

The complexing ability of derivatives of azobenzene (I) is well known (I) and responsible for the production of a wide variety of dyestuffs and analytical chemicals. While the azo group generally participates in the coordination, the determination of the degree of its interaction is complicated by other functional groups which are also coordinated with the transition metal. In a previous publication (2), we reported the preparation of the silver and palladium complexes of benzo[c]cinnoline (II) and proposed that these results might be used to explain the electron donor properties of azobenzene. Different forms of perturbation theory for the calculation of correlation energy in both closed-and open-shell systems are discussed. For closed-shell systems, Epstein–Nesbet perturbation theory is compared with Møller–Plesset (MP ) perturbation theory based on canonical Hartree–Fock orbitals and with MP theory based on internally consistent SCF orbitals. The traditional MP theory gives superior results despite its use of an inferior zeroth-order Hamiltonian. This behavior is rationalized in terms of the larger denominators present in the traditional MP theory. These conclusions are used to support the restricted open-shell perturbation methods proposed recently by Murray and Davidson, and these new methods are compared with spin-restricted Epstein–Nesbet theory and the unrestricted MP (UMP ) approach. © 1992 John Wiley & Sons, Inc.  相似文献

The thermal, hydrolytic and photochemical oxidation of blue water pipe material has been studied using Fourier Transform infra-red microscopy, differential scanning calorimetry (DSC) and hydroperoxide analysis. The results indicate that oxidation of the pipe occurs predominantly on the outer surface and to a lesser extent on the bore, often with little or no change in the middle layers, FTIR analysis of microtomed sections of the pipe supports the DSC analysis (oxidation induction time at 200°C—OIT) and indicates leaching and consumption of the polymer antioxidants at the outer surface of the pipe. Oxidation profiles at 80°C in water, as measured using carbonyl index, indicate an unusual hydrolytic oxidation and extraction of the carbonylic oxidation products only at the outer pipe surface to a depth of about 0·5 mm resulting in high hydroperoxidation levels. These oxidation analyses are consistent with density profile changes through the pipe wall. Whilst water is concluded to have an important influence in controlling pipe stability, which, in turn, is governed by the temperature and extractability of the polymer antioxidants, ultraviolet light is also seen to have a similar detrimental effect. The influence of these various degradative parameters on the long-term stability of pipe is discussed with a view to elucidating the mechanisms involved.  相似文献
