Expression of Growth Hormone-Releasing Hormone and Its Receptor Splice Variants in Primary Human Endometrial Carcinomas: Novel Therapeutic Approaches

Antagonists of growth hormone-releasing hormone (GHRH) inhibit the growth of various tumors, including endometrial carcinomas (EC). However, tumoral receptors that mediate the antiproliferative effects of GHRH antagonists in human ECs have not been fully characterized. In this study, we investigated the expression of mRNA for GHRH and splice variants (SVs) of GHRH receptors (GHRH-R) in 39 human ECs and in 7 normal endometrial tissue samples using RT-PCR. Primers designed for the PCR amplification of mRNA for the full length GHRH-R and SVs were utilized. The PCR products were sequenced, and their specificity was confirmed. Nine ECs cancers (23%) expressed mRNA for SV1, three (7.7%) showed SV2 and eight (20.5%) revealed mRNA for SV4. The presence of SVs for GHRH-Rs could not be detected in any of the normal endometrial tissue specimens. The presence of specific, high affinity GHRH-Rs was also demonstrated in EC specimens using radioligand binding studies. Twenty-four of the investigated thirty-nine tumor samples (61.5%) and three of the seven corresponding normal endometrial tissues (42.9%) expressed mRNA for GHRH ligand. Our findings suggest the possible existence of an autocrine loop in EC based on GHRH and its tumoral SV receptors. The antiproliferative effects of GHRH antagonists on EC are likely to be exerted in part by the local SVs and GHRH system.

Application of Metathesis Protocols to the Stereocontrolled Synthesis of some Functionalized β-Amino Esters and Azaheterocycles

In this account our aim was to give an insight into the application of metathesis protocols (ROM, RCM, RCEYM, CM, RRM) for the synthesis of various azaheterocyclic frameworks. Due to the high biological potential and importance in peptide chemistry and drug design of β-amino acids our intention is to give a highlight on the synthetic procedures and transformation of these class of compounds with the above-mentioned metathesis strategies with emphasis on selectivity, stereocontrol, substrate-directing effect or functional group tolerance.     

Investigation of the regeneration of the passive surface oxide layer of TiAl6V4 alloy printed by WAAM technology using different electrochemical test methods

The kinetics of oxide layer formation on surface of Ti6Al4V alloy samples is a very important property especially if their application as medical implants is planned. Damaged protective surface layer usually heals in ambient condition however; during the self-healing process toxic species can get into the surrounding living tissue. In our experiment the kinetics of the healing process proceeding at 3D printed alloy surface has been studied using electrochemical methods, among them scanning electrochemical microscopy. More than 40 min. time period was found long enough for total healing.     

Studies of Phenol Electrooxidation Performed on Platinum Electrode in Dimethyl Sulphoxide Medium. Determination of Unreacted Phenol by the Effect of 4-Vinylbenzenesulphonate on the Electrooxidation Process

The electrooxidation of phenol was studied in the presence of sodium 4-vinylbenzenesulfonate in various quantities. When the sulphonate was dissolved in equal or higher quantities than phenol the peak currents increased gradually and suppression of electropolymerization was observed. When the sulphonate quantity was smaller than that of phenol brown films fouled the electrode. The observed phenomenon was utilized in an analytical procedure namely determination of unreacted phenol in electrolysis solution with linear dynamic range between 0 and 50 mM and 2.96 mM detection limit. The shape of curve became saturated-like above 50 mM.     

DD‐type Domain Decomposition Method for Frictional Contact Problem

The bilateral or unilateral contact problem with Coulomb friction between two elastic bodies is considered [1]. An algorithm is introduced to solve the resulting finite element system by a non‐overlapping domain decomposition method. The global problem is transformed to a smaller problem on the contact surface. The solution is obtained by using a successive approximation method, in each step of this algorithm we solve two intermediate problems the first with prescribed tangential pressure and the second with prescribed normal pressure.
