Fucosylated oligosaccharides, such as 2′-fucosyllactose in human milk, have important biological functions such as prebiotics and preventing infection. In this work, the effect of an acceptor substrate (lactose) and the donor substrate 4-nitrophenyl-α-l-fucopyranoside (pNP-Fuc) on the synthesis of a fucosylated trisaccharide was studied in a transglycosylation reaction using α-l-fucosidase from Thermotoga maritima. Conducting a matrix-assisted laser desorption ionization time-of-flight mass spectrometry (MALDI-TOF MS), it was demonstrated that synthesized oligosaccharide corresponded to a fucosylated trisaccharide, and high-performance liquid chromatography (HPLC) of the hydrolyzed compound confirmed it was fucosyllactose. As the concentration of the acceptor substrate increased, the concentration and synthesis rate of the fucosylated trisaccharide also increased, and the highest concentration obtained was 0.883 mM (25.2% yield) when using the higher initial lactose concentration (584 mM). Furthermore, the lower donor/acceptor ratio had the highest synthesis, so at the molar ratio of 0.001, a concentration of 0.286 mM was obtained (32.5% yield).  相似文献

Two kilns, one of which collapsed during firing cycle together with its entire pottery load, have been excavated at the Egnazia site in Southern Italy. To understand the reason for the collapse, ‘Broad Line’ typology pottery finds were analysed by complementary analytical techniques. Analytical results not only suggest as cause of collapse sudden overheating in kiln due to uncontrolled increases in temperature, but also indicate a good technological cycle from the recovery of raw materials to the manufacturing and firing process, which tends to disprove the common assumption of non-professional production.  相似文献

Dispersions of a bisphenol A‐based epoxy resin with an organophilic montmorillonite (Nanofil 919) were studied by X‐ray diffraction and oscillatory shear rheometry. X‐ray studies reveal that the clay is intercalated by the epoxy and forms stable dispersions. The viscoelastic behavior of the nanodispersions was measured as a function of the Nanofil concentration and temperature. An increase in both G′ and G″ moduli was detected as the concentration increases. Furthermore, a transition from a liquid‐like behavior, at low temperatures, to a solid‐like behavior, at higher temperatures, was observed for all the samples. This transition is accounted for the formation of a percolated structure of interconnected tactoids through hydrophobic interactions. © 2008 Wiley Periodicals, Inc. Rotationally resolved pulsed-field-ionization zero-kinetic-energy photoelectron spectra of the X 1/2, A(1) 3/2, and A(2) 1/2 electronic states of the ArXe(+) molecular ion have been recorded following resonant (1+1(')) two-photon excitation via selected rovibrational levels of the C 1 and D 0(+) states of selected isotopomers of the ArXe molecule. Using rovibronic selection and propensity rules for the photoionization out of these intermediate molecular states enabled the determination of the parity of the molecular-ion levels and of the magnitude and sign of the Ω-doubling constants of the coupled X 1/2 (p ≈ 4B) and A(2) 1/2 (p ≈ -2B) states of ArXe(+). The results indicate that these molecular-ion states can be approximately described using Mulliken's second variant of Hund's angular momentum coupling case (c), for which J(a), the total electronic and spin angular momentum of the two atoms, is a good quantum number (semi-united atom). The analysis of the rotational structure enabled the derivation of improved values of the dissociation energies, equilibrium distances, and molecular constants for the X 1/2, A(1) 3/2, and A(2) 1/2 states of ArXe(+).  相似文献

The efficiency of headspace solid-phase microextraction (SPME) was evaluated for the qualitative and semi-quantitative analysis of virgin olive oil volatile compounds. The behaviour of four fibre coatings was compared for sensitivity, repeatability and linearity of response. A divinylbenzene-Carboxen-polydimethylsiloxane fibre coating was found to be the most suitable for the analysis of virgin olive oil volatiles. Sampling and chromatographic conditions were examined and the SPME method, coupled to GC with MS and flame ionization detection, was applied to virgin olive oil samples. More than 100 compounds were isolated and characterised. The presence of some of these compounds in virgin olive oil has not previously been reported. The main volatile compounds present in the oil samples were determined quantitatively.  相似文献

The Point-Feature Cartographic Label Placement (PFCLP) problem consists of placing text labels to point features on a map avoiding overlaps to improve map visualization. This paper presents a Clustering Search (CS) metaheuristic as a new alternative to solve the PFCLP problem. Computational experiments were performed over sets of instances with up to 13,206 points. These instances are the same used in several recent and important researches about the PFCLP problem. The results enhance the potential of CS by finding optimal solutions (proven in previous works) and improving the best-known solutions for instances whose optimal solutions are unknown so far.  相似文献

Self-assembling of poly(allylamine) containing an osmium polypyridil complex (PAOs) alternatively with sodium dodecyl sulfate (SDS) and glucose oxidase (GOx) generates a ternary multilayer system, (PAOs/SDS/PAOs/GOx)n. The introduction of this anionic surfactant allows a sensitive increase of the polyelectrolyte and the enzyme uptake at pH 7.0, enhancing its catalytic behavior in presence of glucose more than 5 times, compared to the system (PAOs/GOx)n constructed at the same pH. The balance between ionic and hydrophobic interactions in the construction of this system is discussed.  相似文献

Photodynamic therapy (PDT) may cause tumour cell destruction by direct toxicity or by inducing microcirculatory shutdown. Protoporphyrin IX generated from 5-aminolevulinic acid (ALA) has been widely used as an endogenous photosensitiser in PDT. However, the hydrophilic nature of the ALA molecule limits its penetration through the stratum corneum of the skin and cell membranes and thus, ALA alkyl-esters have been developed to improve ALA permeation.The aim of this work was to study Protoporphyrin IX synthesis from ALA and its derivatives ALA methyl ester (Me-ALA) and ALA hexyl ester (He-ALA) in the microvascular endothelial cell line HMEC-1 derived from normal skin, and to evaluate their response to PDT.We found that lower light doses are required to photosensitise HMEC-1 endothelial cells than to photosensitise PAM212 transformed keratinocytes, showing some possible selectivity of ALA-PDT for vascularisation in skin.Employed at concentrations leading to equal Protoporphyrin IX synthesis, ALA, He-ALA and Me-ALA presented the same efficacy of HMEC-1 photosensitisation. However, He-ALA was a promising compound for the use in the enhancement of Protoporphyrin IX in HMEC-1 cells employed at low concentrations at both short and long time exposures whereas Me-ALA should be employed at high concentrations and longer time periods in order to surpass the Protoporphyrin IX levels obtained with ALA. The advantage of Me-ALA over ALA was based on its lower dark toxicity.This is the first work to report vascular cell photosensitisation employing alkyl-esters of ALA, and we demonstrated that these derivatives could exert the same effect as ALA and under certain conditions enhance photosensitisation of vasculature.  相似文献
