The ability of Trichoderma atroviride F-534 to utilize plant waste byproducts derived from processing of vegetables and fruits, as the major source of organic carbon and nitrogen for growth and protease production, was tested. The submerged cultivation of T. atroviride F-534 in the mineral base of the Czapek–Dox medium supplemented with plant waste byproducts resulted into copious biomass formation and was accompanied by secretion of several proteolytic enzymes. Zymography analysis of fungal culture filtrates showed that the high-molecular weight (HMW) protease(s) (from 100 kDa to 230 kDa) represent the major portion of secreted enzymes. Serine-type proteases and metalloproteases were predominant, although all known types of proteolytic enzymes were detected dependent on the type of inducer (substrate). The most conspicuous feature of secreted proteases was that the zymography patterns were unique for each plant material tested. These results confirm our previous finding obtained with purified proteins. Results also suggest that HMW protease(s) may participate in the heterotrophic/saprophytic/mode of life of this fungus. Their identity remains, however, obscure.  相似文献

A fast and highly sensitive high performance liquid chromatographic/ion-trap mass spectrometric method (LC/MS) has been developed for simultaneous determination of ethynylestradiol (EE2), gestodene (GES), levonorgestrel (LNG), cyproterone acetate (CPA) and desogestrel (DES). Among three types of sorbents tested (C8, C18 and phenyl) from two suppliers, the best separation was achieved on reverse phase Zorbax SB-Phenyl column using aqueous methanol as a mobile phase. A linear gradient profile from 70 up to 100% (v/v) in 7th min, kept constant at 100% up to 10th min and followed by a negative gradient to 70% of methanol up to 12th min was used for elution. Applicability of electrospray ionization (ESI) and atmospheric pressure chemical ionization (APCI) and influence of the mobile phase composition, its flow rate, capillary/vaporizer temperature of API source and in-source fragmentor voltage ionization are discussed. The on-column limits of quantification (10 S/N) were 300 pg of EE2, 14 pg of GES and LNG, 4 pg of CPA and 960 pg of DES per injection (1 μL) using APCI with data collection in selected ion monitoring (SIM) mode. The analytical performance of the method was evaluated using the determination of EE2, GES, LNG, CPA and DES in contraceptives and river water samples.  相似文献

N‐(3‐Azidopropyl)vinylsulfonamide was developed as a new bifunctional bioconjugation reagent suitable for the cross‐linking of biomolecules through copper(I)‐catalyzed azide–alkyne cycloaddition and thiol Michael addition reactions under biorthogonal conditions. The reagent is easily clicked to an acetylene‐containing DNA or protein and then reacts with cysteine‐containing peptides or proteins to form covalent cross‐links. Several examples of bioconjugations of ethynyl‐ or octadiynyl‐modified DNA with peptides, p53 protein, or alkyne‐modified human carbonic anhydrase with peptides are given.  相似文献

We report on an in-situ observation of the colloidal silver nanoparticle self-assembly into a close-packed monolayer at the air/water interface followed by a 2D to 3D transition. Using the fast tracking GISAXS technique, we were able to observe the immediate response to the compression of the self-assembled nanoparticle layer at the air/water interface and to identify all relevant intermediate stages including those far from the equilibrium. In particular, a new nonequilibrium phase before the monolayer collapse via the 2D to 3D transition was found that is inaccessible by the competing direct space imaging techniques such as the scanning and transmission electron microscopies due to the high water vapor pressure and surface tension.  相似文献

Cellulose viscose fibres were functionalized by novel amino cellulose sulfates (ACS), namely 6-deoxy-6-(ω-aminoethyl) amino cellulose-2,3(6)-O-sulfate (AECS), and 6-deoxy-6-(2-(bis-N′,N′-(2-aminoethyl)aminoethyl)) amino cellulose-2,3(6)-O-sulfate (BAECS). In this way an amphoteric characteristics were introduced onto cellulose viscose fibers which is extremely important by fiber applications. Whilst cellulose fibers possess only negligible carboxyl groups’ content, the coating of fibers by AECS and BAECS, respectively, introduces new functional groups to the fibers; as positively-charged amino groups and negatively-charged sulfate groups. The typical functional groups within the non-coated fibers, as well in the ACS-coated fibers, were characterized by means of X-ray photoelectron spectroscopy, conductometric-, potentiometric and polyelectrolyte titrations, as well as conventionally by the spectroscopic methylene-blue method. The electro-kinetic behavior was evaluated by measuring the zeta-potential of the fibers as a function of pH. The amounts of the positive-charges (introduced protonated amino groups) determined by potentiometric titration agreed with the amounts of the positive charges determined by conductometric titration. The total amounts of negatively-charged fiber groups (sulfate and carboxyl) determined by polyelectrolyte titration were 38.8 and 32.1 mMol kg?1 for AECS-Vis and BAECS-Vis, respectively, and these results were in accordance with the conventional methylene-blue method.  相似文献

This paper reports development of the explicitly correlated variant of Mukherjee's state specific multireference coupled cluster method (MkCC-F12). The current implementation is restricted to conventional single and double excitations and to pseudo-double excitations related to the Slater Type Geminal (STG) correlation factor using the SP ansatz. The performance of the MkCCSD-F12 was tested on calculations of singlet methylene, dissociation curve of the fluorine molecule, and the BeH(2) insertion pathway. As expected, the results of the newly developed method reconfirm the significantly faster convergence with respect to the basis set limit compared to the traditional expansion in Slater determinants. Results prove that treating the correlation factor separately for each reference is appropriate.  相似文献

Polypyrrole is one of the most frequently studied conducting polymers, having high electrical conductivity and stability, suitable for multi-functionalised applications. Coatings of chemically synthesised polypyrrole applied onto various organic and inorganic materials, such as polymer particles and films, nanoparticles of metal oxides, clay minerals, and carbon nanotubes are reviewed in this paper. Its primary subject is the formation of new materials and their application in which chemical oxidative polymerisation of pyrrole was used. These combined materials are used in antistatic applications, such as anti-corrosion coating, radiation-shielding, but also as new categories of sensors, batteries, and components for organic electronics are created by coating substrates with conducting polymer layers or imprinting technologies.  相似文献
