Terminal phosphate groups on double-stranded DNA probes bind strongly to glass substrates coated with a zirconium phosphonate monolayer, and probes immobilized in this way as microarrays can be used to detect protein targets. The sensitivity of the microarray was shown to be enhanced by the use of a polyguanine segment ((G)n , n > or = 5) as a spacer between the phosphate linker and the protein interaction domain. More importantly, the presence of phosphate linkers on both ends of the dsDNA probes leads to significant enhancement of target capture. The relevant characteristics of the different probes when bound to the surface were determined, by the original use of a combination of surface characterization techniques (XPS, AFM, and Sarfus). In this context, the location of the phosphate linkers in the duplex probes was found to result in different probe surface coverage and presentation on the surface, which affect subsequent interactions with the target protein.  相似文献

The numerical results for the twist angle profile xi(z) across a slab-shaped nematic cell obtained from a density functional theory (DFT) are compared to the predictions of the macroscopic Frank-Oseen theory. The latter theory predicts that xi"(z)=0, and this is also seen to be the case for the DFT results. These do, however, verify the Frank-Oseen relation, lambda+/-W+/-=K2, between the de Gennes extrapolation length (lambda+/-), the anchoring energy per unit area of the (+/-) cell wall (W+/-), and the elastic constant of the nematic for twist deformations (K2), only if W+/- is nonlinearly related to the amplitude of the anchoring term of the DFT.  相似文献

Cyclopenta-, benzo-, and cyclohepta[b]-annulated azepane scaffolds were prepared in two steps from optically active cyclic α-allyl-β-oxoesters. The first step was ruthenium-catalyzed olefin cross metathesis with acrylonitrile. The second step was palladium-catalyzed dihydrogenation which consists of three consecutive processes: The hydrogenation of the C−C double and C−N triple bonds was followed by the reductive amination via the iminium ion formed in situ from the primary amino function and the endocyclic carbonyl group. This last step gave, stereoselectively, the annulated azepanes with relative trans-configuration. The amino function and the ester group define two points for further diversification of the scaffolds. The trifluoroacetyl derivatives allowed to establish the enantiopurity of the products to be 97–98 % ee by GLC on a chiral phase. The relative trans-configurations and in one case also the absolute (R,R)-configuration was established by X-ray crystallography.  相似文献

The synthesis of a bicyclic germylene from the reaction of a germole dianion with hafnocene dichloride is reported. This germylene is stabilized by a homoconjugative interaction of the dicoordinated germanium atom with a remote C=C double bond. First reactivity studies revealed its nucleophilic character and resulted in the synthesis of bimetallic hafnium/iron and hafnium/tungsten complexes with a germylene group linker.  相似文献

The study describes bioactive compounds as inhibitors of acetylcholinesterase (AChE), from the stem bark extract of Montrouziera cauliflora, selected among 19 dichloromethane extracts from Clusiaceae species. Our work focused on the development of an original normal phase HPLC microfractionation strategy to rapidly assess highly active zones from this crude active non-polar plant extract. Two different microfraction collection methods were evaluated for the assessment of the AChE inhibition. Two guttiferones and a tocotrienol were directly isolated among five compounds identified off-line by NMR after upscaling the fractionation and their AChE inhibition was evaluated. The strengths and weaknesses of the two microfractionation collection methods for HPLC-AChE activity-based profiling are discussed.  相似文献

We establish a “preparatory Sard theorem” for smooth functions with a partially affine structure. By means of this result, we improve a previous result of Rifford [17, 19] concerning the generalized (Clarke) critical values of Lipschitz functions defined as minima of smooth functions. We also establish a nonsmooth Sard theorem for the class of Lipschitz functions from Rd to Rp that can be expressed as finite selections of Ck functions (more generally, continuous selections over a compact countable set). This recovers readily the classical Sard theorem and extends a previous result of Barbet–Daniilidis–Dambrine [1] to the case p > 1. Applications in semi-infinite and Pareto optimization are given.  相似文献
