Platinum metallodrug-protein binding studies by capillary electrophoresis-inductively coupled plasma-mass spectrometry: characterization of interactions between Pt(II) complexes and human serum albumin

Characterizing how platinum metallocomplexes bind to human serum albumin (HSA) is essential in evaluating anticancer drug candidates. Using cisplatin as a reference complex, the application of capillary electrophoresis (CE) to reliably assess drug/HSA interactions was validated. Since this complex is small compared to the size of the protein, the binding response could only be recognized when applying CE coupled to a (platinum) metal-specific mode of detection, namely inductively coupled plasma-mass spectrometry (ICP-MS). This coupling allowed for confirmation of a specific affinity of cisplatin and novel Pt complexes to HSA, measurement of the kinetics of binding reactions, and determination of the number of drug molecules attached to the protein. As the cisplatin/HSA molar ratio increased, the reaction rate became faster with a maximum on the kinetic curve appearing at about 50 h of incubation at 20 times excess of cisplatin. The reaction was characterized as a pseudo-first order reaction with the rate constant k = 0.003 min(-1) at 37 degrees C. When incubated with a 20-fold excess of cisplatin, HSA bound up to 10 mol of Pt per mol of the protein. This is indicative for a strong metal-protein coordination occurring at several HSA sites other than the only protein cysteine residue. Structural analogs of cisplatin, bearing aminoalcohol ligands, showed comparable protein binding reactivity and stoichiometry but a common equilibrium was not reached even after one week of incubation. Also apparent was a two-step mechanism of the binding reaction. Results demonstrated the suitability of CE-ICP-MS as a rapid assay for high-throughput studying of drug/HSA interactions.     

Structural assignment of organotin hydrides containing the oxazoline ligand

A series of triorganotin hydrides and diorganotin dihydrides containing the optically active 2-(4-isopropyl-2-oxazolinyl)-5-phenyl ligand have been characterized by means of the multinuclear low-temperature NMR investigations, the results of which are discussed. In the corresponding organotin hydrides values of the ¹J(¹H-^117/119Sn) couplings appeared to be temperature dependent, supporting an axial/equatorial position of the hydrogen attached to the tin.     

Prediction of adsorption of organic component and water vapour mixture onto activated carbon

A simple isotherm equation is derived for the adsorption of an organic component onto activated carbon in presence of water vapour. The theoretical results are compared with experimental data for toluene-water vapour-activated carbon, which were published byRipperger andGermerdonk [10].

Application of divergent multi-component reactions in the synthesis of a library of peptidomimetics based on γ-amino-α,β-cyclopropyl acids

The multi-component condensation of organozirconocene, aldimine and zinc carbenoid was applied to the stereoselective synthesis of cyclopropane amino acid derivatives. These compounds served as scaffolds for the preparation of a 46-member library. The C- and N-termini of the cyclopropane amino acid derivatives were diversified by condensations with ten amines and ten acylating agents, respectively. To improve yields and accelerate library synthesis, most products were prepared under microwave irradiation and purified by polymer-bound scavengers and SPE methodology. All compounds were analyzed by LC-MS and a representative selection was fully characterized.     

An analysis of mean life and lifetime of unstable elementary particles

A theoretical analysis of the concept of lifetime and mean life of unstable elementary particles is presented. New analytic formulas for lifetime and mean life as a function of decay width and the mass of unstable particle are derived for Breit-Wigner and Matthews-Salam energy distributions. It is demonstrated that, for unstable particles with a larger width or decay energy threshold, the deviation from the generally accepted mean life ^m = ^–1 is significant. The behavior of the decay law P(t) for small times is analyzed, and it is shown that the Breit-Wigner distribution violates the condition P(t = 0) = 0, whereas the Matthews-Salam distribution satisfies it.     

Duality transformations in electrodynamics

Electrodynamics admitting a duality transformation group is considered. For such an electrodynamics an extension of the classical Rainich-Misner-Wheeler theory is presented.On leave of absence from the University of Warsaw, Warsaw, Poland.     

Ergodicity of eigenfunctions for ergodic billiards

We give a simple proof of ergodicity of eigenfunctions of the Laplacian with Dirichlet boundary conditions on compact Riemannian manifolds with piecewise smooth boundaries and ergodic billiards. Examples include the Bunimovich stadium, the Sinai billiard and the generic polygonal billiard tables of Kerckhoff, Masur and Smillie.     

Further studies on the chemistry and structure of n-oxides of sparteine and its derivatives—V: Synthesis and structure of 2-phenylsparteine-n16-oxide,a new case of “sponge” proton

The synthesis of 2-phenylsparteine-N₁₆-oxide (7) and its perchlorate salt (7-H⁺) was carried out. On the basis of spectral data, and by comparison with appropriate sparteine-N-oxides, the mechanism of formation and the structures of the two new compounds were proposed. It was found, the basicity of the new N-oxide is unexpectedly high and comparable to the basicity of quaternary ammonium hydroxides. The structure and the strength of intramolecular H-bond in 7-H⁺ makes 7 an excellent “catcher” proton or specific ”sponge” proton.     

Alkoxy/siloxy group exchange in the system vinyltrialkoxysilane-iridium(I) siloxide complex

A study of reactions of dimeric siloxide iridium complex, [[(cod)Ir(mu-OSiMe3)]2] (1) with vinyltriethoxysilane and vinyltrimethoxysilane has revealed a new type of the reation--alkoxy group transfer from silicon to iridium with a simultaneous transfer of a siloxy group from iridium to silicon--as a result of which vinyldialkoxytrimethyldisiloxane and dimeric alkoxide iridium complex [[(cod)Ir(mu-OR)]2] (3) are formed. The structure of [[(cod)Ir(mu-OEt)]2] (3a) has been solved by X-ray diffraction.
