The method of preparing protein mixtures for electrophoretic analysis of membrane-associated cell proteins was improved. By sonication, about one-half of the proteins of thyroid cells were released into the supernatant, while the other half preferentially comprising membrane proteins still remained in cell fragments, which could be sedimented by centrifugation. After sonication, even those proteins which remained in cell fragments, could completely be dissolved by free-flow isoelectric focusing media. They migrated through the free-flow electrophoresis chamber without forming precipitates. Because of these improvements, it was possible to show that the two thyroid cancer cell lines ML-1 and ONCO-DG1 express cytokeratin 8 at similar rates, but cytokeratins 7 and 18 differently. In addition, the presence of inorganic pyrophosphatase, tubulin-beta-5, and tubulin-beta-1 chains in human thyroid cells was proved for the first time. 相似文献

Although the chemistry of elusive tricyanomethane (cyanoform) has been studied during a period of more than 150 years, this compound has very rarely been utilized in the synthesis or modification of heterocycles. Three-membered heterocycles, such as epoxides, thiirane, aziridines, or 2H-azirines, are now treated with tricyanomethane, which is generated in situ by heating azidomethylidene-malonodinitrile in tetrahydrofuran at 45 °C or by adding sulfuric acid to potassium tricyanomethanide. This leads to ring expansion with formation of 2-(dicyanomethylidene)oxazolidine derivatives or creation of the corresponding thiazolidine, imidazolidine, or imidazoline compounds and opens up a new access to these push–pull-substituted olefinic products. The regio- and stereochemistry of the ring-enlargement processes are discussed, and the proposed reaction mechanisms were confirmed by using 15N-labeled substrates. It turns out that different mechanisms are operating; however, tricyanomethanide is always acting as a nitrogen-centered nucleophile, which is quite unusual if compared to other reactions of this species. 相似文献

Experimental Fourier-transform infrared spectra and DFT calculated infrared spectra are compared to investigate the effect of adsorbed nitrogen on the OH-stretch band complex of water clusters. Using a collisional cooling experiment, pure as well as partially and completely N(2)-covered water clusters consisting of 20-200 water molecules have been generated in thermal equilibrium in the aerosol phase within the temperature range of 5-80 K. Computational IR-spectra simulations have been performed for discrete pure and N(2)-covered water clusters including 10, 15, 20, and 30 water molecules. The adsorbed N(2) molecules especially affect the three-coordinated water molecules at the cluster surface which could be observed as a blue shift of the companion O-H band at 2900 cm(-1) and a red shift of the dangling O-H band at 3700 cm(-1) by about 20 cm(-1) in both cases. The most striking effect of the N(2) adsorbate is an intensity increase of the dangling O-H band by a factor of 3-5. Furthermore, the onset temperature of nitrogen adsorption at the water cluster surface was experimentally found to be roughly 30 K for cluster sizes of about 100 water molecules. Experimental and computational results are in good agreement. The presented results are based on and support the work of V. Buch, J. P. Devlin, and co-workers (e.g., J. Phys. Chem. B, 1997; J. Phys. Nach dem Meßverfahren von Signer können der Auslöschwinkel und die Stärke der Doppelbrechung aus der Kompensationskurve, die eine Sinusschwingung vorstellt, abgelesen werden. Durch fortlaufende Registrierung dieser Kurve können Veränderungen des Auslöschwinkels, der Auskunft über das Längen-Breitenverhältnis der Teilchen gibt, und die Stärke der Doppelbrechung, die von der Teilchenorientierung und bei konstantem Längen-Breitenverhältnis außerdem von der Teilchendicke abhängt, nachgewiesen werden.Bei den untersuchten Lösungen von Karnaubawachs und Kongorot zeigen die geströmten Teilchen ein deutliches Wachstum, das nach den Beobachtungsdaten nicht zu stäbigen, sondern eher zu flächigen bis massigen Sekundärteilchen führt. 相似文献

In the majority of the literature describing green coloured materials used on ancient painting layers (15th or 16th century), two copper greens are mainly cited: malachite [CuCOr3 x Cu(OH)2] and verdigris [Cu(CH3COO)2 x [Cu(OH)2]3 x 2H2O]. It is shown, by micro-Raman spectroscopy, that the artists were actually employing more than these two copper greens, in particular various copper sulfates, among which the most common pigment found is posnjakite [CuSO4 x 3Cu(OH)2 x H2O]. In contrast to the PIXE (particle induced X-ray emission) technique, Raman spectroscopy is a technique of choice, able to distinguish not only a copper sulfate from a carbonate or acetate but also the different sulfates themselves; in this respect, we found that the high wavenumber region (2800-4000 cm(-1)), characteristic of H2O vibrations, is of particular interest. It is also shown that numerous green areas were created with mixtures of a copper sulfate mixed with other pigments, for instance to enhance the colour depth. Finally, in some cases, no green pigment is actually employed but the colour is obtained by intimately mixing yellow and blue pigments. All these results led to a new look at the pigments which were in use on the palettes of the ancient artists. 相似文献
