We describe here an optimization study of the sample preparation conditions for sensitive detection of peptides by matrix‐assisted laser desorption/ionization mass spectrometry (MALDI‐MS). Among many factors in the conditions, we varied the percent acetonitrile in the peptide solution, the percent acetonitrile in the matrix solution and the α‐cyano‐4‐hydroxycinnamic acid (CHCA) concentration in the matrix solution. CHCA was chosen because it is the most frequently used matrix for analyzing peptides. The well‐established dried‐droplet method was employed for sample deposition. The examined range of the concentration of CHCA was from 0.01 to 10 mg/ml, and the MeCN content of the solvent for matrix/analyte was 10% to 50%. The indicator for the detection sensitivity was the S/N ratio of the peaks of peptides used. Highly increased sensitivity (100‐ to 1000‐fold) was observed for the optimal CHCA concentration of 0.1 mg/ml in 20% MeCN/0.1% aq. trifluoroacetic acid (TFA), as compared with the conventional concentration (10 mg/ml) in 50% MeCN/0.1% aq. TFA. For example, the limit of detection of human ACTH 18–39 was 10 amol/well for the optimal condition but 10 fmol/well for the conventional condition. The optimal condition (0.1 mg/ml CHCA in 20% MeCN/0.1% aq. TFA) was verified with five model peptides and provided significant improvement in sensitivity (by two to three orders of magnitude) compared with the conventional conditions. The influence of polarity and rise time of the pulse voltage on the removal of particulate matter (PM) emitted from a diesel engine was investigated using a dielectric barrier discharge reactor. Four kinds of pulse voltage waveforms (positive, negative, positive–negative and negative–positive) were used. It was found that the energy efficiency for PM removal is just a function of energy injection and that there are no obvious influences on PM removal and energy efficiency within the voltage waveforms except the negative pulse voltage of a peak voltage below 8 kV. A comparison of PM removals using various kinds of pulse voltage waveforms and different types of plasma reactors is given.  相似文献

Fifty-one sections of infrared diode-laser spectra of acetonitrile have been measured in the region from 2283.5 to 2235.7 cm?1. About 450 transitions belonging to the ν2 band have been assigned for K ≦ 7 and J ≦ 44. Anomalies found in the rotational structure have been proven to be due to five local resonances. Observed transition frequencies have been fitted by a least-squares method to a model which includes Fermi-type resonances (Δk = 0, Δ? = ± 3n) with ν6±1 + 2ν8±2 and ν4 + ν7±1 + ν8±1 states, x, y-type Coriolis resonances (Δk = ±1, Δ? = ?3n ± 1) with ν4 + ν6±1 and ν4 + ν7±1 + ν8±1 states, and a centrifugal-distortion-type resonance (Δk = ±2, Δ? = ?3n ± 2) with a 2ν70 + ν8±1 state. The 11 × 11 dimensional energy matrix has been diagonalized in order to obtain the perturbed energy levels. The standard deviation for the fit is 1.075 × 10?3 cm?1. Polymerization of benzyl vinyl ether was carried out by BF3·OEt2, and the effects of polymerization conditions on the stereoregularity of the polymer were studied by NMR analysis. The polymerization at ?78°C in toluene gave a highly isotactic polymer. The isotacticity of the polymer was independent of the catalyst concentration but increased with a decrease in the initial monomer concentration and decreased slightly on raising the reaction temperature. When the polymerizations were carried out in toluene—nitroethane mixtures, a gradual decrease in the isotacticity and a rapid decrease in the molecular weight of the polymer were observed with increasing nitroethane in the solvent. The molecular weight of the polymer was almost constant, regardless of the catalyst concentration, and increased with increasing initial monomer concentration and decreasing polymerization temperature. When the polymerization was performed in toluene at ?78°C with a small amount of water or benzyl alcohol, a linear relationship was found between the reciprocal DP of the polymer and water or benzylalcohol concentration. The mechanisms of the initiation reaction and the stereoregulation in the polymerization were also discussed.  相似文献

Polymerization conditions of di-t-butyl fumarate and di-trimethylsilyl fumarate were studied in detail. They cannot be polymerized by either anionic or coordination initiators, but radical and radiation polymerizations are successful. Characterization of poly(di-t-butyl fumarate), obtained thereby, with 1H-NMR spectrum suggests that the backbone of the chain is stiff. From analysis of thermal properties of poly(di-t-butyl fumarate), it is found to be completely converted to poly(fumaric acid) by pyrolysis around 200°C. Poly(di-trimethylsilyl fumarate), on the other hand, can be quantitatively hydrolyzed with acid to the same polyacid, too. The preliminary measurement of the dissociation behaviors of poly(fumaric acid) was done by potentiometric titration, which shows that the titration curves of poly(fumaric acid) are different from those of poly(acrylic acid) and poly(maleic acid).  相似文献

A rapid method for the determination of Pu isotopes in a soil sample within 60 min of starting sample pretreatment was developed. The large reduction in the analysis time was attained by the rapid and perfect digestion of the sample using an alkaline fusion method with an induction heating machine. Pu concentrations were then determined by flow injection/ICP-MS using a solid extraction resin after preconcentration by batch extraction with a chelate resin. The limits of detection for 239Pu and 240Pu were 9.2 fg and 4.3 fg, corresponding to 0.03 and 0.05 Bq kg(-1), respectively, under our analytical conditions, which satisfy the lower detection limits (0.5 Bq kg(-1) of 239Pu, and 2 Bq kg(-1) of 240Pu) required for rapid analysis techniques by the Ministry of Education, Culture, Sports, Science and Technology, Japan. This method provides a powerful and practical technique for emergency monitoring in and around nuclear facilities that handle large amounts of plutonium.  相似文献

We fabricated the ordered Sr2FeMoO6 (SFMO) thin film with a double perovskite structure using the chemical solution deposition (CSD) method. The highly c-axis oriented SFMO thin film with a high degree of Fe/Mo ordering was successfully synthesized on an MgO (001) substrate by optimizing processing conditions. The precise preparation process control of the SFMO precursor solution leads to a typical magnetoresistance effect in a low magnetic field at room temperature.  相似文献

The photoconductivity of bulk glasses of the TeSeSb system is measured as a function of light intensity and photon energy. The relative sensitivity (ΔI/Id) has linear and square-root dependences on light intensity in low and high illumination intensities, respectively, and is nearly proportional to the square-root of the resistivity at room temperature. The spectral response of photoconductivity, which is calculated by taking into account the effect of surface recombination of carriers, agrees qualitatively with the experimental results. The experimentally determined broad spectral response suggests the presence of band tails below the conduction band and above the valence band. The large residual dark conductivity in the decay response is associated with the presence of many deep trapping centers.  相似文献

Fluorescence- and biotin-labeled lipid A analogues were synthesized for the investigation of bacterial lipopolysaccharide (LPS)/lipid A recognition in the innate immune system. For the introduction of the labeling moiety, a hydrophilic glutaryl-glucose linker was used for maintaining the bioactivity and also for preventing self-aggregation, which causes quenching of the fluorescence. The synthesis of immunoregulatory glycoconjugates, namely the active entity of lipopolysaccharide (LPS) from Helicobacter pylori was achieved.
