Application of magdala red as a fluorescence probe in the determination of nucleic acids 总被引:3,自引:0,他引:3
A fluorescence quenching method was developed for the rapid determination of DNA and RNA using magdala red as fluorescence probe. In weakly acidic medium, the fluorescence of magdala red (lambdaex/lambdaem = 540/555 nm) can be largely quenched by DNA or RNA. The calibration graphs are linear over the range 0.01-1.2 microg/mL for both calf thymus DNA (CT DNA) and salmon DNA (SM DNA), and 0.015-1.0 microg/mL for yeast RNA, respectively. The corresponding detection limits are 6.0 ng/mL for CT DNA, 7.0 ng/mL for SM DNA and 15.0 ng/mL for yeast RNA, respectively. CT DNA could be determined in the presence of 20% (w/w) yeast RNA, and the relative standard deviation of six replicate measurements is 3.18% for 400 ng/mL of CT DNA. Interference from coexisting substances in the determination of DNA was also examined. Real samples were determined with satisfactory results. 相似文献

A method for the quantitation of polycyclic aromatic hydrocarbons (PAHs) in indoor and outdoor air by high-performance liquid chromatography (HPLC) with a spectrofluorometric detection and programmed excitation and emission wavelength pairs is proposed. The mobile phase is a linear gradient of methanol-water. The relative standard deviations (n = 5) are in the range 0.38-1.7% at concentration levels of 0.69-11.40 ng ml(-1). The determination limits (S N = 10 ) are 0.5-15.9 pg. The proposed method was successfully applied to quantitate 12 PAHs in gas phase and particulates in indoor and outdoor air. The recoveries of PAHs from gas phase and particulates were 95.7-117.5 and 94.8-112.4%, respectively. This highly sensitive automatic HPLC analysis for PAHs both in gas phase and particulates can be applied to indoor and outdoor survey. 相似文献

The effect of pH on the dilution enthalpies of D-p-hydroxyphenylglycine in phosphate buffer solutions has been investigated by isothermal titration microcalorimetry at 298.15 K. The corresponding homogeneous enthalpic interaction coefficients have been calculated according to the excess enthalpy concept. The results show that the enthalpies of dilution of D-p-hydroxyphenylglycine in phosphate buffer solutions at different pH are all positive. The overall trend is that enthalpies of dilution become more positive with an increase of pH, but there is a minimum of the enthalpy of dilution at pH = 7.0. A new strategy for the preparation of a DNA-modified carbon paste electrode is developed. It is found that the anodic response of mifepristone is greatly enhanced at the dsDNA-modified carbon paste electrode comparing with that obtained at the bare electrode, while the response at a ssDNA-modified electrode is similar to bare electrode. So the dsDNA-modified electrode is employed as a sensitive biosensor for the detection of mifepristone. A linear dependence of the peak currents on the concentration is observed in the range 2.0 x 10(-7) approximately 2.0 x 10(6) mol/L, with a detection limit of 1.0 x 10(-7) mol/L. The relative standard deviation is 4.3% for six successive determinations of 1.0 x 10(6) mol/L mifepristone. In the research on metallic glass, there are arguments against the "rigid band charge transfer", model which assumes a charge transfer from metalloid atoms to transition metal atoms to explain the experimental evidence of linear reduction of average atomic magnetic moment with the increasing concentration of metalloid atoms, but they could not explain the experimental relation of the reduction. In the present work, spin-polarized SCC-DV-X_a calculation for atomic clusters for metallic glass Co-B has been empoloyed to investigate the local electronic structure and magnetic property of the metallic glass. As opposed to the "rigid band charge transfer" model, calculation in the present work indicates that charge transfers from Co4s to both B and Co3d. It is found that there is Co3d4s-B2p hybird bonding in Co-B, which leads to the linear reduction of average atomic magnetic moment. Thus the explanation removes the above controversy. 相似文献
