Affinity and ion exchange conventional chromatography have been used to capture erythropoietin (EPO) from mammalian cell culture supernatant. Currently, chromatographic adsorbent perfusion is available, however a limited number of applications have been found in the literature. In this work, three anion exchange chromatographic supports (gel, membrane and monolithic) were evaluated in the capture step of the recombinant erythropoietin purification process. The influences of load and flow rate on each support performance were analyzed. Also the purity of the EPO molecules was determined. A productivity analysis, as a decision tool for larger scale implementation, was done. As a conclusion, the evaluated supports are technically suitable to capture EPO with adequate recovery and good purity. However, the monolithic column admits high operating velocity, showing the highest adsorption capacity and productivity. 相似文献

The functionalization of magnetite (Fe3O4) nanoparticles with dopamine‐derived clickable biomimetic anchors is reported. Herein, an alkyne‐modified catechol‐derivative is employed as the anchor, as i) the catechol‐functional anchor groups possess irreversible covalent binding affinity to Fe3O4 nanoparticles, and ii) the alkyne terminus enables further functionalization of the nanoparticles by the grafting‐onto approach with various possibilities offered by ‘click’ chemistry. In the present work, azido‐end group functionalized Rhodamine and poly(ethylene glycol) (PEG) are utilized to coat the iron oxide nanoparticles to make them fluorescent and water soluble.

A counterpart of the Mollow triplet (luminescence line shape of a two-level system under coherent excitation) is obtained for the case of incoherent excitation in a cavity. The system acquires coherence through the strong-coupling between the cavity and the emitter. Analytical expressions, in excellent agreement with numerical results, pinpoint analogies and differences between the conventional resonance fluorescence spectrum and its cavity QED analogue under incoherent excitation. Most notably, the satellites broaden and split sublinearly with increasing incoherent pumping. 相似文献

It has been widely accepted that electric fields favor the ferroelectric phase with parallel electric dipoles over the antiferroelectric phase. With detailed measurements in polycrystalline ceramics of Pb(0.99)Nb(0.02[(Zr(0.57)Sn(0.43)(1-y)Ti(y)](0.98)O(3), we demonstrate in this Letter that electric fields can induce an antiferroelectric phase out of a ferroelectric phase, i.e., trigger an apparently unlikely ferroelectric-to-antiferroelectric phase transition. We suggest that it is caused by the volume contraction from the converse piezoelectric effect at the coercive field with a reversed polarity. 相似文献

Theoretical calculations predict 270Hs (Z=108, N=162) to be a doubly magic deformed nucleus, decaying mainly by alpha-particle emission. In this work, based on a rapid chemical isolation of Hs isotopes produced in the 26Mg+248Cm reaction, we observed 15 genetically linked nuclear decay chains. Four chains were attributed to the new nuclide 270Hs, which decays by alpha-particle emission with Qalpha=9.02+/-0.03 MeV to 266Sg which undergoes spontaneous fission with a half-life of 444(-148)(+444) ms. A production cross section of about 3 pb was measured for 270Hs. Thus, 270Hs is the first nucleus for which experimental nuclear decay properties have become available for comparison with theoretical predictions of the N=162 shell stability. 相似文献

Rosmarinus eriocalyx (rosemary or Elyazir) is an endemic species growing in arid steppe and rocky mountain in the South-West Algeria. This plant is well known in Algeria and Morocco due to its medicinal properties. However, little is known about its phytochemical composition. For this purpose, natural antioxidant compounds from R. eriocalyx were recovered by solid-liquid extraction and characterized by reversed-phase high-performance liquid chromatography coupled to quadrupole-time-of-flight mass spectrometry using negative and positive ionization modes. This analytical methodology enabled the characterization of 101 compounds, which were distributed in five major categories namely hydroxycinnamic acid derivatives, hydroxybenzoic acid derivatives, flavonoids, phenolic diterpenes and phenolic triterpenes. Moreover, the studied extract generally showed free radical-scavenging and reductive abilities in the range of butylated hydroxyanisole, butylated hydroxytoluene, α-tocopherol, and ascorbic acid. Therefore, the result suggests that the aqueous-methanolic extract of R. eriocalyx could serve as a potential source of antioxidants. 相似文献

New antibacterial films are designed with the capability to reversibly regulate their killing and repelling functions in response to variations in environmental pH. These systems consist of porous polystyrene surfaces as the main components and a copolymer bearing pH‐sensitive thiazole and triazole groups as the minor components. These pH‐sensitive groups, located on the surfaces, can be partially protonated at acidic pH levels, increasing the positive charge density of the surfaces and their antibacterial activity. Similarly, their bacterial adhesion and killing efficiencies in response to changes in pH are evaluated by analyzing the bacterial viability of Staphylococcus aureus bacteria on the surfaces under acidic and neutral pH values. It is demonstrated that after only 1 h of incubation with the bacterial suspension in acidic conditions, the surfaces killed the bacteria, while at pH = 7.4, some of the adhered bacteria are removed. Furthermore, the surface topography exerts an important role by intensifying this response. 相似文献

We report on the progress towards the implementation of a double trap system for the coupling of ion clouds stored in different traps through the charges they induce in a common electrode. The two traps have cylindrical symmetry and can be used either as Penning or Paul traps. The laser system for Doppler cooling of 40Ca+ clouds is described. The cooling can be performed differentially in the two traps, aiming at the study of the coupling between two clouds of ions at different temperatures. This setup is the starting point for the realization of energy transfer between two Doppler-cooled ions stored in different traps. We report on the status of the experiment and evaluate the performance required for an efficient coupling. 相似文献

Pediatric obstructive sleep apnea (OSA) is a breathing disorder that alters heart rate variability (HRV) dynamics during sleep. HRV in children is commonly assessed through conventional spectral analysis. However, bispectral analysis provides both linearity and stationarity information and has not been applied to the assessment of HRV in pediatric OSA. Here, this work aimed to assess HRV using bispectral analysis in children with OSA for signal characterization and diagnostic purposes in two large pediatric databases (0–13 years). The first database (training set) was composed of 981 overnight ECG recordings obtained during polysomnography. The second database (test set) was a subset of the Childhood Adenotonsillectomy Trial database (757 children). We characterized three bispectral regions based on the classic HRV frequency ranges (very low frequency: 0–0.04 Hz; low frequency: 0.04–0.15 Hz; and high frequency: 0.15–0.40 Hz), as well as three OSA-specific frequency ranges obtained in recent studies (BW1: 0.001–0.005 Hz; BW2: 0.028–0.074 Hz; BWRes: a subject-adaptive respiratory region). In each region, up to 14 bispectral features were computed. The fast correlation-based filter was applied to the features obtained from the classic and OSA-specific regions, showing complementary information regarding OSA alterations in HRV. This information was then used to train multi-layer perceptron (MLP) neural networks aimed at automatically detecting pediatric OSA using three clinically defined severity classifiers. Both classic and OSA-specific MLP models showed high and similar accuracy (Acc) and areas under the receiver operating characteristic curve (AUCs) for moderate (classic regions: Acc = 81.0%, AUC = 0.774; OSA-specific regions: Acc = 81.0%, AUC = 0.791) and severe (classic regions: Acc = 91.7%, AUC = 0.847; OSA-specific regions: Acc = 89.3%, AUC = 0.841) OSA levels. Thus, the current findings highlight the usefulness of bispectral analysis on HRV to characterize and diagnose pediatric OSA. 相似文献
