The mono-N-amino salts of 3-phenylcinnoline, phthalazine, 1-phenylphthalazine, 4-phenylquinazoline, 2-phenylquinoxaline, 1,5- and 1,8-naphthyridines are in high yields prepared by direct N-amination of the parent heterocycles with O-mesitylenesulfonylhydroxylamine. With the one expection of 3-phenylcinnoline, the site of N-amination was determined by mass and nmr spectral techniques. Opioid receptors (ORs) are classified into three types (μ, δ, and κ), and opioid analgesics are mainly mediated by μOR activation; however, their use is sometimes restricted by unfavorable effects. The selective κOR agonist nalfurafine was initially developed as an analgesic, but its indication was changed because of the narrow safety margin. The activation of ORs mainly induces two intracellular signaling pathways: a G-protein-mediated pathway and a β-arrestin-mediated pathway. Recently, the expectations for κOR analgesics that selectively activate these pathways have increased; however, the structural properties required for the selectivity of nalfurafine are still unknown. Therefore, we evaluated the partial structures of nalfurafine that are necessary for the selectivity of these two pathways. We assayed the properties of nalfurafine and six nalfurafine analogs (SYKs) using cells stably expressing κORs. The SYKs activated κORs in a concentration-dependent manner with higher EC50 values than nalfurafine. Upon bias factor assessment, only SYK-309 (possessing the 3S-hydroxy group) showed higher selectivity of G-protein-mediated signaling activities than nalfurafine, suggesting the direction of the 3S-hydroxy group may affect the β-arrestin-mediated pathway. In conclusion, nalfurafine analogs having a 3S-hydroxy group, such as SYK-309, could be considered G-protein-biased κOR agonists. 相似文献

This study demonstrates the reaction behavior during the purification of model automotive exhaust gases over Pd catalysts before and after thermal degradation. In particular, to investigate the relationship between the Pd state and the reaction behavior of Pd/Al2O3 and Pd/CeO2−ZrO2 (CZ), operando X-ray absorption spectroscopy measurements were performed during purifying exhaust gases over real and model catalysts mimicking the degradation of Pd particles and CZ supports after accelerated aging tests. The NO reduction activity of the aggregated Pd metal species was as high as that of the highly dispersed Pd species, but hydrocarbon (HC) poisoning was significantly enhanced by the aggregation of Pd metal particles caused by thermal aging. The existence of a three-phase boundary (TPB) between the CZ, the Pd particles, and the gas phase strongly affected the catalytic activity at low temperatures, and the presence of a sufficient TPB facilitated the combustion of unburned HCs owing to the oxygen storage performance of CZ. Thus, the TPB reduced the poisoning of the precious metal surface by HC species at low temperatures. Gas transport properties of novel hyperbranched polyimide/hydroxy polyimide blends and their silica hybrid membranes were investigated. Gas permeability coefficients of the blend membranes showed positive deviation from a semilogarithmic additive rule. The enhanced gas permeability were resulted from the increase in free volume elements caused by the intermolecular interaction between terminal amine groups of the hyperbranched polyimide and hydroxyl groups of the hydroxy polyimide backbone. Additionally, CO2/CH4 separation ability of the blend membranes was markedly promoted by hybridization with silica. The remarkable CO2/CH4 separation behavior was considered to be due to characteristic distribution and interconnectivity of free volume elements created by the incorporation of silica. For the hyperbranched polyimide/hydroxy polyimide blend system, polymer blending and hybridization techniques synergistically provided the excellent CO2/CH4 separation ability. 相似文献

trans- and cis-1-(4-Dimethylaminophenyl)-6-(4-nitrophenyl)hex-3-ene-1,5-diynes (trans- and cis-DANE) were synthesized and their photochemical properties were studied. The absorption spectra of trans-DANE red-shifted compared with the parent compound bisphenylethynylethene (BEE) due to intramolecular charge transfer. The fluorescence spectra, Stokes shift, fluorescence lifetime, fluorescence quantum yield, and quantum yield of trans-to-cis photoisomerization of trans-DANE showed strong dependence upon the solvent polarity in the less-polar region. No fluorescence emission from trans-DANE was observed in medium-polar and polar solvents. The quantum yield of cis-to-trans isomerization was almost solvent independent. The donor-acceptor substituents shifted the equilibrium between the trans perpendicular triplet state and the trans planar triplet state to the trans triplet state, and resulted in an increase in the triplet lifetime. Comparison of the photochemical properties of trans-DANE with trans-4-dimethylamino-4'-nitrostilbene (DANS) suggests that trans-DANE is a possible fluorescent probe in the non-polar region. 相似文献

A liquid chromatography–electrospray ionization tandem mass spectrometry (LC-ESI-MS/MS) procedure was developed for the simultaneous determination of enantiomers of the prevalent designer drug 3,4-methylenedioxymethamphetamine (MDMA) and its phase I and phase II metabolites in urine with chiral derivatization. The analytes in urine were directly derivatized with chiral Marfey’s reagent, N α- (5-fluoro-2,4-dinitrophenyl)-d-leucinamide, without extraction. The diastereomers of the N α-(2,4-dinitrophenyl)-d-leucinamide derivatives generated were determined by LC-MS/MS. Satisfactory chromatographic separation was achieved for the enantiomers of MDMA and its metabolites 3,4-methylenedioxyamphetamine, 4-hydroxy-3-methoxymethamphetamine (HMMA), HMMA glucuronide, and HMMA sulfate on a semimicro octadecylsilane column using linear gradient elution. With use of multiple reaction monitoring mode, the limits of detection of these analytes ranged from 0.01 to 0.03?μg/mL. Linear calibration curves were obtained for all enantiomers from 0.1 to 20?μg/mL in urine. The method showed sufficient reproducibility and quantitative ability. This is the first report of a simple LC-MS/MS-based analytical procedure with direct chiral derivatization in aqueous media that allows simultaneous enantiomeric determination of drugs and their metabolites, including glucuronide and sulfate derivatives. 相似文献
