In the present work we have modeled the Michaelis complex of the cyclic-Adenosine Monophosphate Dependent (cAMD) Protein Kinase A (PKA) with Mg(2)ATP and the heptapeptide substrate Kemptide by classical molecular dynamics. The chosen synthetic substrate is relevant for its high efficiency and small size, and it has not been used in previous theoretical studies. The structural analysis of the data generated along the 6 ns simulation indicates that the modeled substrate-enzyme complex mimics the substrate binding pattern known for PKA. The values of the average prereactive distances obtained from the simulation do not exclude any of the two limiting situations proposed as mechanisms in the literature for the phosphorylation reaction (dissociative and associative) because the system oscillates between configurations compatible with each of them. Furthermore, the results obtained for the average interaction distances between active site residues concord in suggesting the plausibility of an alternative third reaction mechanism.  相似文献

The complexes [Ni(eta(3)-CH(2)CHCH(2))Br(kappa(1)P-PR(2)CH(2)CH=CH(2))] (R = Ph 1, (i)Pr2 ) and [Ni(eta(3)-CH(2)C(R')CH(2))(kappa(1)P-PR(2)CH(2)CH=CH(2))(2)][BAr'(4)] (R' = H, R = Ph 4a, R = (i)Pr 4b; R' = CH(3), R = Ph 5a, R = (i)Pr 5b; Ar' = 3,5-C(6)H(3)(CF(3))(2)) have been prepared and characterized. The X-ray crystal structures of 1, 2 and 5b have been determined. 4a-b and 5a-b are catalyst precursors for the oligomerization of RC(6)H(4)CH=CH(2) to oligostyrene (R = H) or oligo(4-methylstyrene) (R = CH(3)) respectively, without the need of a co-catalyst such as methylalumoxane. The catalytic activities range from moderate to high. The oligomerization reactions are carried out in the temperature interval 25-40 degrees C in 1,2-dichloroethane, using an olefin/catalyst ratio equal to 200, yielding oligostyrenes with a high isotactic fraction content P(m), with M(n) in the range 700-1900 Dalton, and polydispersities between 1.22 and 1.64. The cationic complexes 4a-b and 5a-b are also effective catalyst precursors for the hydrosilylation reactions of styrene or 4-methylstyrene with PhSiH(3) in 1,2-dichloroethane at 40 degrees C using an olefin/catalyst ratio equal to 100, leading selectively to RC(6)H(4)CH(SiH(2)Ph)CH(3) (R = H, CH(3)) in 50-79% yield.  相似文献

The standard (po = 0.1 MPa) molar enthalpies of formation in the condensed phase of seven isomers of fluoromethylaniline were derived from the standard molar energies of combustion, in oxygen, to yield CO2(g), N2(g) and HF.10H2O(l), at T = 298.15 K, measured by rotating bomb combustion calorimetry. The standard molar enthalpies of vaporization or sublimation of these compounds, also at T = 298.15 K, were determined using Calvet microcalorimetry, while the enthalpies of fusion of the solid compounds were determined by differential scanning calorimetry. The standard molar enthalpies of formation in the gaseous phase, at T = 298.15 K, were derived from the former two experimental quantities. G3MP2//B3LYP calculations were performed for all possible fluoromethylanilines allowing the estimation of data for the isomers that were not studied experimentally. The Cox scheme was applied with two different approaches for the estimation of the standard molar enthalpies of formation of all the isomers studied, and this led to the conclusion that the literature values for the enthalpies of formation of the meta and para isomers of methylaniline seem to be not reliable. Further G3MP2//B3LYPs calculations on the methylaniline isomers yielded new values for the standard molar enthalpies of formation of the isomers of methylaniline, which have been tested under the Cox scheme, resulting in better estimates.  相似文献

We present the first trans-H-bond 13C-15N 3-bond J couplings calculated from entire neutral and protonated alpha-helical polyalanines. The neutral helices considered are those of the capped peptides, acetyl(Ala)NNH2, where N = 8, 16, 17, and 18, while the protonated peptides are the uncapped (Ala)17 protonated at three different positions. The calculated J values correlate well with O...H distances and somewhat less well with N...O distances, particularly if the terminal H-bonds are eliminated from the correlation. The J values calculated using the entire helix are about 6% lower in magnitude than those recently reported for H-bonding chains whose geometries were extracted from the same helices. Aqueous solvation favors protonation of the alpha-helix on the terminal COOH. Experimental measurements of the trans-H-bond 13C-15N 3-bond J couplings in acidic solution should be interpreted with this context.  相似文献

A new protein phosphatase inhibitor, 19-epi-okadaic acid, was isolated from the marine dinoflagellate Prorocentrum belizeanum. Its structure and conformation in solution has been determined, and important differences were found when compared with the lead compound okadaic acid. The new metabolite showed nanomolar activities, and its selectivity for PP2A versus PP1 surpasses that shown by okadaic acid 10-fold, making it one of the most selective inhibitors of this class.  相似文献

We consider a class of kinetic equations, equipped with a single conservation law, which generate -contractions. We discuss the hydrodynamic limit to a scalar conservation law and the diffusive limit to a (possibly) degenerate parabolic equation. The limits are obtained in the ``dissipative' sense, equivalent to the notion of entropy solutions for conservation laws, which permits the use of the perturbed test function method and allows for simple proofs. A general compactness framework is obtained for the diffusive scaling in . The radiative transport equations, satisfied by the Wigner function for random acoustic waves, present such a kinetic model that is endowed with conservation of energy. The general theory is used to validate the diffusive approximation of the radiative transport equation.

Design of miniature and light cameras requires an optical design breakthrough to achieve good optical performance. Solutions inspired by animals' eyes are the most promising. The curvature of the retina offers several advantages, such as uniform intensity and no field curvature, but this feature is not used. The work presented here is a solution to spherically bend monolithic IR detectors. Compared to state-of-the-art methods, a higher fill factor is obtained and the device fabrication process is not modified. We made an IR eye camera with a single lens and a curved IR bolometer. Images captured are well resolved and have good contrast, and the modulation transfer function shows better quality when comparing with planar systems.  相似文献

Our recent work on synthesis and application of thermally gelling nanoparticle dispersions is briefly reviewed here. These nanoparticles consist of interpenetrating polymer networks (IPN) of poly-acrylic acid (PAAc) and poly(N-isopropylacrylamide) (PNIPAM). The aqueous IPN nanoparticle dispersions with polymer concentrations above 2.5 wt % underwent an inverse thermoreversible gelation at about 33 °C. Dextran markers of various molecular weights as model macromolecular pseudodrugs were mixed with the IPN nanoparticle dispersion at room temperature. At body temperature, the dispersion became a gel. The dextran release profiles were then measured using UV-visible spectroscopy. The biocompatibility of this nanoparticle assembly was assessed using an animal implantation model.  相似文献

Solution studies to elucidate the coordination behaviour and the electrochemical response of the ferrocene-functionalized polyazamacrocycle 1,4,7,10,13,16-hexa(ferrocenylmethyl)-1,4,7,10,13,16-hexaazacyclooctadecane (L1) by potentiometric methods and electrochemical techniques have been carried out. Potentiometric methods in the presence of Cd2+, Hg2+, Pb2+ and Zn2+ were carried out in 1,4-dioxane/water (70:30 v/v, 25°C, 0.1 mol dm−3 KNO3). Electrochemical studies were carried out in acetonitrile/dichloromethane (50:50 v/v, 25°C, 0.1 mol dm−3 TBAClO4) in the presence of transition metal ions and anions.  相似文献

Peters and Pennings introduced and studied in [9] and [11] a kind of extensions of dynamical systems induced by certain C*-algebras, which they calltame extensions. In this paper we study the behaviour of metrizable tame extensions when some of the most important dynamical concepts related to the metric (such as sources, sinks, saddles, the shadowing property and distallity) are considered. We will confirm that these extensions can be troublesome in this context. We show, however, that sources and sinks are preserved under certain conditions. Entrata in Redazione il 24 febbraio 1997.
