The global dispersion of hemoglobin variants through population migration has precipitated a need for their identification. A particularly effective mass spectrometry (MS)-based procedure involves analysis of the intact globin chains in diluted blood to detect the variant through mass anomalies, followed by location of the variant amino acid residue by direct analysis of the enzymatically digested globins. Here we demonstrate the use of ion mobility separation in combination with this MS procedure to reduce mass spectral complexity. In one example, the doubly charged tryptic peptide from a low abundance variant (4%) occurred at the same m/z value as a singly and a doubly charged interfering ion. In another example, the singly charged tryptic peptide from an alpha-chain variant (26%) occurred at the same m/z value as a doubly charged interfering ion. Ion mobility was used to separate the variant ions from the interfering ions, thus allowing the variant peptides to be observed and sequenced by tandem mass spectrometry.  相似文献

meso-Aryl substituted rubyrin ([26]hexaphyrin(1.1.0.1.1.0)) 2 and a series of rubyrin-type large expanded porphyrins were obtained from a facile one-pot oxidative coupling reaction of meso-pentafluorophenyl substituted tripyrrane 1. The structures of two of the resulting products were determined by single-crystal X-ray diffraction analysis. Whereas [52]dodecaphyrin(1.1.0.1.1.0.1.1.0.1.1.0) 4 takes a symmetric helical conformation, the larger species, [62]pentadecaphyrin(1.1.0.1.1.0.1.1.0.1.1.0.1.1.0) 5, adopts a nonsymmetric distorted conformation in the solid state that contains an intramolecular helical structure. The ability of rubyrin 2 to act as an anion receptor in its diprotonated form (2(.)2H(+)) was demonstrated in methanolic solutions. Oxidation of 2 with MnO(2) gave [24]rubyrin 6, a species that displays antiaromatic characteristics. [26]Rubyrin 2 and [24]rubyrin 6 both underwent metallation when reacted with Zn(OAc)(2) to give the corresponding bis-zinc(II) complexes 7 and 8 quantitatively without engendering a change in the oxidation state of the ligands. As a result, complexes 7 and 8 exhibit aromatic and antiaromatic character, respectively. NICS calculation on these compounds also supported aromaticity of 2 and 7, and antiaromaticity of 6 and 8.  相似文献

The C4'-oxidized abasic site (C4-AP) is produced in DNA as a result of oxidative stress. A recent report suggests that this lesion forms interstrand cross-links. Using duplexes in which C4-AP is produced from a synthetic precursor, we show that the lesion produces interstrand cross-links in which both strands are in tact and cross-links in which the C4-AP containing strand is cleaved. The yields of these products are dependent upon the surrounding nucleotide sequence. When C4-AP is opposed by dA, cross-link formation occurs exclusively with an adjacent dA on the 5'-side. Moreover, formation of the lower molecular weight cross-link is promoted by an opposing adenine. When the opposing dA is replaced by dT, the activity of the adenine can be rescued by adding the free base. This is a rare example in which DNA promotes its own modification, an observation that is all the more important because of the biological significance of the product produced.  相似文献

The synthesis, photophysical, and anion-binding properties of a series of di-, tri-, and tetrapodal anion-binding hosts based on aminopyridinium units with pyrenyl reporter groups are described. The ditopic mesitylene-derived calix[4]arene-based host 4 binds strongly to dicarboxylates, particularly malonate, in a 2:1 anion:host ratio but is essentially nonemissive in the presence of all anions except chloride because of intramolecular quenching by the pyridinium units. Addition of chloride results in a conformational change, giving an initial increase in emission assigned to intramolecular excimer formation. Further chloride addition also results in an increase in the intensity of the pyrenyl monomer emission as chloride binding reduces the acceptor ability of the pyridinium groups. This behavior is not exhibited by control compounds 5 and 6, which lack the ditopic geometry and calixarene spacer unit; however, tripodal 6 forms 1:2 anion:host complexes with a range of anions.  相似文献

A 12-mer peptide, identified through phage display biopanning, has been used for the first time to induce the rapid formation of ferroelectric (tetragonal) nanocrystalline BaTiO3 at room temperature from an aqueous salt precursor solution at near neutral pH. BaTiO3 is widely used in capacitors, thermistors, displays, and sensors owing to its attractive dielectric, ferroelectric, pyroelectric, optical, and electrochemical properties. Two 12-mer peptides (BT1 and BT2) were selected from a phage-displayed peptide library via binding to tetragonal BaTiO3 powder. While these peptides possessed various types of amino acids, 8 of the 12 amino acids were common to both peptides. Each of these peptides induced the formation of faceted nanoparticles (50-100 nm diameter) from an aqueous precursor solution. X-ray diffraction and selected area electron diffraction patterns obtained from these faceted nanoparticles were consistent with the BaTiO3 compound. Rietveld analyses of the X-ray diffraction patterns yielded good fits to tetragonal crystal structures, with the BaTiO3 formed in the presence of the BT2 peptide exhibiting the most tetragonal character. A coating of the latter BaTiO3 nanoparticles exhibited polarization hysteresis (a well-known characteristic of ferroelectric materials) at room temperature and a relative permittivity of 2200. Such rapid, peptide-induced precipitation at room temperature provides new opportunities for direct BaTiO3 formation on low-melting or reactive materials (e.g., plastics, cloths, bio-organics) and the low temperature integration of BaTiO3 into electronic devices (e.g., on silicon or flexible polymer substrates).  相似文献

Nitric oxide (NO) is endogenously generated by nitric oxide synthase (NOS) enzymes and is involved in a surprisingly wide range of biological functions. As efforts are made to elucidate the regulatory mechanisms of NOS expression and function, there is increasing interest in following NOS activity directly by monitoring NO production. Additionally, spatial and temporal measurements of NO are important for understanding its function and metabolism. In this work, developments in technology enabling NO detection in biological systems are reviewed. Measuring NO at single cell levels is important as NOS is heterogeneously distributed; however, such measurements are difficult as physiological NO levels are in the low nanomolar to low micromolar range. Here, three categories of analytical techniques enabling NO detection at single cell levels are highlighted: fluorescence microscopy, capillary electrophoresis with laser induced fluorescence detection, and electrochemistry. For each, the basic principles, performance, applications, figures of merits and limitations are presented in terms of single cell NO detection.  相似文献

We have demonstrated sensitive label-free virus detection using the opto-fluidic ring resonator (OFRR) sensor. The OFRR is a novel sensing platform that integrates the microfluidics and photonic sensing technology with a low detection limit and small volume. In our experiment, filamentous bacteriophage M13 was used as a safe model system. Virus samples were flowed through the OFRR whose surface was coated with M13-specific antibodies. We studied the sensor performance by monitoring in real-time the virus and antibody interaction. It is shown that OFRR can detect M13 with high specificity and sensitivity. The detection limit is approximately 2.3 x 10(3) pfu mL(-1) and the detection dynamic range spanned seven orders of magnitude. Theoretical analysis was also carried out to confirm the experimental results. Our study will lead to development of novel OFRR-based, sensitive, rapid, and low-cost micro total analysis devices for virus detection.  相似文献

Recent advances in nanotechnology have produced a variety of nanoparticles ranging from semiconductor quantum dots (QDs), magnetic nanoparticles (MNPs), metallic nanoparticles, to polymeric nanoparticles. Their unique electronic, magnetic, and optical properties have enabled a broad spectrum of biomedical applications such as ultrasensitive detection, medical imaging, and specific therapeutics. MNPs made from iron oxide, in particular, have attracted extensive interest and have already been used in clinical studies owing to their capability of deep-tissue imaging, non-immunogenesis, and low toxicity. In this Research Highlight article, we attempt to highlight the recent breakthroughs in MNP synthesis based on a non-hydrolytic approach, nanoparticle (NP) surface engineering, their unique structural and magnetic properties, and current applications in ultrasensitive detection and imaging with a special focus on innovative bioassays. The reaction of oxygen with rhodium complexes containing N-heterocyclic carbenes was found to give dioxygen complexes with rare square planar geometries and unusually short O-O bond lengths. Analysis of the bonding in these complexes by Rh L-edge X-ray absorption spectroscopy (XAS), Raman spectroscopy, and DFT calculations provides evidence for a bonding model in which singlet oxygen is bound to a Rh(I) d8 metal complex, rather than the more common Rh(III) d6 peroxo species with octahedral geometry and O-O bond lengths in the 1.4-1.5 A range.  相似文献

A polymer monolith bearing weak cation-exchange functionality was prepared for the purpose of demonstrating pH-selective extraction and elution in in-line solid-phase extraction-capillary electrophoresis (SPE-CE) utilising a model set of cationic analytes, namely imidazole, lutidine and 3-phenylpropanamine. Optimization of the electrolyte conditions for efficient elution of the adsorbed analytes using a moving pH boundary required that the capillary and monolith be filled with 44 mM sodium acetate at high pH (pH 6) and a low pH electrolyte of 3 mM sodium acetate pH 3 was placed in the electrolyte vials. This combination allowed the adsorbed analytes to be simultaneously eluted and focused into narrow bands, with peak widths of the eluted analytes having a baseline width of 1.2 s immediately after the monolith. Using these optimum elution conditions, the versatility of the SPE-CE approach was demonstrated by removing unwanted adsorbed components after extraction with a wash at a different pH and also by selecting a pH at which only some of the model weak bases were ionised. The analytical performance of the approach was evaluated and the relative standard deviation for peak heights, peak area and migration times were in the ranges of 1.4-5.3, 1.2-3.3 and 0.4-1.2% respectively. Analytes exhibited linear calibrations with r(2) values ranging from 0.996 to 0.999 over two orders of magnitude. Analyte pre-concentration provided excellent sensitivity, and limits of detection for the analyte used in this study were in the range 8.0-30 ng ml(-1), which was an enhancement of 63 when compared to normal hydrodynamic injection occupying 1.3% of the capillary of these bases in water.  相似文献
