A thermal analyzer consisting of a pair of glass thermistors was coupled with a flowthrough immobilized enzyme reactor for the determination of ATP concentration. Hexokinase was covalently bound to poly(chloromethyl-styrene-divinyl benzene) beads and packed in the reactor. The thermal analyzer was designed so as to respond to heat generation in the reactor. A maximum temperature change was exhibited within 1.5 min after the injection of a sample solution containing ATP together with glucose. Thermal decay occurred within 1 min. The thermal analyzer responded reproducibly to ATP at a constant concentration. Assays could be repeated at 3-min intervals. The effects of sample volume and flow rate on the response were examined with regard to an empirical equation. Concentration of ATP was determined with the thermal analyzer in the range of 0-10 mM. The thermal analyzer may be a simple device for the rapid determination of ATP concentration.  相似文献

A non-destructive photon activation procedure with 30-MeV bremsstrahlung followed by high-resolution γ-spectrometry is proposed for multielement analysis of biological materials. The materials tested were the NBS SRM Orchard Leaves and Bovine Liver, Bowen′s kale and Kentucky 1R1 tobacco standards. Simultaneous irradiation of the sample with synthetic multielement standards containing 25 elements showed that up to 12 elements can be determined in a single sample. The method is quite simple and gives good reproducible results. Agreement of the results with published data is excellent.  相似文献

Infrared and visible absorption spectra of the low pressure heated products of S2O6F2 trapped in inert gas matrices at 14 K are presented. The following have been identified as major infrared-absorbing species: SO3F, SO3, HSO3F, and S2O6F2, with matrix spectra of the latter three unequivocally confirming their presence. Reasonable reactions accounting for the production of these species are proposed, and a reassignment of the fundamental vibrations of monomeric HSO3F is offered. The infrared absorptions of the SO8F radical have been observed for the first time, and the six fundamental vibrations determined. These frequencies are in good accord with earlier values deduced indirectly by King and Warren from hot band transitions in the visible absorption spectrum of the vapor. This same visible system (2E-X?2A2) has also been observed here for the matrix isolated species.  相似文献
