Experiences with carrier-mediated transport in liquid-phase microextraction

Different organic borates, phosphates, sulphates, and carboxylic acids are evaluated as extraction carriers in three-phase liquid-phase microextraction (LPME). Hydrophilic basic drugs form ion-pairs with the carriers and are extracted as ion-pair complexes into an organic liquid membrane of n-octanol or peppermint oil immobilized in the pores of a polypropylene hollow fiber. From this point, the basic drugs are released into a 20-microL solution of 50mM HCl placed inside the lumen of the hollow fiber (acceptor solution). Simultaneously, the carrier is neutralized by protons from the acceptor solution (protonated to maintain the charge balance). Both water-soluble and water-insoluble carriers are tested. One promising candidate among the water-soluble carriers is 1-heptanesulfonic acid. This is added to the sample solution to a final concentration of 25mM and served to ion-pair the analytes within the sample solution. Among the less water-soluble candidates, a mixture of di(2-ethylhexyl) phosphate (DEHP) and tris(2-ethylhexyl) phosphate (TEHP) serve as efficient carriers. Ten percent (w/w) of each of DEHP and TEHP are added to the organic liquid membrane, and these carriers principally worked through ion-pairing with the analytes at the interface between the sample solution and the organic liquid membrane. Several carriers are found to be compatible with human plasma samples, and bromthymol blue is particularly efficient in combination with these protein-containing matrices. Following optimization of the conditions for bromthymol blue, including saturation of the plasma samples with sodium sulphate, extraction recoveries between 45% and 75% are obtained for eight model drugs after 60 min of extraction. With bromthymol blue as the carrier, highly acceptable validation data are obtained for phenylpropanolamine and practolol extracted from human plasma.     

Aminopentadiene imines from zincke salts of 3-alkylpyridines. Application to a synthesis of pyridinium salts from amino acids

The reaction of Zincke salts with primary amines to give pyridinium salts generally requires rather elevated temperature to go to completion (50-100 degrees C). It is shown that the addition of 1 equiv of a secondary amine allows formation, at ambient temperature, of intermediate aminopentadiene imine salts which can be isolated and were found to cyclize in acidic medium to give pyridinium salts at temperatures which do not exceed 50 degrees C. If this process has a tendency to give lower yields of pyridinium salt than the standard Zincke procedure, it can be advantageous in some cases, as illustrated by the synthesis of pyridinium salts from amino acids, a challenging reaction which does not work starting from Zincke salt in the absence of diethylamine. More generally, the reaction can be extended to primary amines featuring polar functions, as exemplified by a pyridinium salt synthesis (75 degrees C) in 55% yield from l-carnosine.     

Extraction of ketamine from urine using a miniature silica monolithic cartridge followed by quantification with liquid chromatography tandem mass spectrometry (LC-MS/MS)

The high surface area monolith with reactive hydroxyl group on its surface enables it to function as a miniature solid‐phase extraction (SPE) cartridge in size of 1 cm in diameter and 0.5 cm in length. The prepared silica monolith was characterized by Brunauer–Emmett–Teller method, scanning electron microscopy, X‐ray diffraction and Fourier transform infrared (FTIR) spectroscopy. Ketamine was selected as model analyte to validate the extraction efficiency of the prepared cartridge. The extracted ketamine from urine sample was quantitated by liquid chromatography tandem mass spectrometry (LC‐MS/MS) using positive electrospray ionization. The limit of detection and quantification for ketamine was found to be 0.5 and 1.6 ng/mL, respectively. The analysis exhibited linearity in the range of 10–500 ng/mL with coefficient of correlation >0.99. The recovery was found to be in the range of 89–107% with relative standard deviation (RSD) less than 10%. The prepared cartridge was found robust in extracting ketamine efficiently and repeatedly without any significant deterioration in its performance. Moreover, the batch‐to‐batch variations in the performance of the prepared cartridges in terms of % ion suppression of the extracts and recoveries of samples were small, suggesting the consistency in the properties of the monolith.     

Anti-tumor immunostimulatory effect of heat-killed tumor cells

As a part of our ongoing search for a safe and efficient anti-tumor vaccine, we attempted to determine whether the molecular nature of certain tumor antigens would influence immune responses against tumor cells. As compared with freeze-thawed or formaldehyde-fixed tumor antigens, heat-denatured tumor antigens elicited profound anti-tumor immune responses and greatly inhibited the growth of live tumor cells. The heat-denatured tumor antigens induced a substantial increase in the anti-tumor CTL response in the absence of any adjuvant material. This response appears to be initiated by strong activation of the antigen-presenting cells, which may recognize heat-denatured protein antigens. Upon recognition of the heat-denatured tumor antigens, macrophages and dendritic cells were found to acutely upregulate the expression of co-stimulatory molecules such as B7.2, as well as the secretion of inflammatory cytokines such as IL-12 and TNF-alpha. The results of this study indicate that heat-denatured tumor extracts might elicit protective anti-tumor adaptive immune responses and also raise the possibility that a safe and efficient adjuvant-free tumor vaccine might be developed in conjunction with a dendritic cell-based tumor vaccine.     

Nickel-catalyzed coupling of alkenes, aldehydes, and silyl triflates

A full account of two recently developed nickel-catalyzed coupling reactions of alkenes, aldehydes, and silyl triflates is presented. These reactions provide either allylic alcohol or homoallylic alcohol derivatives selectively, depending on the ligand employed. These processes are believed to be mechanistically distinct from Lewis acid-catalyzed carbonyl-ene reactions, and several lines of evidence supporting this hypothesis are discussed.     

Fermion superfluids of nonzero orbital angular momentum near resonance

We study the pairing of Fermi gases near the scattering resonance of the l not equal 0 partial wave. Using a model potential which reproduces the actual two-body low energy scattering amplitude, we have obtained an analytic solution of the gap equation. We show that the ground state of l=1 and l=3 superfluids are orbital ferromagnets with pairing wave functions Y11 and Y32, respectively. For l=2, there is a degeneracy between Y22 and a "cyclic state." Dipole energy will orient the angular momentum axis. The gap function can be determined by the angular dependence of the momentum distribution of the fermions.     

Compensation for pixel-mismatch errors in shift-multiplexed holographic data storage

To read out correct data from shift-multiplexed holographic data storage, we propose a novel method of pixel matching by use of a tiny plane-parallel refraction plate placed in the path of converging readout beams. Tilting the refraction plate produces a shift in the lateral position of the readout beam incident upon the holographic storage disk, leading to compensation for pixel-mismatch errors by an improvement in bit-error rate of 3 orders of magnitude.     

Two-component Fermi gas on internal-state-dependent optical lattices

We study the phase diagram of a one-dimensional, two-component (i.e., pseudo-"spin"-(1/2)) ultracold atomic Fermi gas. The two atom species can have different hopping or mass. A very rich phase diagram for equal densities of the species is found, containing Mott insulators and superfluids. We also discuss coupling such 1D systems and the experimental signatures of the phases. In particular, we compute the spin-structure factor at small momentum, which should reveal a spin gap.     

Vibronic states in single molecule electron transport

A scanning tunneling microscope was used to study the electron transport through individual copper phthalocyanine molecules adsorbed on an ultrathin Al(2)O(3) film grown on a NiAl(110) surface. The differential conductance spectra display series of equally spaced features, which are attributed to vibronic states of individual molecules. The coupling of the electron current to the vibronic modes was observed to depend on the structures of the adsorbed molecules. Vibronic features were not observed for molecules adsorbed on the bare NiAl(110) surface due to spectral broadening.
