We have developed an on-line strong cation exchange (SCX)-ESI-MS/MS platform for the rapid identification of proteins contained in mixtures. This platform consists of a SCX precolumn followed by a nanoflow SCX column on-line with an electrospray ion trap mass spectrometer. We also used this platform to study the dynamics of peptide separation/extraction by SCX, in particular to understand the parameters affecting the performance of SCX in multidimensional chromatography. For example, we have demonstrated that the buffer typically used for tryptic digestion of protein mixtures can have a detrimental effect on the chromatographic behaviour of peptides during SCX separations, thereby affecting certain peptide quantitation approaches that rely on reproducible peptide fractionation. We have also demonstrated that band broadening results when a step (discontinuous) gradient approach is used to displace peptides from the SCX precolumn, reducing the separation power of SCX in multidimensional chromatography. In contrast, excellent chromatographic peak shapes are observed when a defined (continuous) gradient is used. Finally, using a tryptic digest of a protein extract derived from human K562 cells, we observed that larger molecular weight peptides are identified using this on-line SCX approach compared to the more conventional reverse phase (RP) LC/MS approach. Both methods used in tandem complement each other and can lead to a greater number of peptide identifications from a given sample.  相似文献

Ginseng, the underground parts of plants of Panax species, has been used in oriental traditional medicine for centuries. Unfortunately, because of extensive exploitation over thousands of years, the natural source of these species has been almost exhausted. Recently, we have found a wild ginseng growing in Myanmar. Here, by a combination of chemical composition study and gene sequence analysis, we unambiguously demonstrate that the wild ginseng is actually P. zingiberensis, commonly known as ginger ginseng. This ginseng was an indigenous to the southwestern China. However, now it is seriously threatened to brink of extinction and is put on the highest level of protection in China. Therefore, an appropriate protection measure is highly recommended to preserve this valuable resource, since this Myanmar ginseng might turn out to be the last P. zingiberensis, which could ever be seen in the planet.  相似文献

To realize the effective conversion of renewable energy through water decomposition, efficient electrocatalysts for the oxygen evolution reaction (OER) are essential. In this article, PBA@POM was successfully prepared with a Prussian blue analogue (PBA) as the initial structure. A facile hydrothermal process is reported for obtaining PBA@POM by etching the cubic PBA with a strong Brønsted acid, H3PMo12O40 (HPMo). The hollow cube structure not only exposes more active sites but also promotes electron transport, which results in excellent electrocatalytic activity for the OER. Compared with the PBA, which initially simply adhered to POM, the optimum PBA@POM hybrids display remarkably enhanced OER catalytic activity, with an almost constant overpotential of 440 mV at a current density of 10 mA cm?2 and a small Tafel slope (23.45 mV dec?1). The facilely prepared PBA@POM with good electrochemical activity and stability promises great potential for the OER.  相似文献

The sphene-type solid electrolyte with high ionic conductivity has been designed for solid-state lithium metal battery. However, the practical applications of solid electrolytes are still suffered by the low relative density and long sintering time of tens of hours with large energy consumption. Here, we introduced the spark plasma sintering technology for fabricating the sphene-type Li1.125Ta0.875Zr0.125SiO5 solid electrolyte. The dense electrolyte pellet with high relative density of ca. 97.4% and ionic conductivity of ca. 1.44×10-5 S/cm at 30℃ can be obtained by spark plasma sintering process within the extremely short time of only ca. 0.1 h. Also the solid electrolyte provides stable electrochemical window of ca. 6.0 V(vs. Li+/Li) and high electrochemical interface stability toward Li metal anode. With the enhanced interfacial contacts between electrodes and electrolyte pellet by the in-situ formed polymer electrolyte, the solid-state lithium metal battery with LiFePO4 cathode can deliver the initial discharge capacity of ca. 154 mA·h/g at 0.1 C and the reversible capacity of ca. 132 mA·h/g after 70 cycles with high Coulombic efficiency of 99.5% at 55℃. Therefore, this study demonstrates a rapid and energy efficient sintering strategy for fabricating the solid electrolyte with dense structure and high ionic conductivity that can be practically applied in solid-state lithium metal batteries with high energy densities and safeties.  相似文献

Thermal oxidation of sulfur vulcanized polyisoprene samples was studied by gravimetry and IR mapping of carbonyl groups (to determine the oxidized layer thickness (TOL)) at temperatures ranging from 60 to 150 °C in air. Oxidation appears noticeably lower than that for the starting non-vulcanized polyisoprene, revealing a stabilizing effect of sulfur-containing species. After a short period where mass loss presumably due to water evaporation predominates, the sample mass increases until a plateau corresponding to 6.3% (at 60 °C) to 0.5% (at 140 °C) mass gain. Practically no weight gain (∼0.1%) was observed at 150 °C. The mass uptake is due to oxygen grafting to the chains. TOL varies from about 4.6 mm (70 °C) to about 1 mm (150 °C).A kinetic model, derived from a mechanistic scheme of radical chain oxidation including stabilizing events due to hydroperoxide reduction by sulfur-containing groups and taking into account the diffusion-reaction coupling, was established and numerically resolved. The model predictions for mass changes and TOL values are in good agreement with experimental data.  相似文献

Management of domestic wastes often relies on incineration, a process that eliminates large amount of wastes but also produces toxic residues that concentrate heavy metals. Those hazardous secondary wastes require specific treatment. Vitrification is seen as a powerful way to stabilise them. However, concern exists about the long term behaviour of these glass wastes and the potential release of toxic species into the environment. The answers will come with further investigation into the physico-chemical evolution of the vitrified wastes and the mobility of hazardous elements within the matrix with appropriate analytical methods. Laser ablation coupled with inductively coupled mass spectrometry (LA-ICP-MS) is a challenging technique for the chemical analysis of trace elements in solid materials. This paper presents an evaluation of the potential of LA- ICP-MS for macro and microanalysis of trace metals in domestic vitrified wastes with regards to other physical analytical techniques of solids such as scanning electronprobe X-ray energy dispersive spectroscopy (SEM-EDXS). Two typical samples, vitreous and crystallised, are used to compare the analytical performances of the two techniques. SEM-EDXS was used for mineralogical characterisation and chemical analysis of the mineralogical phases. Relative micro-analysis and bulk quantitative analysis of 30 major, minor and trace elements was performed by LA-ICP-MS: precision was between 10 and 20% for most elements and quantitative analysis proved possible with an accuracy of 20% and relative detection limits of 0.1 mg kg(-1).  相似文献

Thermally stable dipolar and octupolar (D2d, D3) NLO-phores are readily accessible by combining one, two, or three 4,4'-bis(dialkylaminostyryl)-[2,2']-bipyridyl ligands with zinc(II) salts. The off-resonant beta0 values point out the superiority of octupoles versus dipoles in terms of nonlinearity/transparency tradeoff. The octahedral tris(bipyridyl)zinc(II) complex exhibits a very large beta0 value (241 x 10-30 esu), which is the largest ever reported for octupolar molecules.  相似文献
