Experimental cohesive energies were quite often used as bulk energies in semi-empirical or empirical modeling of transition metal and alloy properties (e.g. in TB-SMA, the EAM, the bond order simulation (BOS) model, etc.), thus ignoring the free-atom spin-polarizations and promotion energy contributions. The importance of employing corrected cohesion energy in avoiding erroneous results is emphasized, especially for alloys with similar cohesion energies of the constituents. We demonstrate the effect of the correction in the prediction of equilibrium compositional structures and thermodynamical properties of cuboctahedral 55 atom Pt–Rh nanoclusters.  相似文献

During vocal fold vibration, there may be a mucosal wave in the superior-inferior (vertical) direction, resulting in a convergent shape during opening and a divergent shape during closing. Most of our understanding of the converging/diverging shape of the glottis has come from studies in a hemilarynx model. Previous work has shown that vibratory patterns in the full excised larynx are different than the hemilarynx. This study characterized the dynamics of the medial glottal wall geometry during vibrations in the full excised canine larynx model. Using particle image velocimetry, the intraglottal geometry was measured at the midmembranous coronal plane in an excised canine larynx model. Measurements of the glottal area were taken simultaneously using high-speed imaging. The results show that skewing of the glottal area waveform occurs without the presence of a vocal tract and that the phase-lag of the superior edge relative to the inferior edge is smaller than reported and depends on the subglottal pressure. In addition, it shows that the glottal divergence angle during closing is proportional to the magnitude of the acoustic intensity and the intraglottal negative pressure. This preliminary data suggests that more studies are needed to determine the important mechanisms determining the relationship between intraglottal flow, intraglottal geometry, and acoustics.  相似文献

A polymer therapeutic designed for combination anticancer and antiangiogenic therapy inhibited the proliferation of prostate carcinoma cells and the proliferation, migration, and tube‐formation of endothelial cells. The nanoconjugate was formed from an N‐(2‐hydroxypropyl)methacrylamide (HPMA) copolymer, the bisphosphonate alendronate (for bone targeting), and the chemotherapy agent paclitaxel (PTX), which is cleaved by cathepsin B (see scheme).

Droplet microfluidics offers significant advantages for performing high-throughput screens and sensitive assays. Droplets allow sample volumes to be significantly reduced, leading to concomitant reductions in cost. Manipulation and measurement at kilohertz speeds enable up to 10(8) samples to be screened in one day. Compartmentalization in droplets increases assay sensitivity by increasing the effective concentration of rare species and decreasing the time required to reach detection thresholds. Droplet microfluidics combines these powerful features to enable currently inaccessible high-throughput screening applications, including single-cell and single-molecule assays.  相似文献

The yogurt film confined within a nanogap was investigated by the thin film interferometry between two solid surfaces. Significantly thick film was found even under a low speed, which indicated yogurt might have excellent lubrication performance. The film thickness was much higher than the prediction of elastohydrodynamic lubrication theory. Experiment results indicated that the thick confined film of yogurt can be induced by the adhesion of organic material, which can sustain high pressure without being extruded under low speed. The effect of the rolling speed on film thickness shows that the lubrication under a high rolling speed tends to be the elastohydrodynamic lubrication. Purpose

To assess the feasibility and to optimize imaging parameters of diffusion kurtosis imaging (DKI) in human kidneys.

Methods

The kidneys of ten healthy volunteers were examined on a clinical 3 T MR scanner. For DKI, respiratory triggered EPI sequences were acquired in the coronal plane (3 b-values: 0, 300, 600 s/mm2, 30 diffusion directions). A goodness of fit analysis was performed and the influence of the signal-to-noise ratio (SNR) on the DKI results was evaluated. Region-of-interest (ROI) measurements were performed to determine apparent diffusion coefficient (ADC), fractional anisotropy (FA) and mean kurtosis (MK) of the cortex and the medulla of the kidneys. Intra-observer and inter-observer reproducibility using Bland-Altman plots as well as subjective image quality of DKI were examined and ADC, FA, and MK parameters were compared.

Results

The DKI model fitted better to the experimental data (r = 0.99) with p < 0.05 than the common mono-exponential ADC model (r = 0.96).Calculation of reliable kurtosis parameters in human kidneys requires a minimum SNR of 8.31 on b = 0 s/mm2 images.Corticomedullary differentiation was possible on FA and MK maps. ADC, FA and MK revealed significant differences in medulla (ADC = 2.82 × 10− 3 mm2/s ± 0.25, FA = 0.42 ± 0. 05, MK = 0.78 ± 0.07) and cortex (ADC = 3.60 × 10− 3 mm2/s ± 0.28, FA = 0.18 ± 0.04, MK = 0.94 ± 0.07) with p < 0.001.

Conclusion

Our initial results indicate the feasibility of DKI in the human kidney presuming an adequate SNR. Future studies in patients with kidney diseases are required to determine the value of DKI for functional kidney imaging.  相似文献

Rapid antimicrobial susceptibility tests (ASTs) are essential tool for proper treatment of patients infected by Yersinia pestis (Y. pestis), the causative agent of plague, or for post-exposure prophylaxis of a population exposed to a naturally acquired or deliberately prepared resistant variant. The standard AST of Y. pestis is based on bacterial growth and requires 24–48 h of incubation in addition to the time required for prior isolation of a bacterial culture from the clinical or environmental sample, which may take an additional 24–48 h. In this study, we present a new and rapid AST method based on a fluorescence determination of the minimum inhibitory concentration (MIC). Our method includes the incubation of bacteria with an antibiotic, followed by staining of the bacteria with oxonol dye (SynaptoGreen C4/FM1–43), which enables the rapid detection of an antibiotic’s effect on bacterial viability. We show that stained, non-viable bacteria exhibit a spectral redshift and an increase in fluorescence intensity compared to intact control bacteria. Based on these criteria, we developed a rapid flow cytometer measurement procedure and a unique spectral intensity ratio (SIR) analysis that enables determination of antibiotic susceptibility for Y. pestis within 6 h instead of the 24 to 48 h required for the standard AST. This new rapid determination of antibiotic susceptibility could be crucial for reducing mortality and preventing the spread of disease.  相似文献

We report the first successful optical pumping of cesium atoms to a state of high spin polarization with a CW dye laser tuned to the D1 second resonance line (4593 Å) of the cesium atom. Results of spin polarization versus laser power measurements for cells containing high-density cesium vapors (1013–1015 cm-3) plus mixtures of nitrogen (10–200 torr) and helium (≈1 atmosphere) gases are presented. These experiments have revealed unexpected limits to the spin polarization of dense alkali vapors.  相似文献

We present an integrated approach for highly sensitive identification and validation of substrate-specific kinases as cancer biomarkers. Our approach combines phosphoproteomics for high throughput cancer-related biomarker discovery from patient tissues and an impedimetric kinase activity biosensor for sensitive validation. Using non-small-cell lung cancer (NSCLC) as a proof-of-concept study, label-free quantitative phosphoproteomic analysis of a pair of cancerous and its adjacent normal tissues revealed 198 phosphoproteins that are over-phosphorylated in NSCLC. Among the differentially regulated phosphorylation sites, the most significant alteration was in residue S165 in the Hepatoma Derived Growth Factor (HDGF) protein. Hence, HDGF was selected as a model system for the electrochemical studies. Further motif-based analysis of this altered phosphorylation site revealed that extracellular-signal-regulated kinase 1/2 (ERK1/2) are most likely to be the corresponding kinases. For validation of the kinase–substrate pair, densely packed peptide monolayers corresponding to the HDGF phosphorylation site were coupled to a gold electrode. Phosphorylation of the monolayer by ERK2 and dephosphorylation by alkaline phosphatase (AP) were detected by electrochemical impedance spectroscopy (EIS) and surface roughness analysis. Compared to other methods for quantification of kinase concentration, this label-free electrochemical assay offers the advantages of ultra-sensitivity as well as higher specificity for the detection of cancer-related kinase–substrate pair. With implementation of multiple kinase–substrate biomarker pairs, we expect this integrated approach to become a high throughput platform for discovery and validation of phosphorylation-mediated biomarkers.  相似文献
