The skin acts mainly as a protective barrier from the external environment, thanks to the stratum corneum which is the outermost layer of the skin. As in vitro tests on skin are essential to elaborate new drugs, the development of skin models closer to reality becomes essential. It is now possible to produce in vitro human skin substitutes through tissue engineering by using the self-assembly method developed by the Laboratoire d’Organogénèse Expérimentale. In the present work, infrared microspectroscopy imaging analyses were performed to get in-depth morpho-spectral characterization of the three characteristic layers of human skin substitutes and normal human skin, namely the stratum corneum, living epidermis, and dermis. An infrared spectral analysis of the skin is a powerful tool to gain information on the order and conformation of the lipid chains and the secondary structure of proteins. On one hand, the symmetric stretching mode of the lipid methylene groups (2,850 cm?1) is sensitive to the acyl chain conformational order. The evolution profile of the frequency of this vibrational mode throughout the epidermis suggests that lipids in the stratum corneum are more ordered than those in the living epidermis. On the other hand, the frequencies of the infrared components underneath the envelop of the amide I band provide information about the overall protein conformation. The analysis of this mode establishes that the proteins essentially adopt an α-helix conformation in the epidermis, probably associated with the presence of keratin, while modifications of the protein content are observed in the dermis (extracellular matrix made of collagen). Finally, the lipid organization, as well as the protein composition in the different layers, is similar for human skin substitutes and normal human skin, confirming that the substitutes reproduce essential features of real skin and are appropriate biomimetics.  相似文献

White‐light‐emitting materials and devices have attracted enormous interest because of their great potential for various lighting applications. We herein describe the light‐emitting properties of a series of new difunctional organic molecules of remarkably simple structure consisting of two terminal 4‐pyridone push–pull subunits separated by a polymethylene chain. They were found to emit almost “pure” white light as a single organic compound in the solid state, as well as when incorporated in a polymer film. To the best of our knowledge, they are the simplest white‐light‐emitting organic molecules reported to date.  相似文献

The polymerization kinetics and mechanisms of model compounds (mono- and difunctional) characteristic of prepolymers of ATR (acetylene-terminated resins) have been studied by HPLC, FTIR, UV, NMR (liquid and solid) and HPLC in the temperature range of 150–220°C. Using monofunctional compounds, we were able to show that the thermal polymerization of acetylene functions is not a simple reaction. Initially, the majority of compounds formed have a low polymerization degree, followed by the formation of species with higher and progressive polymerization degrees. Spectroscopic techniques such as NMR and FTIR lead to the unambiguous determination of reaction conversion up to 95% in the case of bifunctional prepolymers. The understanding of network structures, on the other hand, is more delicate in polyaromatic systems, since the chains formed are very similar to the basic skeleton of the prepolymer. The kinetic curves obtained isothermally between 150–220°C have shown that while kinetic monitoring of the disappearance of the first 90% of ethynyl functions poses no particular problems, there is a considerable uncertainty for the last 10%, regardless of the technique chosen. We have demonstrated that me thermal properties are dependent on the molecular weight and the structure of the polymer.  相似文献

Fibrin-Type I collagen composite gels have been widely studied as biomaterials, in which both networks are usually formed simultaneously at a neutral pH. Here, we describe a new protocol in which mixed concentrated solutions of collagen and fibrinogen were first incubated at acidic pH to induce fibrinogen gel formation, followed by a pH change to neutral inducing collagen fiber formation. Thrombin was then added to form fibrin-collagen networks. Using this protocol, mixed gels containing 20 mg.mL−1 fibrin and up to 10 mg.mL−1 collagen could be prepared. Macroscopic observations evidenced that increasing the content of collagen increases the turbidity of the gels and decreases their shrinkage during the fibrinogen-to-fibrin conversion. The presence of collagen had a minor influence on the rheological properties of the gels. Electron microscopy allowed for observation of collagen fibers within the fibrin network. 2D cultures of C2C12 myoblasts on mixed gels revealed that the presence of collagen favors proliferation and local alignment of the cells. However, it interferes with cell differentiation and myotube formation, suggesting that further control of in-gel collagen self-assembly is required to elaborate fully functional biomaterials.  相似文献

Three imine‐based metal complexes, having no overlap in terms of their compositions, have been simultaneously generated from the self‐sorting of a constitutional dynamic library (CDL) containing three amines, three aldehydes, and three metal salts. The hierarchical ordering of the stability of the three metal complexes assembled and the leveraging of the antagonistic and agonistic relationships existing between the constituents within the constitutional dynamic network corresponding to the CDL were pivotal in achieving the sorting. Examination of the process by NMR spectroscopy showed that the self‐sorting of the FeII and ZnII complexes depended on an interplay between the thermodynamic driving forces and a kinetic trap involved in their assembly. These results also exemplify the concept of “simplexity”—the fact that the output of a self‐assembling system may be simplified by increasing its initial compositional complexity—as the two complexes could self‐sort only in the presence of the third pair of organic components, those of the CuI complex.  相似文献

Geometric phase may enable inherently fault-tolerant quantum computation. However, due to potential decoherence effects, it is important to understand how such phases arise for mixed input states. We report the first experiment to measure mixed-state geometric phases in optics, using a Mach-Zehnder interferometer, and polarization mixed states that are produced in two different ways: decohering pure states with birefringent elements; and producing a nonmaximally entangled state of two photons and tracing over one of them, a form of remote state preparation.  相似文献
