We report on a stabilizer of the interaction between 14-3-3ζ and the Estrogen Receptor alpha (ERα). ERα is a driver in the majority of breast cancers and 14-3-3 proteins are negative regulators of this nuclear receptor, making the stabilization of this protein-protein interaction (PPI) an interesting strategy. The stabilizer ( 1 ) consists of three symmetric peptidic arms containing an arginine mimetic, previously described as the GCP motif. 1 stabilizes the 14-3-3ζ/ERα interaction synergistically with the natural product Fusicoccin-A and was thus hypothesized to bind to a different site. This is supported by computational analysis of 1 binding to the binary complex of 14-3-3 and an ERα-derived phosphopeptide. Furthermore, 1 shows selectivity towards 14-3-3ζ/ERα interaction over other 14-3-3 client-derived phosphomotifs. These data provide a solid support of a new binding mode for a supramolecular 14-3-3ζ/ERα PPI stabilizer.  相似文献

Diesel soot (Euro IV and Euro VI) was investigated with spectroscopic methods such as near-edge X-ray absorption fine structure (NEXAFS) spectroscopy and X-ray photoemission spectroscopy (XPS). C and O K-edge NEXAFS show that structural disorder on the surface is accompanied by a higher amount of oxygen functional groups. O K-edge NEXAFS and O1s XPS results are discussed with the aim to elucidate the nature of the oxygen surface species. The analysis of the data presented here allows the postulation of a hypothetical structure for soot samples emitted by diesel engines.  相似文献

Variation of the cultivation conditions for Aspergillus glaucus led to the discovery of two novel spirocyclic aromatic polyketides, aspergiolides C (3) and D (4). Their constitutions were elucidated by a combination of spectroscopic methods and isotope-labeling experiments. Aspergiolides C (3) and D (4) occur as racemic mixtures, the resolution of which was succeeded by HPLC on a chiral phase. The absolute configurations of their enantiomers were assigned online, from the peaks in the chromatogram, by a combination of HPLC-CD and quantum chemical CD calculations. Both compounds were found to inhibit the kinase activities of the receptor tyrosine kinases (RTKs) c-Met, Ron, and c-Src with low-micromolar IC(50)s. The enantiomers of 3 were resolved by HPLC on a chiral phase. Both enantiomers showed a comparable inhibition of the HGF-induced autophosphorylation of c-Met and of subsequent cell migration.  相似文献

We report on reversible changes of the work function (WF) values of indium-tin-oxide (ITO) under prolonged ultraviolet (UV) and air exposure. The WF of ITO is reduced from 4.7 eV to 4.2 eV by photon absorption in ITO under UV illumination or an air mass 1.5 solar simulator (100 mW cm(-2)). Air or oxygen exposure is found to increase the WF of ITO (UV-exposed) to a value of 4.6 eV. These changes of ITO's WF lead to reversible variations of the performance of organic photovoltaic devices where ITO acts primarily as the electron collecting or hole collecting electrode. These variations can be reflected in the disappearance (or appearance) of an S-shaped kink in the J-V characteristics upon continuous UV or solar simulator illumination (or air exposure). This reversible phenomenon is ascribed to the adsorption and desorption of oxygen on the surface and grain boundaries of ITO. The use of surface modifiers to either decrease or increase the WF of ITO in organic photovoltaic devices with inverted and conventional geometries is also shown to be an effective route to stabilize the device performance under UV illumination.  相似文献

We present an immunoassay microarray flow-through system for the surface-enhanced Raman scattering (SERS) analysis of bacteria. The system has been constructed to support and automatize the nondestructive in situ analysis of different microorganisms in aqueous environment. After the immobilization of the desired antibodies to an activated PEG-coated surface, the chip is placed into the flow cell which is then flushed with the contaminated sample. Finally, colloidal metal nanoparticles are added and the cells are detected label-free by SERS. Here, we introduce the successful imaging of single microorganisms in the flow cell as well as the quantification of microorganisms in water by SERS mapping with a linear range between 4.3 × 103 to 4.3 × 105 cells/mL. The method has potential for routine application, e.g. for drinking water control.  相似文献

Products of sorption of uranyl ions on HDTMA-red clay in the presence of phosphates were characterized by thermal analysis. It was established on the basis of DTG curves of the sorption products and FTIR spectra of the gaseous phase of sorption products decomposition that the thermal stability of the mineral increased when P(V) ions were sorbed along with U(VI) ions, i.e., the temperature of defragmentation/oxidation of surfactant increased when going from U(VI)–HDTMA-clay to U(VI)–P(V)–HDTMA-clay to P(V)–HDTMA-clay. The DSC curves of the sorption products showed that defragmentation/oxidation was an exothermic process and dehydration and dehydroxylation had an endothermic character. The investigated sorption system has practical importance, since an evident increase in U(VI) sorption over the entire pH range is observed when going from U(VI)–HDTMA-clay to U(VI)–P(V)–HDTMA-clay.

We introduce "microdeflectometry," a novel technique for measuring the microtopography of specular surfaces. The primary data are the local slope of the surface under test. Measuring the slope instead of the height implies high information efficiency and extreme sensitivity to local shape irregularities. The lateral resolution can be better than 1 microm, whereas the resulting height resolution is in the range of 1nm. Microdeflectometry can be supplemented by methods to expand the depth of field, with the potential to provide quantitative 3D imaging with scanning-electron-microscope-like features.  相似文献
